    Orthopaedic implants have been fixed to bone tissue by other artificial materials such as bone cement or clinical screws. It is important to develop the bonding technique between bone and implant materials directly. Laser technique with high energy density is effective tool for bonding the different materials in short duration precisely. In this study, the laser technique for the bonding of bone and implant material has been proposed. The applications of laser technique were examined for the sintered (HAp), glass-added hydroxyapatite (G-HAp) and bone tissue. As a result of laser irradiation on brittle HAp, the cracks appeared near the melting region. However, on G-HAp containing cordierite-glass ceramics with a low linear expansion coefficient, crack-generation was reduced and performance of laser processing improved. The application for the bonding of bone and G-HAp showed that a sufficient bonding was achieved by the generation of melting region of HAp and bone with cavities by the evaporation of bone tissue.

    Naturally occurring holes in bone called foramina are not detected as region of crack initiation or growth. Lumbar vertebrae, which are subjected to complex external loads, have considerable number of such big and small foramina. The mechanical characteristics of bone profoundly depend on the mineral content (HAp crystals) and collagen fibers alignment, where c-crystallographic axis of HAp crystals are preferentially oriented parallel to the long axis of collagen fibers. In the current work, we investigated the crystal orientation and mineral distribution around the foramen using X-rays to explore the reason of foramen not being region of crack initiation or growth to the precise level. Specimens were harvested from bovine lumbar vertebrae. The orientations of HAp crystals were determined from the diffracted intensity profile of X-rays using imaging plate. Mineral fractions were measured from the fraction of X-rays transmitted through the specimen. The tangential orientation of the crystals revealed around the foramen edge was considered as the main reinforcing parameter for the foramen. The minerals were distributed in the specimen without any predictable tendency. Hence, they were not considered as a strengthening parameter. A thicker cortical region was discovered near the surface around the upper edge of the foramen, which was also extended within the cancellous mass as a thin cylindrical cortical shell covering the foramen from inside. Such structures were regarded to result the foramen a damage resistant site.

    Since the cortical bone has higher elastic modulus compared to the cancellous bone, its geometry is very important for stress analysis of bone structure. During finite element modeling of bone structure, cortical bone is generally determined as the region having higher CT values with respect to specific threshold value. However, it is difficult to determine the thin cortical bone regions by considering a specific threshold value. This study proposes a method to select regions of cortical bone from clinical CT images by considering CT value distributions of cortical and cancellous bone. Applying the method to bovine proximal femur, the mean error in cortical thickness compared to the actual bone was found to be less than one pixel (0.39×0.39 mm). Hence, the proposed method could accurately determine the cortical bone regions from clinical CT images. The method was also applied to develop a finite element model with the precise cortical bone structure.

    The aim of the study was to elucidate the difference of elastic modulus and hydroxyapatite (HAp) crystal properties between single trabecula and cortical bone. The study used trabecular specimens obtained from the cancellous bone in proximal bovine femurs. In the experiments, the angle between the longitudinal axis of the trabecular specimen and the longitudinal axis of the femur was measured by CT imaging. To measure the elastic modulus of the trabecular specimens, tensile test was conducted using a material testing machine. The c-axis orientation of HAp crystals and the degree of orientation were measured by X-ray diffraction technique. Further, the HAp crystal strain was also measured under tensile loadings. As a result, elastic modulus had a statistical negative correlation with the angle between the longitudinal axis of the trabecular specimen and the longitudinal axis of the femur. The c-axis of HAp crystals was aligned along the longitudinal axis of the trabecula and the degree of orientation was lower than that in bovine cortical bone Further, the HAp crystal strain was lower than that in bovine cortical bone.

    Osteoarthritis (OA) is the most common degenerative joint disease in the field of orthopaedics. In advanced stages of OA, the patients suffer from severe pain, functional limitations in the affected joints, and restriction of mobility. For detecting early OA and the prevention of progression in OA by novel therapeutic strategies, they are important to understand the functional properties of synovial joint and to establish the precise measurement technique. Pendulum friction test is one of the mechanical evaluations for lubrication properties of joints. The advantages of this method are the natural motion without the constraints of joint and also preservation of the joint capsule, the synovial fluid in it and the tendons and ligaments around the knee. In this study, in addition to the friction properties, the viscous and elastic effects were evaluated. The equation of motion of joint cartilage were proposed including the friction, viscous and elastic effect. The each mechanical parameters were identified by fitting the damping oscillation curve measured by pendulum terster with two laser displacement sensors and an accelerometer.

