    Resveratrol (RSV) can protect against non-communicable diseases by improving cholesterol metabolism. However, it is unclear that effects of maternal RSV intake on health of adult offspring. In this study, we examined effects of maternal RSV intake during lactation on cholesterol metabolism in adult male rat offspring. Female Wistar rats were fed a control diet (CON) supplemented with or without RSV (20 mg/kg body weight/day) during their lactation period. Male offspring were weaned onto a standard diet and maintained on this diet for 36 weeks. As a result, plasma cholesterol level significantly decreased in RSV offspring compared to CON offspring. Furthermore, a decrease in hepatic 3-hydroxy-3-methylglutaryl-CoA reductase level and an increase in hepatic LDL-receptor level were observed in the RSV offspring. These results indicate that maternal RSV intake causes long-term decrease in plasma cholesterol level in the offspring through suppression of hepatic cholesterol biosynthesis and promotion of hepatic cholesterol uptake.

    Silver nanoparticles (AgNPs) were successfully synthesized from the reduction of Ag+ using AgNO3 solution as a precursor and Brassica rapa var. japonica leaf extract as a reducing and capping agent. This study was aimed at synthesis of AgNPs, exhibiting less toxicity with high antibacterial activity. The characterization of AgNPs was carried out using UV–Vis spectrometry, energy dispersive X-ray spectrometry, fourier transform infrared spectrometry, field emission scanning electron microscopy, X-ray diffraction, atomic absorption spectrometry, and transmission electron microscopy analyses. The analyses data revealed the successful synthesis of nano-crystalline Ag possessing more stability than commercial AgNPs. The cytotoxicity of Brassica AgNPs was compared with commercial AgNPs using in vitro PC12 cell model. Commercial AgNPs reduced cell viability to 23% (control 97%) and increased lactate dehydrogenase activity at a concentration of 3 ppm, whereas, Brassica AgNPs did not show any effects on both of the cytotoxicity parameters up to a concentration level of 10 ppm in PC12 cells. Moreover, Brassica AgNPs exhibited antibacterial activity in terms of zone of inhibition against E. coli (11.1 ± 0.5 mm) and Enterobacter sp. (15 ± 0.5 mm) which was higher than some previously reported green-synthesised AgNPs. Thus, this finding can be a matter of interest for the production and safe use of green-AgNPs in consumer products.

    Senescence-accelerated mice are known to display a variety of deficits and signs of accelerated aging, but the specific mechanisms involved in this process are still unclear. In this study, we examined the expression levels of antioxidant enzymes, transcription factors responsible for the regulation of expression of these enzymes, and mitochondrial proteins in the liver of SAMP10 and SAMR1 mice at 3 and 12 months of age using western blotting analysis. To investigate the amount of oxidative damage to DNA, levels of 8-OHdG were measured in the liver of these mice. At 3 months of age, the levels of catalase, Mn-SOD, GPx, UQCRC2 and COXIV were significantly upregulated in SAMP10 mice compared with that in SAMR1 mice. However, NDUFS3 levels were not significantly different at this young age. In contrast, the expression level of catalase was significantly lower, and the levels of phosphorylated FoxO-1a and UQCRC2 were significantly higher in SAMP10 mice compared to those in SAMR1 mice; however, at 12 months of age, there were no significant differences in Mn-SOD, GPx, total -FoxO-1a, COXIV, and NDUFS3 expression between the two groups of mice. The levels of 8-OHdG in the liver were markedly higher in 12-month-old SAMP10 mice than those in 3-month-old SAMP10 and SAMR1 mice. These results suggest that an increase in number of mitochondria or a collapse in the balance between the levels of complexes I and III results in an increase in the amount of ROS and induces the expression of antioxidant enzymes in the liver of SAMP10 mice at 3 months of age. Although young SAMP10 mice produce a large amount of ROS, they also produce suitable levels of antioxidant enzymes that decompose ROS; consequently accelerated aging does not occur in young SAMP10 mice. In addition to excessive ROS production which is an important cause of aging, the level of catalase was significantly lower in SAMP10 than that in SAMR1 mice. These results suggested that overexpression of ROS and a decrease in the levels of catalase resulted in the accelerated aging observed in older SAMP10 mice. Moreover, the level of phosphorylated FoxO-1a was increased in SAMP10 compared to that in SAMR1 mice though the total amount of FoxO-1a was not significantly different between the two groups in old age. These results suggest that some impairment in the regulation mechanism of FoxO-1a phosphorylation is responsible for abnormal catalase expression and that a significant decrease in the level of catalase with aging decisively affects the metabolic balance of ROS; thus, ROS that cannot be metabolized contributes to the accelerated aging of SAMP10 mice.

    Purpose of this study is to investigate mechanism/s of cyto-protection by selenium (Na2SeO3 Se4+) against cadmium (CdCl2 Cd2+)-induced cytotoxicity using PC12 cells. In addition, Se (5, 10, 20 and 40 μM) and Cd (2.5, 5 and 10 μM)-induced cytotoxicity is determined. Cytotoxicity assays and western blot analyses confirmed that Se (≥10 μM) promotes autophagic cell death via inhibition of mTOR activation and p62 accumulation due to increase of cellular oxidative stress. On the other hand, co-presence of non-toxic Se (5 μM) and toxic Cd (5 μM) showed to increase cell viability, glutathione and glutathione peroxidase 1 (GPx1) levels, and to decrease DNA fragmentation and lactate dehydrogenase (LDH) activity compared to Cd-treated (5 μM) cells alone. Furthermore, western blot analyses of cytochrome c and ERK1 indicated that Cd-induced apoptotic cell death in PC12 cells. However, the co-exposure of Se with Cd significantly decreases the release of cytochrome c into cytosol from mitochondria, and up-regulates ERK1 protein to inhibit Cd-induced apoptosis. In conclusion, Se (≥10 μM) possess cytotoxicity in PC12 cells however, co-presence of Se (5 μM) with Cd (5 μM) protects against Cd-induced apoptosis in PC12 cells due to inhibition of Cd-induced oxidative stress and subsequently suppression of mitochondrial apoptosis pathway.

    Arsenic is well known toxicant responsible for human diseases including cancers. On the other hand, selenium is an essential trace element with significant chemopreventive effects, anticancer potentials and antioxidant properties. Although previous studies have reported antagonism/synergism between arsenic and selenium in biological systems, the biomolecular mechanism/s is still inconclusive. Therefore, to elucidate the molecular phenomena in cellular level, we hypothesized that co-exposure of selenium with arsenic may have suppressive effects on arsenic-induced cytotoxicity. We found that selenium in co-exposure with arsenic increases cell viability, and suppresses oxidative stress induced by arsenic in PC12 cells. Consequently, DNA fragmentation due to arsenic exposure was also reduced by arsenic and selenium co-exposure. Furthermore, western blot analyses revealed that simultaneous exposure of both metals significantly inhibited autophagy which further suppressed apoptosis through positively regulation of key proteins p-mTOR, p-Akt, p-Foxo1A, p62, and expression of ubiquitin, Bax, Bcl2, NFкB, and caspases 3 and 9, although those are negatively regulated by arsenic. In addition, reverse transcriptase PCR analysis confirmed the involvement of caspase cascade in cell death process induced by arsenic and subsequent inhibition by co-exposure of selenium with arsenic. The cellular accumulation study of arsenic in presence/absence of selenium via inductively coupled plasma mass spectrometry confirmed that selenium effectively retarded the uptake of arsenic in PC12 cells. Finally, these findings imply that selenium is capable to modulate arsenic-induced intrinsic apoptosis pathway via enhancement of mTOR/Akt autophagy signaling pathway through employing antioxidant potentials and through inhibiting the cellular accumulation of arsenic in PC12 cells. Selenium is capable to modulate arsenic-induced intrinsic apoptosis pathway via enhancement of mTOR/Akt autophagy signaling pathway in PC12 cells.

    Polyphenols have many beneficial effects and an effective disease therapeutic auxiliary drug. Previously, myricetin, a polyphenol, had been reported to possess various biological effects on human physiology. However, mechanism of myricetin on apoptosis induced in PC12 cells is still unclear. PC12 cells were treated with myricetin in two concentration levels comprising 0.1 and 1 μM under serum-free condition. As a result, morphological changes were observed using trypan blue assay. DNA fragmentation was determined by DNA ladder assay to evaluate DNA damage levels. Western blotting results showed that cytosolic cytochrome c which was released from mitochondria. Subsequently, tumor suppressor gene p53, pro-apoptotic and anti-apoptotic Bcl-2 family proteins Bax and Bcl-2 were expressed. The caspase cascade reaction was induced through caspase 3 and 9 expression. From these results, it is suggested that myricetin significantly enhanced the apoptosis induced by serum deprivation in a dose-dependent manner in PC12 cells.

