    AIM: To evaluate the dental pulp response to a novel mineral trioxide aggregate containing phosphorylated pullulan (MTAPPL) in rats after direct pulp capping. METHODS: Ninety-six cavities were prepared in the maxillary first molars of 56 male Wistar rats. The dental pulps were intentionally exposed and randomly divided into four groups according to the application of pulp capping materials: MTAPPL; phosphorylated pullulan (PPL); a conventional MTA (Nex-Cem MTA, NCMTA; positive control); and Super-Bond (SB; negative control). All cavities were restored with SB and observed for pulpal responses at 1-, 3-, 7- and 28-day intervals using a histological scoring system. Statistical analysis was performed using Kruskal-Wallis and Mann-Whitney U-test with Bonferroni's correction, and the level of significance was set at 0.05. DMP1 and CD34 antigen were used to evaluate odontoblast differentiation and pulpal vascularization, respectively. RESULTS: On day 1, mild inflammatory cells were present in MTAPPL and NCMTA groups; fewer inflammatory cells were present in the PPL, whereas SB was associated with a mild-to-moderate inflammatory response. A significant difference was observed between PPL and SB (p < .05). No mineralized tissue deposition was observed. On day 3, moderate-to-severe inflammatory cells were present in PPL and SB, whereas MTAPPL and NCMTA had a mild inflammatory response. Initial mineralized tissue deposition was observed in the NCMTA, MTAPPL and SB. A significant difference was observed between MTAPPL and PPL (p < .05). On day 7, a thin layer of mineralized tissue was observed in all tested groups with no or mild inflammatory response. On day 28, no inflammatory response was observed in MTAPPL, whereas NCMTA, PPL and SB had mild inflammatory responses. A significant difference was observed between MTAPPL and SB (p < .05). Complete mineralized tissue barrier formation was observed in MTAPPL, NCMTA and PPL with no significant difference (p > .05). SB exhibited incomplete mineralized tissue barriers, significantly different from NCMTA, MTAPPL and PPL (p < .05). The staining with CD34 was positive in all the groups on all observation days. CONCLUSION: The favourable pulpal responses and induction of mineralized tissue formation associated with MTAPPL indicate its potential application as a direct pulp capping material.

    This study aimed to evaluate the pulpal responses of monkey's pulp after direct pulp capping (DPC) with the novel mineral trioxide aggregate containing phosphorylated pullulan-based material (MTAPPL). Seventy-two teeth were randomly divided into four groups: MTAPPL; Nex-Cem MTA (NX); TheraCal LC (TH); and Dycal (DY). Histopathological changes in the pulps were observed at days 3, 7 and 70. On day 3, mild inflammatory responses were observed in the MTAPPL, no to moderate inflammatory responses in the TH, whereas moderate inflammatory responses in the NX and DY. No mineralized tissue formation (MTF) was observed in all groups. On day 7, no or mild inflammatory responses were observed in all groups. Initial MTF was observed except for DY. No inflammation with complete MTF including presence of odontoblast-like cells was observed in the MTAPPL, NX and TH groups at day 70. These findings indicate that MTAPPL could be an efficient DPC material.

    目的:多機能性表面処理ガラス(Surface Pre-Reacted Glass ionomer:S-PRG)フィラーは、イオン放出性を有することからさまざまな生体活性効果を示すことが報告されている。本研究の目的は、S-PRGフィラーと酸化亜鉛を基材としたS-PRGフィラー含有根管充填用シーラー(S-PRGS)の生体親和性を、ユージノール系シーラー(ZOES)および非ユージノール系シーラー(ZOS)と比較検討することである。材料と方法:S-PRGS、ZOESおよびZOSの硬化体へMC3T3-E1細胞を播種培養し、走査電子顕微鏡(SEM)にて観察、細胞生存性を測定した。次にラットの背部皮下に硬化させた各シーラーを埋植し、術後10日および35日の炎症性細胞浸潤の程度について光学顕微鏡で観察を行い、数値化して評価した。結果:SEM観察の結果、S-PRGS上には細胞の伸展がみられたが、ZOESおよびZOS上では細胞の伸展は観察されなかった。細胞生存性を測定した結果、S-PRGSはZOESおよびZOSと比較して有意に高い値を示した。皮下埋入試験の結果、術後10日ではすべてのシーラー周囲に炎症細胞浸潤が観察されたが、S-PRGSでは軽度であった。S-PRGS、ZOESおよびZOSの術後10日の炎症スコアは、それぞれ2.16、2.67および2.83であった。術後35日では、S-PRGS周囲に炎症性細胞はほとんど観察されなかった。術後35日の炎症スコアは、それぞれ0.00、0.67および0.67であった。結論:S-PRGSは、ZOESおよびZOSと比較して良好な生体親和性を示した。(著者抄録)

