    Although the original Saccharina japonica populations in China are assumed to have been accidentally introduced from Japan in 1920s, few investigations have been performed to prove it. Genetic structure analysis is expected to shed light on this question. In the present study, 11 highly polymorphic microsatellite markers were employed to analyze the genetic structure of representative populations of S. japonica in the Northwest Pacific, including the four varieties from Japan. Populations from China had a much lower level of genetic diversity than the ones from Japan in terms of the number of alleles, private alleles and locally common alleles, and the values of heterozygosity, suggestive of the founder effect in the introduced populations. Both the Bayesian model-based and genetic distance-based analyses revealed three major clusters: (1) S. japonica var. japonica, var. religiosa, and var. ochotensis from Hokkaido, as well as the populations from Korea and Russia; (2) populations from China; and (3) var. diabolica. Saccharina japonica var. diabolica harbored the highest number of private alleles, with three of them having a frequency of 1.00 at three loci, which have the potential to be used as specific markers to distinguish this variety from others. Significant genetic differentiation was found between almost all the populations except for a few cultivated populations from China. Populations from China were most closely related to S. japonica var. japonica, which was in turn most close to var. religiosa and the Korean population. The microsatellite markers used in this study were shown to be polymorphic enough to decipher the subtle genetic structure of S. japonica, including the different varieties.

    Acoustic methods can be used to assess seaweed meadows. The accuracy of the methods is a key factor in the estimation of seaweed distribution and conditions. We obtained and verified thickness and spatial distribution values measured using an echo sounder. We determined the thickness of seaweed growing in the coastal waters off Higashidoori-mura, Aomori, Japan on June 5-6, 2013. Acoustic data were collected using an on-board quantitative echosounder at 120 kHz. The thickness was also directly measured at 14 points. The root mean square error (RMSE) of the thickness determined by the acoustic and direct methods was calculated. A survey to determine the spatial distribution was performed in Miyako-shi, Iwate on July 18, 2014. The estimated spatial distribution was determined from the thickness data collected by the acoustic method of varied transect lines by changing transect orientation and intervals. The kelp forest distribution was also observed directly at 106 points. Then, the concordance rates of the visual observations and estimated spatial distribution were obtained. The RMSE of the acoustic and directly measured thickness of the kelp forests (Saccharina japonica), was 0.06 m, similar to the vertical resolution of the echosounder. The concordance rate between the acoustic and directly measured values was 92% when the maximum transect interval was 21 m. Smaller transect intervals yielded higher accuracy. High accuracy for the thickness and acoustically derived spatial distribution were obtained by the acoustic method when suitable thresholds were used, which has important applications for the evaluation of seaweed stands.

    Morphological, molecular and chromosomal studies were carried out on Tauya basicrassa, an endemic kelp species distributed on the northern continental coast of the Sea of Okhotsk in Russia. The sporophytes of T basicrassa grow up to 3-6 m long, 1.8-2.2 m wide, and 6.5-7 kg wet weight. The thallus has a blade with very thick narrow basal portion and thinner and much broader upper portion, which usually splits into 3 bullated lobes. A dwarf laminariacean alga, which did not show any morphological similarity to the other species of the order Laminariales, was found from the same locality. The blade of this alga is thin and soft, reached 26-34 cm long and 6-6.5 cm wide and had 4 longitudinal rows of bullations that covered the entire blade. Molecular analysis showed that the dwarf alga has 100% sequence identity in plastid-encoded RuBisCo spacer, mitochondrial cytochrome c oxidase subunit 1 and nuclear-encoded rDNA genes with normal sporophytes of T basicrassa, indicating that they are different life forms of the same species. Fluorescent DAPI staining showed that the nucleus in the normal sporophyte was 50-65% larger than those of the dwarf ones. Chromosome count using acetocarmine staining showed n = ca. 20 for the normal sporophytes of T basicrassa and n = ca. 10 for the dwarf one. These results suggest that the dwarf thallus is a haploid parthenosporophyte of T. basicrassa, which developed in nature. This is the first evidence of parthenosporophytes of the laminariacean algae occurring naturally in the field.