    With the development of nanotechnology, silver nanoparticles (Ag-NPs) have become one of the most in-demand nanoparticles owing to their exponential number of uses in various sectors. The increased use of Ag-NPs-enhanced products may result in an increased level of toxicity affecting both the environment and living organisms. Several studies have used different model cell lines to exhibit the cytotoxicity of Ag-NPs, and their underlying molecular mechanisms. This review aimed to elucidate different properties of Ag-NPs that are responsible for the induction of cellular toxicity along with the critical mechanism of action and subsequent defense mechanisms observed in vitro. Our results show that the properties of Ag-NPs largely vary based on the diversified synthesis processes. The physiochemical properties of Ag-NPs (e.g., size, shape, concentration, agglomeration, or aggregation interaction with a biological system) can cause impairment of mitochondrial function prior to their penetration and accumulation in the mitochondrial membrane. Thus, Ag-NPs exhibit properties that play a central role in their use as biocides along with their applicability in environmental cleaning. We herein report a current review of the synthesis, applicability, and toxicity of Ag-NPs in relation to their detailed characteristics.

    Humic acid (HA) has been implicated as a contributory factor for blackfoot disease, which is an endemic peripheral vascular disease. We investigated the effect of HA on the regulation of endothelial nitric oxide (NO) synthase (eNOS) in human umbilical vein endothelial cells (HUVECs) to evaluate the involvement of eNOS and related factors in peripheral vascular impairment with HA exposure. Treatment of HUVECs with HA induced upregulation of eNOS. This result coincides with those of previous studies. Furthermore this is the first study to report that HA induces upregulation of heat shock protein (Hsp)90, Hsp90, eNOS phosphorylation at Ser1177, and eNOS phosphorylation at Thr495, as compared to that in the control. In contrast, treatment with BAPTA, an intracellular Ca2+ chelator, inhibited upregulation of these proteins induced by HA. This study demonstrates that HA treatment leads to increases in both Hsp90 and Hsp90 proteins and indicates that Hsp90 leads to eNOS phosphorylation at Ser1177 and that Hsp90 leads to eNOS phosphorylation at Thr495, respectively. Upregulation of eNOS, Hsp90, and Hsp90 in HUVECs is regulated by intracellular Ca2+ accumulation induced by HA. These results suggest that upregulation of eNOS phosphorylation at Ser1177 and eNOS phosphorylation at Thr495 produce NO and superoxide anions, respectively, resulting in generation of peroxynitrite, which causes impairment of vascular endothelial cells.     When peat forest fires happen, it leads to burn soil and also humic acids as a dominant organic matter contained in peat soil as well as the forest. The structure and properties of humic acids vary depending on their origin and environment, therefore the transformation of humic acid is also diverse. The impacts of the peat fires on peat soil from Central Kalimantan, Indonesia were investigated through the characterization of humic acids, extracted from soil in burnt and unburnt sites. The characterization of humic acids was performed by elemental composition, functional groups, molecular weight by HPSEC, pyrolysate compounds by pyrolysis-GC/MS, fluorescence spectrum by 3DEEM spectrofluorometer, and thermogravimetry. The elemental composition of each humic substance indicated that the value of H/C and O/C of humic acids from burnt sites were lower than that from unburnt sites. The molecular weight of humic acids from burnt sites was also lower than that from unburnt sites. Pyrolysate compounds of humic acids from unburnt sites differed from those of humic acids from burnt soil. The heating experiment showed that burning process caused the significant change in the properties of humic acids such as increasing the aromaticity and decreasing the molecular weight.

    The impacts of peat fire on the water quality were investigated by comparing the water quality of the Sebangau River and the Canal Kalampangan. pH and DOC are important parameters related to the specific properties of water in Central Kalimantan. The pH value of the Sebangau River and the Canal water are about 4. The average concentration of DOC in the Sebangau River was about 43.8 mg/L, while that in the Canal was about 37.2 mg/L. The DOC concentration in the Canal was lower than that in the Sebangau River, it was supposed that peat soil around the Canal had been burnt, and therefore the supply of dissolved organic matters to the Canal decreased. In this chapter, it is also shown that DOC concentration of soil collected from burnt area was lower than that from unburnt area. It was found that the tap water in Palangka Raya contained high concentration of NH4-N and DOC, including humic substances. Polyaluminium chloride (PAC) with CaCO3 was one of the effective coagulants that could reduce humic substances contained in tap water in Palangka Raya, to more than 91 % removal. The assessment of the toxicity of humic acid using the trypan blue exclusion method is also discussed in this chapter. The peat fire also influenced the chemical characteristics of aquatic humic substances (fulvic acid and humic acid), especially in the H/C and O/C value from elemental analysis data, molecular weights, and 3DEEM fluorescence spectra. The effect of peat fire on the properties of soil organic matter was investigated, and it was clarified that the peat fire affected not only on surface soil but it reached into the subsurface soil until 30-50 cm depth.

    Humic acid (HA), a group of high-molecular weight organic compounds characterized by an ability to bind heavy metals, is normally found in natural water. Although the impairment of vascular endothelial cells in the presence of humic substances has been reported to be involved in some diseases, the mechanisms responsible for this involvement remain unclear. In this study, we examined the cytotoxicity of HA obtained from peatland in Central Kalimantan, Indonesia, to human vascular endothelial cells, as well as the mechanisms behind these effects. It was found that 50 mg/L HA showed cytotoxicity, which we considered to be mediated by apoptosis through the mitochondrial pathway because of an increase in the expression of caspases 6 and 9 in response to HA administration. In addition, this cytotoxicity was enhanced when cells in this experimental system were exposed to oxidative stress, while it was decreased by the addition of vitamin C. Thus, we conclude that the apoptosis induced by HA depends upon oxidative stress. Furthermore, an iron chelator, DFO, showed a tendency to decrease HA-induced cytotoxicity, suggesting that iron may potentially mediate HA-induced oxidative stress. In conclusion, long-term consumption of HA-rich water obtained from our study area may cause damage to endothelial cells and subsequent chronic health problems. (C) 2012 Wiley Periodicals, Inc.

    Despite extensive research on copper toxicity the mechanisms involved are not fully characterized. There have been many recent reports concerning the relationship between epigenetic factors and cell metabolism, but the effects of copper exposure on epigenetic factors have not been investigated. In this study, an in vitro culture system was employed to study the influence of copper on apoptosis and epigenetic factors in PC12 cells. When PC12 cells were exposed to copper, DNA damage was observed as DNA fragmentation. In addition, cytosolic cytochrome c levels were increased by copper treatment. These results suggested that copper induced apoptosis via an oxidative stress pathway. This was consistent with the observation that copper-induced apoptosis was enhanced by further oxidative stress induced by exposing cells to H2O2. In addition, the epigenetic factors were significantly increased in apoptotic cells following exposure to copper and oxidative stress.

    The pollution of water by heavy metals is one of the most serious problems in the developing countries, where watercourses play important roles in transport and economic activities. The aim of this study was to examine whether Hyposarcus pardalis, a fish species widespread in the freshwater environment in Indonesia, could be used as a biomarker for environmental pollution by metals. To this effect, the concentrations of metallothioneins and metals in the livers and kidneys of H. pardalis were measured. In addition, to clarify the relationship between metallothionein concentrations and metal exposure, the concentrations of metallothioneins and metals were determined in the liver and the kidney of fish exposed to 50 and 500 ppb Cu and 500 ppb Mn, compared with those kept in clean water. Sufficient concentrations of metallothionein were detected in fish captured from Lake Rawakalong located in an industrial area in the suburbs of Jakarta. The results of exposure experiments suggested that H. pardalis retained a history of pollution in its organs for a long duration, and the metals bound to metallothioneins in the liver and kidney could be replaced with Cu following exposure. In conclusion, the hepatic and renal metallothioneins in H. pardalis are a useful candidate biomarker for monitoring heavy metal contamination.