    In this study, we investigated the behaviors and cytocompatibility response of human cervical carcinoma (HeLa) cells expose to nano-sized particles. Cultivated cells exposed to titanium oxide and indium oxide nanoparticles remained highly viable. In the presence of copper oxide (CuO); however, the cells became seriously inflamed. To understand the mechanism by which CuO causes cell death, we evaluated cell death and apoptosis cytometry. CuO induced cells apoptosis more strongly than exposure to titania nanoparticles. Confocal fluorescence microscopy revealed that the nano-sized particles penetrate the cells.
  • 1ステップ型接着システムによる臨床的スミヤー層の除去能とその接着性能

    本研究の目的は、6種の1ステップ型接着システムを用い、研削条件の異なる象牙質スミヤー層の除去能を検討することと、その微小引張り接着強さとの関係性を明らかにすることである。ヒト抜去大臼歯の歯冠象牙質をレギュラーダイヤモンドポイントあるいはSiCペーパー#2000により研削を行い、2種の象牙質研削面を得た(レギュラー面、#2000面)後、G-Bond Plus(GP)、Absolute 2(AB)、Bond Force(BF)、BeautiBond(BB)、Hybrid Coat II(HC)、およびClearfil S3 Bond(TS)で処理し、処理面の走査型電子顕微鏡観察、コンポジットレジン築盛後の微小引張り接着試験、および接着界面の透過型電子顕微鏡観察を行った。その結果、ABはスミヤー層を溶解し、強い脱灰能を示したが、その他のシステムではスミヤー層の残留が認められた。各システムの接着強さは、GPはレギュラー面に対する方が有意に高かったが、AB、TSは#2000面に対する方が有意に高かった。BF、BB、HCは両者間で有意差がなかった。以上より、スミヤー層の除去能と短期的な接着強さの関係は使用するシステムにより異なることが示唆された。(著者抄録)

    Purpose: To evaluate the hydrolytic stability of three one-step self-etching adhesives (1-SEAs) bonded to dentin through bond strength testing and ultra-morphological interfacial analysis before and after long-term thermocycling. Materials and Methods: Eighteen flattened mid-coronal dentin surfaces of extracted human molars were subjected to bonding treatment using three 1-SEAs (Clearfil S(3) Bond, Kuraray (S3), G-Bond, GC (GB), Absolute, Dentsply-Sankin (AB)), after which the bonded surfaces were built up with composite. After storage overnight at 37 C, the specimens were sectioned into slabs and further trimmed into an hourglass shape with an interface area of approximately 1 mm(2). The specimens were left untouched (control) or were thermocycled for 100,000 cycles. The microtensile bond strength (mu TBS) was measured and the ultrastructure of the adhesive/dentin interface characterized using transmission electron microscopy (TEM). Results: Long-term thermocycling significantly decreased the mu TBS of all one-step adhesives tested (p < 0.05, one-way ANOVA and Games-Howell test). TEM revealed a similar interfacial ultrastructure before and after thermocycling for S3. For GB, many voids were observed at the interface after 100,000 thermocycles. Regarding AB, collagen fibrils could no longer be clearly observed upon staining, while the adjacent unaffected dentin was rich in voids. Conclusion: The bond strength and ultramorphological data demonstrated that the bond of 1-SEAs to dentin degrades with time, although the degree of degradation is obviously material dependent.

    トータルエッチング(エッチ&リンス)、セルフエッチングといったレジン系歯質接着システムやグラスアイオノマーセメントは、歯の再建に不可欠な材料として、今日の歯科医療現場で幅広く用いられている。この歯質接着、特に象牙質接着は、樹脂含浸層を形成することによる機械的な嵌合で接着すると考えられてきた。それに基づき、歯質接着の評価も形態学的な観察、接着強さの測定、辺縁封鎖性を中心に検討されてきた。しかし、異なるメカニズムで歯質と接着するこれらの材料も、その成否は材料と歯質との界面に大きく左右される。すなわち、修復材料の寿命は、樹脂含浸層の質に大きく依存する。より機能性に優れた接着を得るためには、アパタイトやコラーゲンなどさまざまな物質より構成される歯質に対し、接着性モノマーをより強く接着させ、かつ、ナノスケール(あるいは化学結合のレベル)で両者の相互作用を制御する必要がある。このような観点から本稿では、歯質と材料との界面の形成メカニズムの解析について化学分析を中心に解説した。(著者抄録)