    Saccharina japonica is a commercially and ecologically important seaweed and is an excellent system for understanding the effects of domestication on marine crops. In this study, we used 19 selected simple sequence repeat (SSR) markers to investigate the influence of domestication on the genetic diversity and structure of S. japonica populations. Wild kelp populations exhibited higher genetic diversity than cultivated populations based on total N-A, H-E, H-O, N-P and A(R). Discriminant analysis of principal components (DAPC), a neighbour-joining (NJ) tree and STRUCTURE analyses indicated that S. japonica populations could be divided into two groups (a cultivated/introduced group and a wild indigenous group) with significant genetic differentiation (P < 0.0001). Divergent selection, continuous inbreeding and inter-specific hybridization have caused the divergence of these two genetically separate gene pools. The significant genetic differentiation between northern and southern cultivated populations appears to be due to inter-specific hybridization and wild germplasm introduction during the domestication process. In addition, the cultivation of S. japonica has not resulted in any serious genetic disturbance of wild introduced S. japonica populations. An understanding of the genetic diversity and genetic structure of domesticated S. japonica will be necessary for further genetic improvement and effective use of germplasm.

    The genetic diversity of Saccharina japonica inhabiting the Hokkaido coastline and the surrounding area was investigated by AFLP and SSR analyses. A STRUCTURE analysis based on the AFLP data identified two clusters in the species, although a few local populations with genetic structures different from that of the neighboring populations were detected. The two clusters could be identified even within each current variety. In the SSR analysis, genetic variation in nucleotide sequences in all samples was located in four DNA regions (MS10, MS11, MS16, and MS29), and it is thought that these regions can be used as markers to detect individual-level variation in S. japonica. The STRUCTURE analysis of the SSR data identified four clusters in the species. Since the geographical distribution of these four different clusters does not correspond exactly to the currently recognized varieties in this species, it will be necessary to re-evaluate the distribution of the varieties of S. japonica.

    The taxonomy of Saccharina longipedalis, a kelp species endemic to the brackish Lake Akkeshi in eastern Hokkaido, was reviewed. The external morphology of sporophytes collected from seven sites around the distribution boundary of S. longipedalis and Saccharina japonica var. diabolica were compared; it was confirmed that the diagnostic morphology of S. longipedalis, such as a long stipe and thin blade, was not solely characteristic of individuals that grew in and around the center of the lake, which is thought to be the unique distribution area of S. longipedalis. Sequence comparisons of a 5S rDNA spacer and four microsatellite regions and AFLP analysis revealed that genetic similarity was high between S. longipedalis and S. japonica var. diabolica. Also based on observations that the early development in hybrids of sporelings from the seven sites was normal, we conclude that S. longipedalis should be treated as S. japonica var. diabolica.

    The DNA fragment analysis can become an effective tool to study genetic differences between not only species but also individuals on saccharinan kelp from which the little genetic diversity was reported. This time, extraction methods of suitable DNA for use in the analysis with a capillary sequencer was examined on Saccharina japonica var. diabolica that contains polysaccharide abundantly. When AFLP was performed using genomic DNA extracted by seven different methods: (1) commercial kit, (2) original CTAB method, (3)-(5) three types of modified CTAB method, (6) modified SDS method, (7) combination of CTAB method and SDS method, high reproducible peak that was worth for the analysis was noticeable in the electropherogram in the experiment with the last combination method (7). It is considered that the pretreatment washing of polysaccharide and the subsequent purification for protein and RNA in SDS method and for polysaccharide in CTAB method are effective to obtain the high purity DNA.

    The inhibition of germination and development of zoospores originating from Laminaria angustata originated on various kinds of hydrogels is studied. The effects of the water content of the gel (the degree of swelling), the electrical nature (neutral, positive charge, negative charge), the charge density, the counterions of hydrogels on the inhibition of zoospore germination and development of gametophytes are phenomenologically investigated. Among the gels investigated, poly(acrylic acid) (PAA) gel showed a dramatic inhibition to germination, and might serve as a novel, environmentally safe inhibitor of zoospore adhesion.