    6R-l-Erythro-5,6,7,8-tetrahydrobiopterin (BH4) is an essential cofactor for tyrosine hydroxylase (TH) activity and is a risk factor for cognitive decline and brain atrophy. Previous studies have shown that the decline in TH activity in the cerebral cortex of senescence-accelerated mouse prone 10 (SAMP10) mice is caused, at least in part, by a decrease in Fe, ferritin, and TH phosphorylation. We determined the concentrations of BH4 and the enzymes GTP cyclohydrolase-1,6-pyruvoyltetrahydropterin synthase and sepiapterin reductase (SPR) in the de novo pathway of BH4 biosynthesis. Dihydrofolate reductase (DHFR), which converts BH2 to BH4 in the salvage pathway of BH4 synthesis was also determined in the cerebral cortex of SAM mice at 3 and 12 months of age. The BH4 concentration was measured by HPLC, and the protein levels of enzymes involved in BH4 synthesis were measured by western blot analysis. At 12 months of age, BH4 concentration in the cerebral cortex of SAMP10 mice showed significantly higher values as compared to that of control mice. Further, the protein level of SPR in SAMP10 mice was significantly higher than that in SAMR1 mice at 3 and 12 months of age. In contrast to SPR, the protein level of DHFR in SAMP10 mice was significantly lower than that in SAMR1 mice. These results indicate that abnormal regulation of BH4 metabolism occurs in the cerebral cortex of SAMP10 where the dysfunction of the salvage pathway of BH4 synthesis may cause overproduction of BH4 through the de novo pathway, which is considered characteristic in the cerebral cortex of SAMP10 with aging. Therefore, there is a possibility that the excess amounts of BH4 lead to age-related brain dysfunction in the cerebral cortex of SAMP10. © 2013 Springer Science+Business Media Dordrecht.

    The purpose of this study was to elucidate the alteration of catecholamine metabolism and the contribution of catecholamines to the decline of learning and memory in the brain of the senescence-accelerated mouse prone 10 (SAMP10) with aging. Catecholamines and their metabolites in the cerebral cortex were measured by HPLC-ECD. The protein levels of tyrosine hydroxylase (TH) as well as TH phosphorylated at Ser19 or Ser40, dopamine-beta-hydroxylase (D beta H), and cAMP-dependent protein kinase (PKA) were determined by western blot analysis. Dopamine (DA) and norepinephrine (NE) levels in SAMP10 were significantly lower than those in control animals. However, no significant difference was observed in catecholamine metabolite levels between SAMP10 and control mice. The level of TH phosphorylation at Ser40 in SAMP10 was significantly lower than that in control mice, but no significant difference was observed in the levels of TH, TH phosphorylated at Ser19, or D beta H. The amount of PKA, which regulates the phosphorylation of TH at Ser40, was significantly lower in SAMP10 than in control mice. The present study demonstrated that a decline in DA and NE concentrations was observed in the cerebral cortex of SAMP10 with aging, and this decrease of catecholamine levels was caused by impairment of their synthetic pathway. These impairments are considered to be caused by downregulation of TH phosphorylation at Ser40 as a result of PKA deficiency. The present study suggests that the decline of learning and memory abilities of SAMP10 is caused by a decrease in catecholamine synthesis in the cerebral cortex with aging.     To compare water quality in rivers of developed and developing countries, a study based on physicochemical parameters and dissolved metals levels was conducted. Water samples were collected from selected sites in Dhaka, Bangladesh; Hokkaido and Osaka, Japan; Erdenet, Mongolia and West Java, Indonesia. Analysis of least significant differences revealed that most water quality parameters were within comparable low levels in both developed and developing countries. The dissolved metals concentrations were found to be similar and below those of water standards except for manganese and cadmium at every sampling point, and lead in Erdenet, Mongolia. Some metals showed high enrichment factors in the rivers of Osaka, Japan and Erdenet, Mongolia, indicating accumulation possibility of metals in the river-bed sediments. High concentrations of dissolved organic carbon, Escherichia coli and dissolved metals suggested greater water pollution in some rivers of developing countries than in the rivers of Japan. Principal component analysis showed strong correlations between dissolved organic carbon and chemical oxygen demand and conductivity and total dissolved solids at each sampling point, and E. coli, nitrate (NO?3-), nitrite (NO?2-), and pH levels were found to be higher in the rivers of Dhaka and Erdenet. In addition, there were high levels of Al and Zn in West Java, Pb in Erdenet, and Mn, Fe, and Cr in the rivers of Dhaka and Japan. Based on pressures and impacts, it is evident that dissolved metal, organic, and fecal pollution in the rivers of developing countries are in somewhat dreadful condition in comparison with the rivers of developed country.

    The adsorption of cadmium and chromium from aqueous solutions with epichlorohydrin cross-linked , , and -cyclodextrin polymers (CDPs) was investigated under a wide range of experimental conditions including pH, metal concentration, and CDP amounts. Recent studies have shown that , , and -CDPs follow the Freundlich, Langmuir, and BrunauerEmmettTeller models and are capable of reducing heavy metal ion concentrations in water to desirable levels with enhanced sorption capabilities. The removal of cadmium and chromium achieved equilibrium in 4h. The pH of effective cadmium removal was 6.5 to 7.5, and phosphate buffer favored the removal of cadmium in the presence of potassium ion. Chromium removal was optimal at pH 3.6 to 6.5. Current technologies can remove 99% cadmium ions from solution, yet this process is not very efficient for chromium ions. Furthermore, a small amount of CDPs can remove large quantities of heavy metal ions and can then be regenerated with acid for reuse. The present results are promising for using inexpensive CDPs as a low-cost material that is effective in remediating waters contaminated with heavy metal species. The sorption kinetics of CDPs along with the conditions to adsorb cadmium and chromium are reported here for the first time.

    In this study, to examine the mechanism of diethyl phthalate toxicity to cells, the effects of diethyl phthalate on apoptosis in a PC12 cell system were investigated by assaying apoptotic factors such as caspase-3, Bax, cytochrome c and DNA damage. Diethyl phthalate was shown to enhance the apoptosis induced by serum deprivation according to the results of DNA electrophoresis and TUNEL signal assays, although it could not induce apoptosis itself in the cells. This enhancement was thought to be because of an increase in caspase-3-like activity. In addition, the expression of bax and contents of cytochrome c in the cytosol showed a tendency to increase the cells exposed to diethyl phthalate. These results indicated that diethyl phthalate, a potential endocrine disrupter, affects the apoptotic system in PC12 cells. Diethyl phthalate may enhance oxidative stress such as that induced by reactive oxygen species in PC12 cells.

    In the laboratory, using a PC12 cell system, studies have been conducted on the effects of various chemicals on apoptosis, as it is considered to be an essential part of normal development, maintenance, and defense in organisms. Stevioside is a natural sweetener extracted from the leaves of Stevia rebaudiana. Since it is widely used as a sugar replacement, it was decided to evaluate the toxicological effects of low concentrations of stevioside on apoptosis induced by serum deprivation using the PC12 cell system. It was found that based on data from DNA electrophoresis and TUNEL signal assays stevioside enhanced apoptosis induced by serum deprivation. This enhancement was caused by increased expression of Bax and of cytochrome c released into the cytosol. These findings suggest that stevioside affects the regulation of the normal apoptotic condition. Further investigation will be needed to clarify the detailed mechanism of the enhancement due to the treatment with stevioside.

    To determine the water environment and pollutants in West Java, the contents of metals and general water quality of the Ciliwung River in the Jakarta area were measured. High Escherichia coli number (116-149/mL) was detected downstream in the Ciliwung River. In addition to evaluate mercury pollution caused by gold mining, mercury contents of water and sediment samples from the Cikaniki River, and from paddy samples were determined. The water was not badly polluted. However, toxic metals such as mercury were detected at levels close to the baseline environmental standard of Indonesia (0.83-1.07 mu g/g of sediments in the Cikaniki River). From analyses of the paddy samples (0.08 mu g/g), it is considered that there is a health risk caused by mercury.

    An endocrine disrupter, bisphenol-A is widely used in the production of plastics and coatings. Recently, it was reported that bisphenol-A affected neurotransmitters in the mammalian brain. On the basis of these reports, it was considered that bisphenol-A affected neuronal differentiation. In this study, the morphological changes in nerve growth factor (NFG)-induced differentiation caused by bisphenol-A were confirmed using a PC12 cell system. When a low concentration of bisphenol-A was added to medium containing NGF, it inhibited neurite extension. In addition, to clarify whether bisphenol-A affects the early and late stages of the NGF-signaling pathway in cell differentiation, changes of phosphorylation of MAP kinases and cAMP-response element binding protein (CREB) in PC12 cells treated with and without BPA in medium containing NGF were investigated using western blot analysis.     In Java and Kalimantan in Indonesia, river water plays important roles in human life; for example, for transportation, and economic activities of the inhabitants. However, industrial, agricultural and domestic water is discarded into rivers directly in many developing countries, including Indonesia, since drainage systems have not been completely constructed. In this study, to evaluate the water quality and to compare those levels of environmental contaminants in developing and developed countries, water quality and contents of endocrine disrupters were measured in a total of 64 water samples (Indonesia; 28 samples and Japan; 36 samples) from 53 sites. The results indicated that, rivers in both capital cities, Jakarta and Tokyo, were contaminated. Water in rivers in Indonesia was not so heavily polluted as in Japan. Pollution in the river water in Indonesia appeared to be caused by the lack of sewerage systems. In addition, the findings on endocrine disrupters indicated that the concentration of alkylphenol in water samples was large enough to affect living organisms.