    The purpose of this study was to evaluate the morphological appearance of resin-dentin interface produced by 1-step adhesive systems in order to classify them, using TEM. It was hypothesized that all interfaces produced by each 1-step adhesive show characteristics similar to that produced by the conventional self-etching system rather than that produced by etch-and-rinse system. The flattened dentin surfaces were subjected to bonding treatment using either a 2-step adhesive: single bond, mega bond, unifil bond or a 1-step adhesive: Prompt L-Pop, absolute, xeno CF, AQ bond, one-up bond F, G bond, and SSB-200. A flowable resin was then applied to these surfaces. The results revealed that resin-dentin interfaces produced by 1-step systems were classified into three categories. Additionally, the interface produced by G bond and SSB-200 showed a different morphological appearance from that of the others. The interface produced by these adhesives was thinner and less demineralized, with the presence of hydroxyapatite-coated collagen fibrils. It is proposed that "Nanointeraction Zone (NIZ)" is a new complementary classification for partially demineralized resin-dentin interdiffusion zones. Thus, one-step self-etching adhesive systems can be classified into three categories, etch-and-rinse type, mild self-etching type, and NIZ type, by their morphological appearance of the interface. © 2006 Wiley Periodicals, Inc.

    The micro-shear bond strengths (MSBSs) of five single-step self-etch adhesives (Adper Prompt L-Pop [APL], AQ Bond plus [AQP], OBF-2 [OB2], Reactmer Bond [RB], and Xeno III [XIII]) were compared with that of a two-step self-etch adhesive, Clearfil SE Bond [SE]. The adhesives were applied on dentin surfaces, according to manufacturers' instructions, for bonding of resin composite to dentin. After 24 hours, a micro-shear bond test was carried out and the data were analyzed by one-way ANOVA and Bonferroni test (p<0.05). The mean MSBSs in MPa were APL: 22.8, AQP: 37.4, OB2: 34.7, RB: 28.3, XIII: 30.3, and SE: 47.2. Among the single-step adhesives, the MSBS of AQP and OB2 were significantly higher than APL (p<0.05). In conclusion, the single-step adhesives used in this study, apart from AQP, produced significantly lower MSBS than the two-step adhesive (p<0.05).

    Functional monomers chemically interact with hydroxyapatite that remains within submicron hybrid layers produced by mild self- etch adhesives. The functional monomer 10- MDP interacts most intensively with hydroxyapatite, and its calcium salt appeared most hydrolytically stable, as compared with 4- MET and phenyl- P. We investigated the hypothesis that additional chemical interaction of self- etch adhesives improves bond stability. The micro- tensile bond strength (mu TBS) of the 10- MDP- based adhesive did not decrease significantly after 100,000 cycles, but did after 50,000 and 30,000 cycles, respectively, for the 4- MET- based and the phenyl-P- based adhesives. Likewise, the interfacial ultrastructure was unchanged after 100,000 thermocycles for the 10- MDP- based adhesive, while that of both the 4- MET- and phenyl- P- based adhesives contained voids and less- defined collagen. The findings of this study support the concept that long- term durability of adhesive-dentin bonds depends on the chemical bonding potential of the functional monomer.