    Early detection of fracture risk is important as effectiveness of treatment with bone diseases. Over the years, loss of bone mineral or bone mass has been considered the major cause of bone fractures. However, recent studies suggest that collagen also contributes to bone strength by using physiological and artificial aging bone. It was found that fracture risk is related to not just a simple loss of bone mass, but related to significant changes in the biochemical and physical properties of the collagen matrix. Therefore the microscopic interaction between the mineral and organic material may determine the mechanical properties of bone. In this study, bovine cortical bone specimens with different ages were analyzed by Raman spectroscopy. As the results, Raman spectrum of bovine cortical bone with different mechanical properties with aging showed the different Raman spectrum with aging. The effectiveness of Raman spectroscopy was confirmed as a technique for the mechanical evaluation of bone aging.

    Cortical bone has a hierarchical structure. In microstructure, cortical bone is a composite structure of the osteonal and non-osteonal regions and it is regarded as "osteon composites". In nanostructure, cortical bone is a multi-phase nanocomposite material consisting of collagen and hydroxyapatite (HAp). The relationship between the mechanical properties and the hierarchical structure of cortical bone has not been fully investigated and elucidated. This study used strip specimens aligned with the bone axis taken from the cortical bone of a bovine femur. In the experiments, elastic modulus and strength of the specimens were measured by tensile tests. The degree of c-axis orientation of HAp crystals in the specimens was measured by X-ray diffraction method. The area ratio of osteonal region at the cross sections of the specimens was determined. Indentation tests using an AFM probe were conducted at the osteonal and non-osteonal regions in the cross-section. As a result, the elastic modulus and strength were higher in specimens with lower area ratio of the osteonal region. Indentation tests show that the contact stiffness of the osteonal region was lower than that of the non-osteonal region; however, the difference was not so large. The degree of HAp orientation was higher in specimens with lower area ratio of the osteonal region. Furtehr, the elastic modulus in the bone axial direction depended on the degree of c-axis orientation of HAp crystals.

    Bone has a hierarchical structure from macroscopic to microscopic levels and is often regarded as a composite material consisting of apatite crystals and organic matrix in microscopic scale. Microscopic bone quality changes with age. Recent studies suggest that osteoporosis is not just a simple loss of bone mass, but involves significant changes in the biochemical and physical properties of the collagen matrix. Irradiating high intensity X-ray into bone, both of small angle scattering by collagen matrix and wide angle diffraction by HAp crystals occur simultaneously. In this experiment, both structural changes between collagen matrix and HAp crystals in bone are observed by synchrotron X-ray diffraction in combination with micro-compressive device for dynamic image guided failure assessment. Then the difference of behaviors by aging, the aging effects on structural and mechanical properties of bone will be cleared.

    The etiology of idiopathic scoliosis is still unknown. The effects of limitation of spinal mobility in forward flexion on the development of scoliosis was studied in experimental animals. The spinous processes of lumbar vertebrae of young female rats were sutured together to make persistent limitation in the forward flexion of the spine. These findings indicated that the experimentally produced scoliosis was structural.     北海道大学In this study, to propose a novel bone quality evaluation as diagnosis of posteoporosis, the mechanical analysis of nano-composite bone structure of apatite/collagen phases was carried out.At first, the microscopic mechanical behaviors of both mineral particles of apatite and collagen matrix in bone are observed by wide-angle X-ray scattering caused by the lattice structure of apatite crystals and small-angle X-ray scattering caused by the periodic structure of collagen molecules, respectively. In combination with micro-compressive device, both strains of mineral and collagen phases were measured simultanously. Based on these knowledges on bone micro-structure, to evaluate the possibility of Raman spectroscopy as novel bone quality assessment, the mechanical behaviors of mineral and collagen phases in bone tissue were measured by using Raman microscope. And, the effects of mechanical properties and ageing of bone were evaluated.

    北海道大学Bone shows a hierarchical structure from a nano scale of hydroxyapatite crystal and collagen molecular to a macro scale of cortical and cancellous bone. This work proposed a new method of detecting stress of tissue level using the information of HAp crystal deformation. X-ray diffraction (XRD) experimental procedure was developed to measure the characteristics of hierarchical structure. Some structural parameters were introduced to represent these characteristics. Also, the XRD method was applied to detect residual stress in bone, and the stress was observed at various regions in animal extremities.