    Copper, an essential trace element, induces apoptosis in mammalian cells. However, the precise mechanism of copper-induced apoptosis is still unclear. In this study, to determine the apoptotic pathway initiated by copper treatment, apoptotic factors such as Bax, Bad and Bcl-2, and the caspase family in PC 12 cells treated with copper were measured by Western blot and RT-PCR analyses. The expression of Bax, Bad, cytochrome c and caspases 3 and 9 were increased by copper treatment. From these results, two pathways for copper-induced apoptosis were suggested. At first, an increase of Bax induces the release of cytochrome c from the mitochondria into the cytoplasm owing to binding to apoptotic activating caspase 9 leading to the activation of caspases 3. In the other pathway an increase of Bax and reactive oxygen species activates the release of AIF from the mitochondria. The AIF induces apoptosis via a caspase-independent pathway.     In November 2005, Hokkaido University held the 8th Workshop on Education. This workshop was introduced to improve the education of Hokkaido University and it has been held once a year since 1998. The main theme of this year's workshop was to plan courses respecting the requirements for credits more rigidly. Our basic understanding about credits given in a university is that a credit corresponds to 45 hour study of students,including lectures, preparations,reviews,assignments etc. The workshop had (1) three sessions about planning of courses,each of which consisted of minilectures,small group discussions and a general discussion,and (2) two lectures,one of which was about the purpose of faculty development and the other was about the meaning of 'respecting the requirements for credits more strictly' It had 30 participants from all over Hokkaido University,7 participants from three other universities and 8 persons of the task force,including the president of Hokkaido University and a lecturer,that is,45 persons in all.This report contains an explanation about what was done in the workshop,especially the syllabi of the courses worked out in the sessions.

    北海道大学では,2006年度に入学してくる新カリキュラム卒業生に対応した初習理科教育を2004年度から試験的に導入している。そのうち「基礎物理学」では,e-Learningシステムの利用,大人数講義,演示実験,演習の導入,TAの活用など,これまでにない教育手法を採用し,効果的な授業を展開している。この教育手法を紹介するとともに,今後検討すべき課題も提示する。

    Metallothionein-3/growth inhibitory factor is a brain-specific member of the metallothionein gene family, and impairs the survival and neurite formation of cultured neurons. Metallothionein-3 can bind heavy metals such as Zn, Cu, or Cd almost in the same way as other metallothionein family. However, its biological function as growth inhibitory factor apparently distinguishes metallothionein-3 from other metallothioneins. To better understanding of the relationship between the growth inhibitory activity of metallothionein-3 and metals bound to metallothionein-3, the metal-binding ability of metallothionein-3 was analyzed in comparison with those of metallothionein-1 and -2. The metal-binding ability of metallothionein-3 was evaluated by pH titration and 5-5' dithiobis (2-nitrobenzoic) acid (DTNB) analysis as compared with those of the other metallothioneins. The affinity of metal ions for metallothionein-3 was indicated as follows, Cu>Cd>Zn, same as metallothionein-1 and -2. However, the affinity of metallothionein-3 to Cu was much higher than that of metallothionein-1 and -2. The strong affinity to Cu of metallothionein-3 might be related to its growth inhibitory activity.

    We examined synaptic plasticity in the optic tectum of rainbow trout by extracellular recordings. We found that the field-excitatory postsynaptic potential in the retinotectal synapses was potentiated by repetitive stimuli of 1.0 Hz for 20 s to the retinotectal afferents. The long-term potentiation (LTP) developed slowly, and was maintained for at least 2 h. Applications of an antagonist for N-methyl-D-aspartic acid (NMDA) receptors or Mg2+-free saline showed that activation of NMDA receptors was required to form the UP beyond the induction period. The present findings indicate that presynaptic stimulation in the retinotectal synapses causes LTP mediated by NMDA receptors in the optic tectum of rainbow trout. (C) 2004 Elsevier Ireland Ltd.     Although nonylphenol is well known as an endocrine disrupting chemical, there is little information concerning biological effect of nonylphenol. In this study, we investigated effect of nonylphenol on apoptosis induced by serum deprivation in PC 12 cells using TUNEL and DNA fragmentation assays. In addition, changes in contents of proapoptotic factors, Bad and Bax, and antiapoptotic factor, Bcl-2, and enzyme activity of caspase-3 were studied. Below 100 ng/ml of nonylphenol increased TUNEL signals, DNA fragmentation and content of proapoptotic factor, Bad as compared to those by serum deprivation without nonylphenol. Furthermore, addition of nonylphenol enhanced caspase-3 activity and Z-VAD, caspase-3 inhibitor, diminished such effect. These results indicated that below 100 ng/ml of nonylphenol enhanced apoptosis induced by serum deprivation via caspase-3 activation in PC12 cell. (C) 2004 Elsevier Inc. All rights reserved.

    Objectives: In the present study, it was examined whether the metal-binding dopamine melanins and the metal-free dopamine melanin showed SOD-like activities, al though eumelanin and pheomelanin protect against oxygen toxicity caused by their superoxide dismutase activity, physiological roles of neuromelanin, dopamine melanin, have not been elucidated. Materials and methods: Dopamine melanin was prepared by the method of oxidative polymerization. Results: Both Cu- or Fe-binding dopamine melanin was proved higher SOD-like activity as compared to Zn-binding and metal-free dopamine melanins. This activity of Cu- or Fe-binding dopamine melanin was increased depending on these metal contents. In contrast, the SOD-like activity of Zn-binding dopamine melanin did not depend on Zn contents. Conclusions: It is to be expected that neuromelanin protects against oxidative stress by both O-2-scavenging and transition metal-binding abilities in the brain.

    Macrophage migration inhibitory factor (MIF) is a molecule known to regulate macrophage accumulation at sites of inflammation. To elucidate the role of MIF in progression of liver fibrosis, the immunohistochemical localization of MIF and macrophages in the liver were examined. Male Wistar rats received thioacetamide (TA) injections (200 mg/kg, i.p.) for 1 or 6 weeks. In biochemical and histological tests, it was confirmed that liver fibrosis was induced. In immunohistochemical analyses, the expression of MIF protein was seen in hepatocytes in the areas extending out from the central veins to the portal tracts. In particular, at 6 weeks, immunoreactivity was detected in degenerated hepatocytes adjacent to the fibrotic areas but hardly observed in the fibrotic areas. On the other hand, a number of exudate macrophages stained by antibody ED1 were seen in the areas from the central veins to the portal tracts at 1 week and in the fibrotic areas at 6 weeks. Macrophages also showed a significant increase in number as compared with controls. These results revealed that there was a close relationship between the appearance of MIF expression and ED1-positive exudate macrophages in degenerated hepatocytes during the progression of TA-induced liver fibrosis.

    Spatio-temporal patterns of neuronal activity before and after the induction of long-term potentiation in mouse hippocampal slices were studied using a real-time high-resolution optical recording system. After staining the slices with voltage-sensitive dye, transmitted light images and extracellular field potentials were recorded in response to stimuli applied to CA1 stratum radiatum. Optical and electrical signals in response to single test pulses were enhanced for at least 30 minutes after brief high-frequency stimulation at the same site. In two-pathway experiments, potentiation was restricted to the tetanized pathway. The optical signals demonstrated that both the amplitude and area of the synaptic response were increased, in patterns not predictable from the initial, pretetanus, pattern of activation. Optical signals will be useful for investigating spatio-temporal patterns of synaptic enhancement underlying information storage in the brain.

    Seven kinds of angiotensin I-converting enzyme (ACE) inhibitory peptides were isolated from the hydrolysates of wakame (Undaria pinnatifida) by Protease S "Amano" (from Bacillus stearothermophilus) by using three-step high-performance liquid chromatography (HPLC) on a reverse-phase column. These peptides were identified by amino acid composition analysis, sequence analysis, and liquid chromatography-mass spectrometry (LC-MS), as Val-Tyr (IC50 = 35.2 muM), Ile-Tyr (6.1 muM), Ala-Trp (18.8 muM), Phe-Tyr (42.3 muM), Val-Trp (3.3 muM), Ile-Trp (1.5 muM), and Leu-Trp (23.6 muM). These peptides have resistance against gastrointestinal proteases in vitro. Each peptide was determined to have an anti hypertensive effect after a single oral administration in spontaneously hypertensive rats (SHR). Among them, the blood pressure significantly decreased by Val-Tyr, Ile-Tyr, Phe-Tyr, and Ile-Trp in a dose of 1 mg/kg of body weight (BW). The present study showed that antihypertensive effect in the hydrolysates of wakame by Protease S "Amano" was attributed to these peptides.