    Objective. The purpose of the present study was to evaluate the effect of evaporation of primer components on the ultimate tensile strength (UTS) of primer-adhesive mixtures. Methods. The UTSs of 1:3 by weight primer-adhesive mixtures and of the pure adhesives (controls) were measured after 0-, 2-, 5-min and 24-h storage in a 37 degrees C oven for the three-step etchftrinse adhesive OptiBond FL (Kerr) and the two-step self-etch adhesive Ctearfil SE (Kuraray). Results. Evaporation of primer components increased with time for both adhesives investigated. OptiBond FL showed a higher evaporation of primer components than Ctearfil SE at the 2-, 5-min and 24-h storage time. As compared to the pure adhesive, the UTS of the primer-adhesive mixture was lower after 0- and 2-min storage for OptiBond FL and after 0-, 2- and 5-min storage for Clearfil SE. The UTS of OptiBond FL was higher than that of Clearfil SE at the 0- and 2-min storage time, though there was no difference at the 5-min and 24-h storage time. Significance. The degree of evaporation of primer components depended largely on the primer solvents employed. The significant decrease in UTS of adhesive resin when mixed with primer must be attributed to incomplete evaporation of primer components.     Objectives: To evaluate the effect of double-application of all-in-one adhesives using the micro-shear bond test. Methods: The occlusal surfaces of extracted human third molars were ground perpendicular to the long axis of each tooth to expose a flat dentin surface. Three commercially available and one experimental bonding system were used in this study: Adper Prompt L-Pop (APL, 3M ESPE), REACTMER BOND (RB, Shofu), XENO III (Xeno, Dentsply-Sankin) and newly developed OBF-2 (0132, Tokuyama Dental). These adhesives were applied on the dentin surfaces by either the manufacturers' instructions or by an experimental method (single-application or double-application). Resin composite was then placed and light-cured for 40 s. After 24 h immersion in water, a micro-shear bond test was carried out and the fractured dentin surfaces were observed microscopically. The data were analyzed by one-way ANOVA and Tukey's HSD tests (p < 0.05). Results: The mean bond strengths of APL, RB, Xeno and OB2 with single-application were 22.7, 28.3, 30.3 and 34.6 MPa, respectively. The mean bond strengths of APL, RB, Xeno and OB2 with double-application were 29.5, 27.2, 29.6 and 32.5 MPa, respectively. There were no statistically significant differences in micro-shear bond strengths between the single- and double-application methods for each adhesive system (p > 0.05). The morphological observation of the fractured dentin surfaces revealed differences between the single- and double-application groups especially for APL and OB2. Conclusions: Micro-shear bond strengths of all-in-one adhesives in this study showed no statistically significant differences between the single-application and the double-application method.     Resin-dentin bonds are known to degrade in the relatively aggressive oral environment. In order to obtain greater insight into the interfacial degradation process, we examined, by using transmission electron microscopy (TEM), the interfacial ultrastructure of two adhesives bonded to dentin after 1 yr in vivo. Class V cavities were prepared on the buccal surfaces of 14 intact teeth of two monkeys and then restored by using either the two-step self-etch adhesive, Unifil Bond, or the two-step etch-and-rinse adhesive, Single Bond, in combination with the restorative microhybrid composite, Z250. After 1 yr, 10 other teeth were restored by using the same materials (controls). One day later, the monkeys were killed, following which the microtensile bond strength to dentin was determined and the interfacial ultrastructure was examined by TEM. Whereas no noticeable changes in the morphology of the resin-dentin interface were observed between the 1-d and 1-yr specimens for Unifil Bond, Single Bond exhibited signs of interfacial degradation, in particular at the bottom of the 3 mu m-deep hybrid layer. In conclusion, the adhesive interface produced by the etch-and-rinse adhesive was less resistant to degradation than that produced by the self-etch system.

    The aim of this study was to investigate the flexural fatigue behavior of 11 packable composites. Of each material 30 rectangular samples (1.2 x 5 x 35 mm) for flexural fatigue test were prepared. The clamped fracture strength and flexural fatigue limit (FFL) of each material were determined using a custom-made fatigue machine, after storage of the samples for one month in water at 37degreesC. All data were analyzed using one-way ANOVA and Bonferroni/Dunn's test for multiple comparisons (p < 0.05). Regression analysis was used to evaluate the relationship between elastic modulus (Abe et al., 2001), clamped fracture strength or FFL and inorganic filler fraction (vol%). The fracture strengths of all but two materials were in between those of the compact-filled densified composites and the microfine ones. The FFL of the packable composites tested were significantly lower than those of the compact-filled densified composites. Three of the tested materials showed even significantly lower FFL than did the microfine composites. There were statistically significant relationships between both the elastic modulus and the volumetric filler fraction (R-2 = 0.974, p = 1.990 x 10(-7)). The great diversity in packable composites makes clinical assessment necessary with regard to a justified use in posterior teeth. (C) 2004 Elsevier Ltd. All rights reserved.