    大阪大学Bone, major component of skeletal system, is blood vessel-rich tissue, and the bone tissue structure is not only of structural architecture but also of vascular network for bone circulation. In this study, the microstructure of bone is analyzed from the viewpoints of the load bearing structure and of the vascular network of microcirculation. The network structure consisting of Haversian canal Volkmann's canal in cortical bone was analyzed by using radiation synchrotron CT, and the biphasic structural change of canal network was revealed in the cortical bone of growing rats tibiae. It was also found that the rarefaction of the canal network was enhanced by the disuse-mediated load reduction. The X-ray diffraction analysis and Vickers hardness test for trabeculae showed that the macroscopic anisotropy of cancellous bone was coming from the structural anisotropy of trabecular network structure and from the orientation dependent property of trabeculae as well. The material and mechanical properties of cortical bone was examined using nano-indentation test and FTIR microscopy, and much importance of bone quality is demonstrated in cortical bone of mature rats. A technique was established for the direct blood flow measurement of periosteal microcirculation by using the pensile-type CCD video microscope. The material transport in cortical bone through the blood vessel in canal network and the lacuno-canalicular system in bone matrix was modeled in terms of the convective diffusion in tubes and the diffusion in homogenized continuum, and the physiological material concentration was analyzed based on the canal network structure of cortical bone. Simulation model of bone remodeling for physiological window was extended to a unified model involving models of the osteocyte apoptosis for disuse window and the targeted remodeling for overuse windows, and its capability was examined through a couple of simulation studies.

    大阪大学Individual geometry of temporomandibular joint(TMJ) was retrieved from MR images and was used for the finite element modeling of individual patient. The condyle trajectory was determined by referring to its position at different mouth opening positions, and used for the stress analysis of soft tissue components of temporomandibular joints. The typical stress conditions in the TMJ disc and the retrodiscal connective tissue were investigated for normal TMJs and for anterior-disc-displaced TMJs. The sensitivity analysis with condyle position demonstrated that the perturbation in inferior direction at the fully-mouth-open position and the perturbation in inferior direction with small rotation at the mouth-closed position were effective for the disc reduction during the mouth opening motion of anterior-disc-displaced TMJs. The individual finite element model was extended with the frame structures for the mandible bone connected to the finite elements of condyle and for the temporal bone behind the elements of glenoid fossa. These frames were interconnected by muscles of mastication under the contacting conditions at anterior/posterior tooth, and the extended model enabled us to examine the stress condition of soft tissue components of viscoelasticity under the clenching. The parameter studies suggested us that the comparison of the results of individual finite element analysis with the results of medical imaging analysis has the potential to estimate the elastic characteristics of the soft tissue components towards the diagnosis and to probe the treatment condition of the adjustment of relative position of bony components leading the condyle trajectory for the reasonable stress condition of the soft tissue components.

The methodology for establishing patient-specific individual modeling was developed for temporomandibular joints based on magnetic resonance images available at medical examination, and a prototype modeling system has been demonstrated for the clinical dental researchers.2. The jaw bone movement was reconstructed interpolating the jaw bone position observed by magnetic resonance images at different mouth opening stages of individual subject. It gives us a rationale to determine the typical jaw movement condition used for the computational biomechanics analysis of temporomandibular joints.3. Stress analysis concerning on the soft tissue components of articular disc and surrounding connective tissue have showed that normal temporomandibular joints exhibited the high stress region at the anteromedial portion of the articular disc and that permanently anterior-disc-displaced temporomandibular joint resulted the relatively high stress in retrodiscal connective tissue in which tissue ligamentization was observed.4. Stress redistribution and tentative displacement in articular disc was very dependent not only on irregular spatial arrangement of components of anterior-disc-displaced temporomandibular joint but also on biomechanical property of articular disc and retrodiscal connective tissue. This fact suggested us the importance of biomechanical tests of retorodiscal connective tissue as well as articular disc.5. Indiyidual computational biomechanics analysis revealed that the tentative articular disc displacement in anterior direction due to the mouth opening was dependent on the elastic modulus of the articular disc in the case of normal temporomandibular joints, while the disc displacement was dependent on the elastic modulus of the retrodiscal tissue in the case of permanently anterior-disc-displaced joints. This feet showed the possibility to estimate the mechanical property of soft tissue components by comparing tentative disc displacement by image analysis and that by computational biomechanics analysis.

    大阪大学An orthogonal lattice continuum is studied as a model of trabecular architecture of cancellous bone and the relation between parameters of lattice geometry and mechanical characteristics as a continuum is considered in the context of unit cell analysis. A rate equation of orthogonal lattice parameters is described for a phenomenological model of adaptation by bone remodeling by paying attention to some optimality in mechanical function at equilibrium. The lattice continuum model is extended to permit the non-orthogonal lattice intersection, and the lattice parameter determination is considered. The residual stress in trabecular architecture is observed and estimated based on a lattice model.As an application of cancellous bone-like internal structure to a structural design problem, a topology and shape determination problem is formulated for a variable stiffness structure that realize different stiffness at different level of external load, and a solution procedure is demonstrated. This problem is extended for a design of a material structural system with specified elastic material constants depending on working load level.