    We examined the linkage of nitric oxide (NO)-induced apoptosis to acceleration of brain aging of senescence-accelerated mouse prone 10 (SAMP10). The expression of neuronal nitric oxide synthase (nNOS) increased in the cerebral cortex of the brain of SAMP 10 in an age-dependent manner and significantly higher levels of neuronal nitric oxide synthase (nNOS) were observed in both young and old SAMP10 as compared to age-matched controls. Moreover, a lower level of anti-apoptotic protein Bcl-2 and a higher level of pro-apoptotic protein cytochrome c in cytosol were observed in SAMP10 compared to the control. However, there was no significant difference in the expression of pro-apoptotic protein p53 between SAMP10 and the control. Furthermore, terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL)-positive apoptotic cells were more abundant in the cerebral cortex of aged SAMP10 than in the control. The present results suggest that an age-dependent increase of NO by up-regulation of nNOS promotes the Bcl-2-linked apoptosis in the cerebral cortex of SAMP10 and this may cause the acceleration of brain aging of SAMP10.     The present study was carried out to investigate the effect of gold (Au) injection on copper (Cu) and two types of ceruloplasmin (Cp), total Cp (ID1) and active Cp (ID2), metallothionein (MT) in the serum, kidney and liver, and 8-hydroxydeoxyguanosine (8-OHdG) in the rat kidney. The Cu contents in sera and kidneys of Au-injected rats were 1.7 and 5.5 times higher than those in sera and kidneys of control rats, respectively. The most of Cu in the sera of the control rats or Au-injected rats were observed in the Cp fractions from a Sephacryl S-200 column. The Cu concentration in the Cp fractions was increased by Au injection. Significant increases of ID1 and ID2 were found in the sera of the control rats and Au-injected rats, while there was no significant difference in those concentrations of livers or kidneys between the control rats and Au-injected rats. Our results indicated that the most of Cp existed as active ID1. The immunoreaetivity of 8-OHdG was located in the cortex of the Au-injected rat. These results indicated that the oxidative DNA damage occurred in the renal cortex of the Au-injected rat and the localization of DNA damage did not coincide with that of Cu-MT. These findings suggest that the oxidative DNA damage in the kidneys of rats injected with Au is associated with Cu except Cu-MT. (C) 2002 Elsevier Science Ireland Ltd. All rights reserved.

    This study was undertaken to investigate the effect of taurine on the infiltration of macrophages and the progression of renal interstitial fibrosis in the kidneys of rats treated with cisplatin (CDDP). Male rats in different groups were treated as follows: (1) saline as control, (2) CDDP and (3) CDDP plus taurine in drinking water. At weeks 2, 4 and 6 after the fifth CDDP injection, we examined platinum content, the kinetics of macrophages and the areas of fibrosis. In the histologic analyses, fibrotic areas were found to have developed around dilated or atrophic tubules in the corticomedullary junction in the kidneys of CDDP-treated rats, while CDDP-plus-taurine-treated rats showed a reduction of the extent and magnitude of damage. These findings reflect the fact that the percentage of fibrotic areas in the kidney of CDDP-plus-taurine-treated rats was significantly lower than in that of CDDP-injected rats throughout the recovery period (P<0.05). Compared with normal rats, the macrophages in CDDP-injected rats showed significant increases in number at weeks 2, 4 and 6 (P<0.05). In contrast, the number of macrophages in CDDP-plus-taurine-treated rats was significantly smaller than that of CDDP-injected rats. These results suggested that taurine suppressed the increase of infiltrating macrophages, and led to the attenuation of renal interstitial fibrosis (P<0.05).

    To understand the possible physiological roles of the neuromelanins, we examined effects of synthetic dopamine-melanins on oxygen radical formation by metal ions with dopamine. Oxygen radical formation was detected by DNA cleavage analysis. Addition of metal-free synthetic dopamine-melanins inhibited DNA cleavage induced by Fe or Cu ions with dopamine. On the other hand, in the presence of dopamine, dopamine-melanins loaded with Cu and Fe induced DNA cleavage without addition of free metal ions. It was clarified that dopamine-melanins loaded with Cu and Fe generated hydroxyl radicals in the presence of dopamine, on the basis of the findings that catalase and mannitol inhibited the DNA cleavage. Our results suggest that metal-free neuromelanins inhibit hydroxyl radical formation induced by metal ions with dopamine by sequestering metal ions.

    2,4,5-Trichlorophenoxyacetic acid (2,4,5-T) is an endocrine disrupter that exerts cytotoxic effects on organisms. In this study, the influence of 2,4,5-T at low concentrations on apoptosis in PC12 cells was investigated. Although no apoptotic features were observed in PC12 cells treated with 2,4,5-T, it inhibited the DNA fragmentation induced by serum deprivation. In addition, the cell viability of PC12 cells increased after treatment with 2,4,5-T. In conclusion, 2,4,5-T suppressed the apoptosis of the cultured cells. Since apoptosis is a morphological and biochemical description of a physiological mechanism of cell death that is commonly associated with programmed events necessary for development of individuals and organs, the inhibitory effect of 2,4,5-T on apoptosis might cause serious damage to cell homeostasis and differentiation.     Effects of styrene exposure on middle latency auditory-evoked potentials (MAEPs), which reflect the central auditory function, were investigated. As significant delay in latencies of No. Po, Na, and Pa components and increases in the peak to peak intervals of MAEPs, but not amplitudes, were observed in a dose-dependent manner. The present and our previous studies demonstrate that electrophysiological alterations occur before neuronal cell damage induced by styrene exposure. The present results suggested that styrene has a marked effect on the processing of auditory information related to MAEPs

    The combined effects of city noise and luminance of the computer display were evaluated from the changes in lymphocytes and mental activities of participants. Healthy male students were tested under the following four experimental conditions: (1) a calculating task on a video display terminal (VDT) with luminance of 90 cd m(-2) without city noise; (2) a calculating task on a VDT with luminance of 20 cd m(-2) without city noise; (3) a calculating task on a VDT with luminance of 90 cd m(-2) with city noise of 70 dB(A); and (4) a calculating task on a VDT with luminance of 20 cd m(-2) with city noise of 70 dB(A). A visual reaction test (VRT) was performed, and critical flicker fusion frequency (CFF), heart rate (HR), numbers of circulating white blood cells (WBCs), lymphocyte subsets and subjective symptoms of fatigue were measured (1) before; (2) just after; and (3) 30 min after each 60 min test. Subjective symptoms of fatigue significantly increased just after experiments conducted under the two noisy conditions. VRT and CFF showed significant changes in the case of the high-luminance display with noise. WBCs and neutrophils showed significant increases in the two quiet conditions. These results suggested that high luminance with noise had the most effect on subjective fatigue and mental activities.

    We examined the distribution and induction of Cu-metallothionein (MT) in the liver of the rats with thioacetamide-induced cirrhosis. The cirrhotic rats received intraperitoneal injections with or without Cu. Subsequently, immunohistochemical assessments of MT, metal analyses and purification of MT by gel filtration were carried out. By immunohistochemistry, the presence of MT in the parenchymal areas but not in the fibrotic areas of the cirrhotic liver of Cu-injected rats was confirmed. In Cu-injected cirrhotic rats, the Cu contents in the liver increased from 3.74 +/- 0.06 (means +/- SE) mug/g to 89.44 +/- 7.89 mug/g, depending upon the dose of Cu. The Zn contents in the liver also increased significantly. The results of gel filtration also showed that the concentrations of Cu in MT fractions obtained from the cirrhotic liver increased with the doses of Cu, whereas the Zn in MT fractions hardly increased. The results obtained from this study suggested that although Cu-MT was induced in the cirrhotic liver following Cu administrations, Cu metabolism in the parenchymal and fibrotic areas may be altered.

    The effect of trichloroethylene (TCE) on long-term potentiation (LTP) was studied using both electrical and optical recording. The hippocampi from mice injected with 300 mg/kg or 1000 mg/kg TCE were sliced 24 h after administration. The field potential from the CAI was recorded. After the application of tetanus, population spikes (PS) were potentiated in all groups, but the post-per-pre ratio of PS was smaller in TCE groups than in the control. Optical recording was also carried out in 1000 mg/kg TCE-injected mice and a new analytical method using a high speed camera was employed. After the induction of tetanus, the optical signal was potentiated in both TCE and control groups. However, the post-per-pre ratio of the optical signals and response area were smaller in the TCE groups than in the control. It was suggested that the impairment of LTP is one of the mechanisms of the impairment of immediate memory after acute exposure to TCE in humans.