    Objective. The purpose of this study was to measure the micro-tensile bond strength (JBS) of two adhesives to dentin and to correlate this JBS with the micro-tensile fracture strength (mu TFS) of primer-adhesive mixtures of the two adhesives. Methods. The three-step etchEtrinse adhesive OptiBond FL (Kerr) and the two-step self-etch adhesive Clearfil SE Bond (Kuraray) were used to bond Filtek Z100 (3M ESPE) to twenty mid-coronal dentin surfaces. The mu TBS was determined at 1 and 24 h after tight-curing. In addition, the mu TFS of primer-adhesive mixtures (primer: adhesive= 1:3 by weight) and of the pure adhesives (controls) were measured after the same time intervals. Results. The JBS to dentin of OptiBond FL at 24 h was significantly higher than at 1 h and than the mu TBS of Clearfil SE Bond at 24 h. The mean mu TFS of the primeradhesive mixtures was always lower than the mu TFS of the respective adhesives. The highest reduction in mu TFS due to mixing primer with the adhesive resin was measured for Clearfil. SE Bond. Both the mu TFS of the primer-adhesive mixtures and the pure adhesives of OptiBond FL and Clearfil SE Bond increased with time. Significance. The early 1 h mu TBS to dentin was lower than the 24 h mu TFS for OptiBond FL, but not for Clearfil SE Bond. Differences in polymerization efficiency as well as in the resulting interfacial strength may explain such varying early bond strengths to dentin. (c) 2004 Academy of Dental Materials.     This study investigated, mechanically and morphologically, whether the dentin surface irradiated by CO2 laser could be a possible adherent when bonded with simplified-step adhesives. Buccal enamel and cementum of extracted human premolars were removed to expose a flat dentin surface. The dentin surfaces were irradiated continuously with CO2 laser at 1.0 W. Before bonding with either a single-bottle adhesive (Single Bond) or a self-etching priming system (Mega Bond), the irradiated dentin surface was treated as follows: no treatment, NaHCO3 powder abrasion and wet-grinding with 600-grit SiC paper. The treated dentin surfaces were bonded to resin composite with either of the two adhesives. Non-irradiated dentin surfaces were also used as control. Resin bonded specimens were stored in water at 37 degrees C for 24 hours and subjected to microtensile bond test. Additionally, to observe the resin/irradiated dentin interface, resin-bonded specimens were similarly prepared, sectioned into slabs, embedded in epoxy resin, polished with diamond pastes, sputter coated Au-Pd and examined with scanning electron microscopy (SEM). After SEM observation, the specimens were further polished with diamond paste to remove the Au-Pd sputter-coat, immersed in HCL and NaOCl and finally observed by SEM again. In the presence of carbonized dentin, microtensile bond strength drastically decreased but recovered to the control value by removing the carbonized dentin layer visually with SiC paper for both adhesive systems. However, the laser-affected dentin that remained on the bonded interface was easily dissolved with NaOCl and HCl.

    Objectives: The purpose of this study was to determine the effect polymerization contraction stress may have on bond durability. Methods: Bonding effectiveness was assessed by micro-tensile bond strength testing (muTBS) and electron microscopy. The muTBS to flat dentin surfaces and in standardized cavities was determined (this after 1 day as well as 1 year water storage). Six adhesives representing all current classes were applied: two etch-and-rinse (OptiBond FL, Kerr; Scotchbond 1, 3M ESPE), two self-etch (Clearfil SE Bond, Kuraray; Adper Prompt, 3M, ESPE) and two glass-ionomer (Fuji Bond LC, GC; Reactmer, Shofu) adhesives. Results: The conventional 3-step etch-and-rinse adhesive OptiBond FL bonded most effectively to dentin, and appeared insensitive to polymerization shrinkage stress and water degradation. The 2-step self-etch adhesive Clearfit SE Bond most closely approached this superior bonding effectiveness and only slightly lost bond strength after 1-year water exposure. The 2-step etch-and-rinse adhesive Scotchbond 1 and the 'strong' 1-step self-etch adhesive Adper Prompt appeared very sensitive to cavity configuration and water-aging effects. The 2-step resin-modified glass-ionomer adhesive Fuji Bond LC only suffered from shrinkage stress, but not from 1-year water-exposure. Remarkable also is the apparent repairability of the 'mild' 1-step glass-ionomer adhesive Reactmer when stored for 1 year in water, in spite of the very tow 1-day muTBS.     The effect of a polyalkenoic acid conditioner pretreatment on the bond strength of a glass-ionomer adhesive to tooth substrates with or without smear layer was evaluated. Smear-layer covered and smear-layer free dentin and enamel surfaces were prepared from 24 extracted human molars. Resin composite was bonded to the sur faces using FuJiBond LC with or without a polyalkenoic acid conditioner and subjected to microtensile bond strength (muTBS) testing. Failure modes were determined using scanning electron microscopy. For dentin, smear-layer coverage and conditioner treatment did not reveal significant differences in muTBS, which ranged from 20 to 29 MPa. For enamel, smear-layer coverage did not significantly affect muTBS, whereas, the use of conditioner significantly improved MTBS, reaching the same MTBS-values as when bonded to dentin. Regarding failure mode, most dentin specimens failed mixed adhesive cohesively. For enamel, adhesive failures mostly occurred when no conditioner was used, though mixed failures were predominant when the specimens were conditioned beforehand. Bonding of the glass ionomer adhesive to dentin can be achieved without the separate use of a polyalkenoic acid conditioner, even with the interposition of a smear layer. However, instrumented and non-instrumented enamel requires separate conditioning to provide sufficient micro-mechanical retention.

    Background: Self-etching primers are reported to produce considerable etching on flat enamel surfaces. However, little is known about the influence of different enamel surface textures. In this study the influence of grinding enamel on bond strength of two all-in-one and two-step adhesives was investigated. Methods: Resin composite was bonded to the ground enamel of extracted human third molars that was reduced 0.5mm from the buccal or lingual surfaces using either regular or superfine-grit diamond burs with each of the three adhesives. After 24 hours in 37degreesC water, the specimens were sectioned into slabs of 0.7mm thickness, trimmed to an hour glass configuration, and subjected to micro-tensile bond strength (MTBS) testing. Results: For all adhesive systems, MTBS to enamel ground with a regular-grit diamond bur was not significantly different from that with a superfine grit diamond bur. The etching patterns of there adhesives were partly varied according to the aggressiveness of the adhesives. Conclusions: The use of two different burs does not affect the tensile bond strength of the adhesives to enamel.

    The purpose of this study was to evaluate the long-term durability of in vivo bond strengths and the morphological changes of interfaces between dentin and two adhesive systems. Class V preparations were prepared on the facial surfaces of 14 intact teeth of two monkeys and restored with a combination of Unifil Bond/Z250 or Single Bond/Z250. One year later, 10 additional teeth were restored with the same materials and the monkeys were killed after 24 h. All of the restored teeth were subjected to microtensile bond strength (muTBS) testing. The debonded surfaces of the dentin sides were morphologically observed using Fe-scanning electron microscopy (SEM), as were the polished cross-sections of resin-lentin interfaces. For both Unifil Bond and Single Bond, the muTBS at 24 h was significantly higher than that at 1 yr. Fe-SEM observations of polished cross-sectioned and fractured surfaces showed that porosity within the hybrid layers produced by Single Bond increased over time. However, the interface produced by Unifil Bond revealed no noticeable changes in morphology between 24-h and 1-yr specimens. It is concluded that even though the bond strengths of both adhesive systems declined over time, the bonding interface using self-etching primers was relatively stable over time compared to the wet bonding system.

    Mild self-etch adhesives demineralize dentin only partially, leaving hydroxyapatite around collagen within a submicron hybrid layer. We hypothesized that this residual hydroxyapatite may serve as a receptor for chemical interaction with the functional monomer and, subsequently, contribute to adhesive performance in addition to micro-mechanical hybridization. We therefore chemically characterized the adhesive interaction of 3 functional monomers with synthetic hydroxyapatite, using x-ray photoelectron spectroscopy and atomic absorption spectrophotometry. We further characterized their interaction with dentin ultra-morphologically, using transmission electron microscopy. The monomer 10methacryloxydecyl dihydrogen phosphate ( 10-MDP) readily adhered to hydroxyapatite. This bond appeared very stable, as confirmed by the low dissolution rate of its calcium salt in water. The bonding potential of 4-methacryloxyethyl trimellitic acid ( 4-MET) was substantially lower. The monomer 2-methacryloxyethyl phenyl hydrogen phosphate ( phenyl-P) and its bond to hydroxyapatite did not appear to be hydrolytically stable. Besides self-etching dentin, specific functional monomers have additional chemical bonding efficacy that is expected to contribute to their adhesive potential to tooth tissue.