    Effects of styrene exposure on middle latency auditory-evoked potentials (MAEPs), which reflect the central auditory function, were investigated. As significant delay in latencies of No, Po, Na, and Pa components and increases in the peak to peak intervals of MAEPs, but not amplitudes, were observed in a dose-dependent manner. The present and our previous studies demonstrate that electrophysiological alterations occur before neuronal cell damage induced by styrene exposure. The present results suggested that styrene has a marked effect on the processing of auditory information related to MAEPs.

    This study was undertaken in order to investigate the effect of zinc (Zn) administration on induction of Zn-binding metallothionein in rat liver with thioacetamide-induced cirrhosis, and the localization of metallothionein in the liver. Normal and cirrhotic rats received intraperitoneal injections with or without Zn. Subsequently, metal analyses, purification of metallothionein by gel filtration and immunohistochemical assessments of metallothionein were carried out. Although in Zn-injected cirrhotic rats, the Zn contents in the liver and plasma increased significantly depending upon the dose of Zn, the Zn contents in the liver and plasma of the cirrhotic rats were lower than those of normal rats after the same dose of Zn. The results of gel filtration also showed that the levels of Zn-metallothionein in the cirrhotic liver were reduced in comparison with those of the normal liver. By the immunohistochemical method, the presence of metallothionein in the parenchymal areas but not in the fibrotic areas of the cirrhotic liver was confirmed. These results suggested that the induced metallothionein was only located in the parenchymal areas. The metallothionein induced in the parenchymal areas was considered to play a role in protecting the parenchymal cells against the progression of fibrosis, because metallothionein has been thought to be involved in the cellular defense against oxidative stress.

    We examined the link of hippocampal Zn to the functional impairments with aging using senescence-accelerated mouse prone 10 (SAMP10) with deficits in learning and memory. Zn in hippocampal mossy fiber pathway was less distributed in aged SAMP10 than that in the age-matched control. Furthermore, expression of Zn transporter 3, ZnT3, which plays to accumulate Zn in synaptic vesicles in the mossy fiber pathway, was markedly reduced in the hippocampal region even in young SAMP10. Moreover, excessive presynaptic release of glutamate as well as glycine and expression of glial fibrillary acidic protein, a marker of neuronal cell injury, were observed in the hippocampus of aged SAMP10 compared to the control. The present results suggest that age-dependent deficiencies of Zn in synaptic vesicles of the mossy fiber pathway induced by low expression of ZnT3 cause glutamatergic excitotoxicity in the hippocampal neurons and the deterioration of learning and memory in SAMP10.     It has not been fully elucidated how endocrine-disrupting chemicals disrupt hormone functions or how strong their effects are compared with natural hormones. There is little information concerning the effects of tributyltin (TBT), one of the endocrine disrupters on living organisms. Although TBT at high cocentration induced apoptosis in PC12 cells, TBT at low concentration inhibited the DNA fragmentation in the cells cultured in serum-free medium or in medium containing 6-hydroxydopamine. The cell viability grown in both medium conditions increased after treatment with TBT, These findings suggest that TBT exerted a apoptosis-inducing and -inhibiting effect, These diverse effect of TBT on apoptosis would cause serious damages on cell differentiation.     Cis-diamminedichloroplatinum (cis-DDP) has been used as an anticancer agent but it also causes nephrotoxicity. To study the cis-DDP metabolism and its effects, the induction of metallothioneins and DNA damage caused by cis-DDP were observed. Cis-DDP or trans-DDP was administered to seven-week-old Wistar male rats as three daily injections of 8.0 mg/kg body wt., intraperitoneally. Using the obtained kidneys, gel filtration assay, metal analysis, immunohistochemistry and terminal deoxy transferase-mediated deoxy uracil triphosphate nick end labeling (TUNEL) method were carried out to examine localization of metallothioneins and DNA damage caused by cis-DDP. Platinum (Pt) contents, 26.05+/-12.01 mu g/g body wt. and 51.29+/-4.59 mu g/g body wt. (average+/-S.E.) were detected in the kidney of rats injected with both cis- and trans-DDP, respectively. Metallothionein was detected in the cortex of the kidney in rats administrated cis-DDP or trans-DDP. The mRNA was also detected in the same region. On the other hand cis-DDP showed induction of DNA damage on the cells in the outer stripe of the outer medulla but trans-DDP did not show any damage. The region-induced DNA damage differed from that induced by metallothioneins. Cis-DDP is suggested to be mainly trapped at the proximal tubules by metallothioneins, and the rest of cis-DDP induces DNA damage at the outer stripe of the outer medulla. Metallothioneins are considered to contribute to the protection against cis-DDP in the rat cortex.

    Histochemical characterizations of Ag-induced metallothionein (MT) in the kidney of the rat have been reported. Ag, Cu and Zn contents increased in kidney and liver after Ag injection. In particular, the Cu content in kidneys increased dramatically after three injections of Ag. Sephadex G-75 elution profiles of the renal cytosol of rats injected with Ag revealed that the accumulated Cu in the kidney was bound to MT as were Ag and Zn. In addition, localization of Cu- and Ag-MT in the kidney was studied using autofluorescent signals, which are dependent on Cu- or Ag-thiol clusters, and immunohistochemistry. Although the MT induced by Ag was predominantly observed in the cortex of the kidney, some MT signals were also detected in the outer stripe of the outer medulla, as well as in the kidneys of LEC rats, an animal model of Wilson disease (a hereditary disorder of Cu metabolism). In these LEC rats, the Cu-MT also accumulated in the outer stripe of the outer medulla of the kidney. From these results, one possibility could explain that the Cu-MT detected in the outer stripe of the outer medulla in the kidney of Ag-injected rat was associated with the Cu transporter affected by Ag.     Immunohistochemical localization of Copper-transporting P-type ATPase (ATP7B), a gene product responsible for Wilson disease, was visualized in the brain tissues of the Long-Evans agouti rat in detail using tissue-blotting technique and confocal laser microscopy. The ATP7B was intensely detected in neuronal cells of the hippocampal formation, olfactory bulbs, cerebellum, cerebral cortex and nuclei in the brainstem in which high amounts of copper and cuproenzymes, dopamine beta hydroxylase and Cu,Zn-superoxide dismutase (Cu,Zn-SOD) were detected. The present results suggest that ATP7B plays key roles in neurotransmissions of catecholamine pathway and preventing brain tissues from injury by superoxide radicals to regulate the cellular Cu concentration and/or activities of cuproenzymes related to neurotransmissions and a free radical metabolism. Furthermore, it is reasonable to assume that neurotoxicity due to abnormal copper accumulation or irregular regulation of cuproenzymes in the critical brain regions by mutation of the ATP7B gene leads to neurological failures of Wilson disease.     We report for the first time that autofluorescent signals were found in the kidney of Long Evans Cinnamon rats, a model for Wilson's disease, under the excitation at 488 nm using a confocal laser microscopy. Autofluorescence signal was observed in lysosome-like organelle ill proximal convoluted tubule cells of S1 and S2 adjacent to glomerulus in the cortex as described previously. These fluorescent materials were identified as Cu-metallothionein (Cu-MT) by the detection specificity of Cu-MZ referred by the chemical properties of the protein such as quenching test using Hg (II) and immunohistochemistry using MT monoclonal antibody. In addition, the emission was able to be observed after fixation by formaldehyde. The visualization principle of the autofluorescent emission from Cu-MT and the availability of the presented technique using a confocal laser microscopy are discussed.

    In order to investigate the neuropathological effects on the central nervous system of the rat by styrene exposure, identification of glial fibrillary acidic protein (GFAP) was observed using immunohistochemical staining and confocal laser scanning microscopy in the rat treated with styrene at 400 mg/kg and 800 mg/kg for 2 weeks. Both styrene-treated groups showed a marked increase of GFAP expression in cerebral cortex and hippocampus compared with the control group. These changes showed a dose- and time-dependent manner. Increased expression of GFAP in cerebral cortex and hippocampus provided the effects of styrene on the structure of cerebral cortex and hippocampus objectively.

    Several aspects of a new, small and inexpensive fatigue assessment tool, the Portable Fatigue Meter (PFM), were studied. In the first experiment the effects of room illumination and distance between the PFM and the eyes on critical flicker fusion frequency (CFF) values were examined. The results revealed that the PFM CFF value varied significantly as a function of the distance between the indicator and the eyes, but it was not affected by room illumination. The effect of ageing on the PFM CFF value was assessed in a second experiment. The results suggested that PFM CFF values are significantly smaller in middle-aged females than in younger females.