    Glass-ionomers are auto-adhesive to tooth tissue through combined micro-mechanical and chemical bonding. How much each of the two bonding components contributes to the actual bonding effectiveness is, however, not known and there is not much information available on long-term stability. The objective of this study was to assess the bonding effectiveness of a resin-modified glass-ionomer adhesive to dentin after 4 yr of water storage. Fuji Bond LC (GC) was applied without (i) and with pretreatment using (ii) a polyalkenoic acid conditioner and (iii) a 37.5% phosphoric acid etchant. The etchant was used to exclude any chemical interaction with hydroxyapatite. The micro-tensile bond strength (muTBS) to dentin decreased significantly over the 4 yr period in all three experimental groups. After 24 h and 4 yr, the lowest muTBS was recorded when dentin was not pretreated. The highest muTBS was obtained following polyalkenoic acid pretreatment, although this was not significantly different from specimens that were pretreated using phosphoric acid. Pretreating dentin with phosphoric acid intensified micromechanical interlocking at the expense of chemical bonding potential to hydroxyapatite. Nevertheless, correlating the muTBS data with failure analysis through scanning electron microscopy and transmission electron microscopy indicated that combined micro-mechanical and chemical bonding involving pretreatment with the polyalkenoic acid conditioner yielded the most durable bond.

    Purpose: To compare the microtensile bond strength (muTBS) to enamel of 10 contemporary adhesives, including three one-step self-etch systems, four two-step self-etch systems and three two-step total-etch systems, with that of a conventional three-step total-etch adhesive. Methods: Resin composite (Z100, 3M) was bonded to flat, #600-grit wet-sanded enamel surfaces of 18 extracted human third molars using the adhesives strictly according to the respective manufacturer's instructions. After storage overnight in 37degreesC water, the bonded specimens were sectioned into 2-4 thin slabs of approximately 1 mm thickness and 2.5 mm width. They were then trimmed into an hourglass shape with an interface area of approximately 1 mm(2), and subsequently subjected to muTBS-testing with a cross-head speed of 1 mm/minute. Results: The muTBS to enamel varied from 3.2 MPa for the experimental one-step self-etch adhesive PQ/Universal (self-etch) to 43.9 MPa for the two-step total-etch adhesive Scotchbond 1. When compared with the conventional three-step total-etch adhesive OptiBond FL, the bond strengths of most adhesives with simplified application procedures were not significantly different, except for two one-step self-etch adhesives, experimental PQ/Universal (self-etch) and One-up Bond F, that showed lower bond strengths. Specimen failures during sample preparation were recorded for the latter adhesives as well.

    Previously, we introduced a methodology to determine the chemical bonding potential of polyalkenoic acids to mineralized tissues through quantification of the degree of ionic bond formation between the carboxyl groups of a polyalkenoic acid with calcium of hydroxyapatite. In a continuation of that study, we now investigated in how far the chemical bonding potential to synthetic hydroxyapatite is influenced by the molecular structure of the polyalkenoic acid and if this also may affect the self-adhesiveness to enamel and dentin. X-ray photoelectron spectroscopy and inductively coupled plasma atomic emission spectrometry were used to quantitatively analyze the chemical bonding efficacy of a polyalkenoic acid consisting of acrylic acid units (PAA) to synthetic hydroxyapatite (HAp) in comparison to the chemical bonding potential of the previously investigated synthesized polyalkenoic acid (s-PA) co-polymer consisting of 90 w/w% acrylic and 10 w/w% maleic acid units. In addition, the analysis was carried out for enamel and dentin samples. PAA revealed a significantly lower bonding effectiveness with only half of its carboxyl groups bonded to HAp versus about two-third of the carboxyl groups of s-PA. The difference in bonding potential was confirmed by the considerably lower adhesiveness of PAA to enamel and dentin as compared to that of s-PA The present findings indicate that the molecular structure of the polyalkenoic acid significantly influences the chemical bonding efficacy to Hap-based substrates.     The purpose of this study was to evaluate the effect of dentin depth and tubule direction on the ultimate tensile strength (UTS) of human dentin. Dentin slabs of 0.5-mm thickness were trimmed either from the mesial and distal (for specimens with the tubules parallel to the tensile force; parallel group) or from the occlusal and pulpal surfaces (perpendicular group) to reduce the cross-sectional area of the superficial, middle, and deep regions to 0.25 mm(2), and subjected to microtensile testing. From SEM photomicrographs of the fractured specimens of the parallel group, the tubule density was investigated. For both parallel and perpendicular groups, superficial dentin showed a significantly higher UTS than deep dentin. The tubule density of superficial dentin was significantly lower than that of middle and deep dentin. When performing the microtensile bond test to deep dentin, it is possible that cohesive failure of dentin can occur at relatively low tensile stresses.