    Using confocal microscopy in conjunction with microdrop application of Dil, we have imaged and measured individual dendritic spines of living hippocampal CA1 pyramidal neurons in acute brain slices, before and approximately 3 hr after induction of long-term potentiation by chemical means. Statistical analysis' of changes in the length of individual spines, and comparison with results of Monte Carlo simulations, suggests that two forms of structural change occur in chemically induced long-term potentiation: growth of a subpopulation of small spines, and angular displacement of spines, These changes could provide a structural basis for the expression of long-term potentiation.

    An epidemiologic survey was conducted of the 818 households in a community consisting of aggregated dwelling units in Tokyo, Japan, in order to assess the relationship between housing environment and psychological or physical health status of the female residents. Psychological health status was assessed by the 28-item General Health Questionnaire in Japanese translation. Main housing factors such as internal density (person-to-room ratio) and floor level (vertical location) of the dwelling unit did not have significant effects on the psychological health status of the sample of Japanese women, but the internal density generally did not exceed 1.5 persons per room. However, physical health status might be associated with floor level: women living at the highest levels complained of more symptoms than did those at lower floor levels, when the type of dwelling unit (i.e., high-rise, low or medium height building) was not taken into account. Poor psychological health status was found among women who were dissatisfied with the plan of their house or with the room arrangement, were anxious about earthquake or other accidents, perceived the house as inadequate for the children, were annoyed by indoor noise, and complained of outdoor noise.

    A number of theories on the cellular basis of learning and memory propose that long-term changes in synaptic strength are encoded by changes in the properties of dendritic spines. Such theories imply that individual spines retain their identity over time. Using a new technique for fluorescent labelling of synaptic structures together with confocal microscopy, we have found that the appearance of individual spines on viable cells remains unchanged over observations periods of up to 5 hours. Even in slices exposed to kainic acid at concentrations that abolished synaptic transmission, groups of spines remained identifiable on cells clearly damaged by the toxin.     The effects of background tone (BGT) on the waveform of the auditory evoked potentials obtained from nine male students exposed to 95 ms tone bursts were studied at 250 Hz, 500 Hz, 1 kHz, 2kHz and 4 kHz. The sound intensity of the toneburst was 80 dB (SL), and that of BGT was 60 dB (SL). A decrease in the amplitudes, and an increase in the latency of the N1 component with BGT, were shown. BGT had more effect on the N1 component than on the P2 component. Frequency changes affected the amplitude of the N1 component and the latency of the P2 component. © 1989.

    Copper can be found in many wastewater sources including, printed circuit board manufacturing, electronics plating, plating, wire drawing, copper polishing, paint manufacturing, wood preservatives and printing operations. Typical concentrations vary from several thousand ppm from plating bath waste to less than 1 ppm from copper cleaning operations. Copper can be removed from wastewater by precipitation as an insoluble hydroxide salt or by ion exchange. In this study, single batch adsorption experiment of copper derivatives, e.g. CuCl2.2H2O, Cu(CH3COO)2 and CuSO4.5H2O from aqueous solutions with epichlorohydrin linked α, β and γ cyclodextrin polyurethanes (CDPs) have been investigated for a wide range of experimental conditions: pH, metal concentration and CDP amounts. The aim of this study was to specify highly effective derivatives of copper to fit on the CDP backbone as the size of the copper molecules is different to each other. Recent studies have shown that α, β and γ CDPs are capable of removing copper ions of all derivatives from water to desirable levels with enhanced sorption capabilities. The adsorption efficiencies of CDPs were surprisingly similar and found to be independent for each type of copper derivatives. The phosphate buffer with pH 6.5-7.5 was found the best alkaline environment to remove the ions. Current technologies have been removed 95% copper ions from solution and unit gram of β-CDP was capable to remove 118 mg of copper ions. These polymers already established as the good carries of organic substances and by combining the inorganic compounds removal, it may be possible to develop a simultaneous system to confiscate both organic and inorganic compounds from waste water. © 2011 WIT Press.

    健康な男子学生6名を対象として,A:騒音なしで高い画面輝度,B:騒音なしで低い画面輝度,C:騒音ありで高い画面輝度,D:騒音ありで低い画面輝度の4条件で60分間のVDT作業を行わせ検討した. 1)疲労自覚症状は,どの条件でも負荷直後の限局した身体違和感で最も高く,負荷直後に自覚的疲労度が増加していた.有意な増加は騒音のある2条件で認められた. 2)VRT-SDとフリッカー値は,どの条件でも負荷終了30分後に最も大きな変動を示し,騒音のある高い画面輝度の条件で有意に増加した. 3)ノルエピネフリンとL/H比も,どの条件においても負荷終了30分後で最も高かった.有意な増加は,ノルエピネフリンでは騒音のない2条件と騒音のある低い画面輝度の条件で認められ,L/H比では騒音のある低い画面輝度の条件で認められた. 4)副交感神経系の指標であるHF成分は,騒音のある高い画面輝度の条件で負荷終了直前に減少した

    本研究では、飛躍的に増加する新たな化学物質の種類や量、有害作用の多様性、生体の感受性などを考慮し、環境汚染化学物質の作用機構として、性ホルモン系に対する作用だけでなく新たな作用機構の解明をも包括できる評価法の確立を念頭に置き、環境汚染化学物質の影響を最も受けやすい胎生期および乳幼児期の中枢神経系を対象として、環境汚染化学物質の中枢神経の分化・組織形成、機能発現に対する影響を包括的に把握でき、かつその作用機構の解明に直結する評価法の確立をはかることを目的とする。本年度は、神経細胞が有機的に結合し、組織形成および機能を発現するためには、.プログラムされた細胞死(アポトーシス)の段階が必要であること、種々の化学物質がアポトーシスを誘導することに着目し、この現象を利用した、神経由来の培養細胞でアポトーシスを人工的に誘導できる細胞系を選択し、神経細胞のアポトーシス誘導に対する環境汚染化学物質の影響をDNA切断を指標として定量的に計測できる評価系を確立するために、DNA切断とその切断部位の修飾による定量的な測定法を確立した。さらに、本定量技術を応用し、人工食品添加物であるアスパルテームの細胞死誘導を検討し、アスパルテームはごく微量でもアポトーシスを誘導することを明らかにした。さらに、ホウ素についても検討したが、ホウ素はアポトーシスを誘導しないことが明らかになった。

    1 新任教員のためのワークショップ型研修の再設計と試行北海道大学ではすでに10年前から先駆的なワークショップ型FDを実施しており,全国的にも良く知られている。新任教員研修のプログラムに,職業規範の学習と新しい教育技術(e-ラーニング,クリッカー)を組み込み実施した。職業規範については論文として発表した(安藤他2011)。また,このタイプの研修に経験のある小笠原,細川が中心になって,次世代の新任教員研修プログラムを検討し,実施した。これらの成果は報告書としてまとめるとともにホームページに掲載した。2 世界の大学におけるティーチング研修の基礎的研究宇田川が中心となって,北米西海岸ベイエリア地区の様々なタイプの大学におけるリサーチ・ティーチングバランスを調査し,さらにこの地域から講師を招いてFDのノウハウの移植をはかった。さらに,最近すさまじい速さで教育改革を進めている中国,インドネシアの大学を訪問し,教育・学習センターの機能と組織を調査した。3 アクティブラーニングのモデル授業の開発本研究では大学初年次の基礎的科学を大学レベルのscience for allと位置づけ,志望する専門分野にかかわらず身に着けるべき基本的な素養と考えた。大学レベルの勉学に必須の項目を絞りこんだ上で,単に知識を覚えこむだけでなく,自分で考え,自分で確かめることにより高度の概念的な理解に到達するアクティブ...

    アメリカのカリフォルニア大学バークレー校のGSIのための「ティーチング・ガイド」と研究代表者が企画編集した「北海道大学TAマニュアル」を比較し,検討したUCバークレー校のGSIセンターから講師を招いて,日本での「教員を目指す大学院生教育(PFF)」を日本の大学院生に対して行う取り組みを行い,どの要素がマッチするか観察した。ソウル大学の「学習と教育のためのセンター」を訪問し,「センター」の視察と意見交換を行った。「センター」のスタッフの数は40名程度で研究代表者の所属するセンターの何倍もあり,ソウル大学の意気込みが感じられた。シンガポール国立大学を訪問した。ライティングセンターなどを視察したが,英語教育などでアジアからの留学生を受け入れることにより,大学が生き延びようとしていることが分かった。UCバークレーを訪問し1週間に渡って,GSIが授業をするところを観察した。どのGSIも熱心で教育スキルが高く,長年のGSI研修の成果が現れていることが分かった。ティーチング・メソッドとしては物理の演習においても学生にグループ討論指せる方法が効果的に使われていた。北海道大学の大学院の授業として,PFFを実施した。日本の大学ではその意義が未だ認知されていないが,数名の受講生が出席し,PFFのいくつかの要素を実際に行ってみることが出来た。

    1.北米の大学におけるTA研修・PFFシステムの調査・研究(1)2009年8月にカリフォルニア州立大学サンフランシスコ校で新任教員研修等について調査を行った。(2)2010年2月にカリフォルニア大学バークレー校(UCB)で,TA研修・PFFプログラムの調査と,2010年3月の北大PFFワークショップの打ち合わせを行った。2.国際ワークショップ・シンポジウムの開催(1)2009年7月に米国,カナダ,韓国,中国の第一線の研究者・実践家を招き,高等教育におけるプロフェッショナル・ディベロップメントについて筑波大学・北海道大学共催の国際ワークショップとシンポジウムを開催した。(2)2010年3月にUCBの講師2人を招き,北海道大学で国際PFFワークショップ(5日間)を開催し,大学院生27名(うち留学生16名,他大学生4名),テューター(TA)7名,他大学の見学者10数名が参加し,ティーチングとライティングの指導を基本とする「北大型」PFFプログラムの基本形ができ上がった。3.北大における外国語教育TA研修・PFFシステムの需要の調査(1)筑波大学でのUCBの講師による国際PFFワークショップ(2009年7月)に,北大から大学院生等約15名を派遣し,UCBのPFFプログラムが北大の大学院生の要望と能力に適合することが確認できた。(2)北大での国際PFFワークショップ(2010年3月)で...

    第一に、微量金属の神経細胞移行時の選択的透過性や細胞内動態および脳内微量金属の代謝制御機構に関わる金属イオン膜輸送蛋白およびメタロシャペロンの役割を明らかにする。これらを基礎として、微量金属が中枢神経伝達機構の構築および脳機能の発現・保持・増進に果たす神経科学的役割を明らかにする。さらに、微量金属が促進因子として関与するとされる老化、神経変性疾患に伴う脳機能の低下の機構解明や予防・治療法の開発を図ることを目的とする。本年度の主な研究内容を示す。老化促進マウスなどの老化モデル動物を用いて、胎児期から老年期に至る脳内微量元素濃度を測定した。その結果、加齢に伴い記憶学習機能と密接な関連を有する亜鉛濃度の有意な低下、モリブデンなど活性酸素発生に関与する金属の増加が認められた。さらに、金属イオンと神経細胞死の関連について、神経系由来のPC12細胞を用いて検討した。その結果、銅単独投与では、細胞死の一つであるネクローシスは今回用いた投与条件ではほとんど認められなかった。その理由として、銅の細胞内吸収が十分に行われないためと思われる。しかし、キレート作用を有する物質を同時に投与すると、ネクローシスだけでなくアポトーシスといわれる細胞死も誘導されることが明らかになった。そのアポトーシス誘導のメカニズムを明らかにするため、アポトーシス誘導関連因子等の変動解析を行い、銅によるアポトーシス誘導は、...

    今年度は実際にソフトウエアのプログラミングを行った。まず、基本的なクリッカーの機能:クイズ問題テキスト・画像・解答番号の読み込み、学生リストの読み込み、クイズ画面表示、投票率のグラフ表示、解答表示、ペン先が解答欄の数字部分をタッチした際の投票、解答データの書き出し、そして、手書きに関する基本的な機能:手書き欄に解答した場合の教員画面への手書き一覧表示、手書き拡大表示、手書き解答数表示、手書き軌跡の書き出し、手書き一覧画面-クイズ画面の切り替え機能を構成した。さらに、本ソフトウエアで重要となる機能・特製の構成は次のように行った。複数の学生からのペンの軌跡をリアルタイムで観覧するためのソフトウエアであるため、表示方法の切り替え等のオプション的な機能に対応するために、逐一再描画することで表示の更新を行うのでは、非常に動作が重くなってしまう。よって、データ書き出しのための軌跡データを保存する一方で、再描画により軌跡の描画を更新するのではなく、画像に絵として書きこんでゆくシャドウビットマップ方式を採用した。模範的な解答例を見せる前に、学生に添削式で指導すると学習効果が得られる。そのような典型的ミスを含む解答を教員側でピックアップできるようにするためには、従来のクリッカーとは異なり、教員側は事前に解答を一望できる管理者画面にする必要がある。しかし、パワーポイントへ本ソフトウエアを組み込む...

    本調査研究においてインドネシア西ジャワ地方を中心に重金属汚染、環境ホルモン汚染の実態を調査し、併せて日本の河川の調査を行ない汚染の構造が日本とは異なっていることを示した。また、インドネシアにある重金属汚染の健康影響の調査を行ない農産物の一部に汚染が広がっていること、その汚染を除去する能力のある細菌の同定、およびインドネシアに多く生息する魚類を用いた重金属汚染バイオマーカーの策定を試み成果を上げた。

    従来の内分泌撹乱化学物質に関する研究は主として性ホルモン作用の生体影響とその作用機序に関して力が注がれ、生体内における他の重要な生理的機能、ことに胎児期において重要な要素である細胞の分化や増殖等にどのような影響を与えるか否かに関する研究はほとんど行なわれていない。本研究では、培養細胞系の実験系を用いて、その培養細胞に内分泌撹乱化学物質を含む環境汚染化学物質を低濃度曝露することにより、細胞の分化や増殖に重要な役割を果たしているアポトーシスにどのような影響を与えるのかを検討し、併せてその作用機序を解明することにより、環境汚染化学物質の発生分化に及ぼす影響評価法を構築しようとするものである。本年度はまず種々の濃度の内分泌撹乱化学物質(ビスフェノールA、ノニルフェノール、トリブチルスズ)をPC12細胞に加え、それらの化学物質自体がアポトーシスを誘起させるか否かを、ラダー法およびTunel法により検討したところ、その化学物質自体ではアポトーシスは誘起されないことが確かめられた。次にPC12細胞の培養培地から牛胎児血清を除いた培地を用いる系および6 Hydroxyl dopamineを培地に添加した系等アポトーシスが誘起される条件下で種々の内分泌撹乱化学物質を低濃度曝露させると、ビスフェノールAでは変化がなかったがトリブチルスズでは誘起させたアポトーシスがほぼ抑制され、ノニルフェノール添...

    本研究ではストレス負荷による神経細胞の形態学的変化ならびに信号伝搬機能の変化を光学計測の手法を用いて観察する計画である。昨年度は,これらの光学手法を実現するために,より取り扱いが容易である海馬スライスの培養法(オーガノティピックカルチャー)を確立した。今年度はこれらの成果をふまえ,ストレスホルモンであるコルチコイドならびにエストロゲン投与後の海馬を対象にした光学計測を実施し,報告書としてまとめた。すなわち,ラットの海馬スライスを顕微鏡のステージ上に設置された潅流槽にセットし,肝油に染色液(DiI)とホルモン(いずれも10μg/μl)を溶解しスライスのCA1上に小球(dye ball)として4時間静置した。エストロゲンは潅流液に溶解することも試みた。その後,共焦点顕微鏡により樹状突起とそれに連なるシナプス(スパイン)を撮影し,樹状突起の単位長さあたりのスパイン数(スパイン密度)を求めた。その結果コントロール条件(3.62±0.54)に比較して,コルチゾールを添加したスライス(2.30±0.32)のスパイン密度が有意に小さくなっていたが(p<0.05),エストロゲンは肝油に含ませた場合も潅流液に含ませた場合も有意な変動は示さなかった。このことから,ストレスにより誘導されるコルチゾールはその濃度が高い場合,わずか4時間でも神経組織にダメージを与えうることが示唆された。今後は,さらに...

    In this project, dynamics of trace elements ( copper, zinc, manganese, selenium ) and the related enzymes such as superoxide dismutasa (SOD), catalase, qlutathione peroxidase (GSH-Px) in metabolism of superoxide radicals were examined in normal healthy subjects and in patients with various diseases.Firstyl, the concentrations of trace elements and activities and/or contents of those enzymes were determined in blood samples from healthy subjects.Secondly, levels of trace elements and superoxide metabolic enzymes were determined in patients with stomach cancer.     In this project the authors designed a computer-controlled apparatus to evaluate fatigue. The design of this apparatus was based on the TAF test, which was originated 20 years ago. Measuring VRT, a subject was instructed to follow a dot target with a cross-shaped marker controlled by a paddle, and to superimpose the marker on the target. The computer calculated the distance between the marker and the target every 0.2 sec, and recorded the data per minute. This process was repeated three times.