    Resin-dentin bonds degrade over time. The objective of this study was to evaluate the influence of variables like hybridization effectiveness and diffusion/elution of interface components on degradation. Hypotheses tested were: (1) There is no difference in degradation over time between two- and three-step total-etch adhesives; and (2) a composite-enamel bond protects the adjacent composite-dentin bond against degradation. The micro-tensile bond strength (muTBS) to dentin of 2 three-step total-etch adhesives was compared with that of 2 two-step total-etch adhesives after 4 years of storage in water. Quantitative and qualitative failure analyses were conducted correlating Fe-SEM and TEM. Indirect exposure to water did not significantly reduce the muTBS of any adhesive, while direct exposure resulted in a significantly reduced muTBS of both two-step adhesives. It is concluded that resin bonded to enamel protected the resin-dentin bond against degradation, while direct exposure to water for 4 years affected bonds produced by two-step total-etch adhesives.

    The aim of this study was to investigate the bond strength between indirect resin core-buildup and dentin treated with or without hypochlorite (NaOCl) gel. The post-space was made in five extracted human molars with obturated root canals, and the indirect resin core was produced on a gypsum model. Then, the dentin surface was treated with/without NaOCl, followed by luting of the resin core into the cavity. After storage for 24 h in 37degreesC water, the specimens were sectioned for measuring bond strength to the root and coronal dentin, and were subjected to micro-tensile bond strength (MTBS) testing. Two-way ANOVA showed that significant differences in MTBS were revealed for both factors of NaOCl treatment and dentin region. It is concluded that, for indirect resin core-buildup, the proper use of the NaOCl gel to dentin could increase the bond strength.

    Ineffective silane coupling between filler and matrix within dental composites is prone to accelerated in vivo degradation. In this study, we examined to what degree a procedure involving chemical decontamination of filler prior to silanization could improve the filler-matrix bonding, and thus the physico-mechanical properties, of composites. X-ray photoelectron spectroscopy revealed that filler-matrix coupling largely depended upon siloxane bridge (Si-O-Si) formation between the silica surface and the silane molecule, rather than on intermolecular bonding between adjacent mane molecules. Pre-silanization decontamination based upon boiling silica in 0.05-5% sodium peroxodisulfate, followed by ultrasonic rinsing in acetone, most effectively decontaminated filler. Consequently, it significantly improved the bonding of silane molecules to silanol groups at the silica surface. Experimental composites produced following pre-silanization decontamination of filler revealed a diametral tensile strength that was resistant to degradation by thermocycling.

    In order to study adhesion/decalcification mechanisms of acid interactions with human hard tissues such as bones and teeth, the chemical interaction of five carboxylic acids (acetic, citric, lactic, maleic, and oxalic) and two inorganic acids (hydrochloric and nitric) with enamel and two synthetic hydroxyapatite (HAp) powders with, respectively, a high and a low crystallinity were analyzed using X-ray photoelectron spectroscopy (XPS), atomic absorption spectrophotometry (AAS), and spectrophotometry (S). X-ray diffraction revealed that the crystallinity of the highly crystallized HAp was considerably higher than that of enamel while the crystallinity of the poorly crystallized HAp was similar to that of dentin and bone. XPS of acid-treated enamel demonstrated for all carboxylic acids ionic bonding to calcium of HAP. AAS and S showed for both HAps that all carboxylic and inorganic acids except oxalic acid extracted Ca significantly more than P, leading to a Ca/P ratio close to that of synthetic HAp (2.16 w/w). Oxalic acid extracted hardly any Ca, but substantially more P, leading to a significantly smaller Ca/P ratio than that of HAp. AAS showed that the calcium salt of oxalic acid hardly could be dissolved, whereas the calcium salts of all the other acids were very soluble in their respective acid solution. These results confirm the adhesion/decalcification concept (AD-concept) previously advanced. Depending on the dissolution rate of the respective calcium salts, acids either adhere to or decalcify apatitic substrates. It is concluded that the AD-concept that originally dictated the interaction of carboxylic acids with human hard tissues can be extended to inorganic acids, such as hydrochloric and nitric acid. Furthermore, HAp crystallinity was found not to affect the adhesion/decalcification behavior of acids when interacting with apatitic substrates, so that the AD-concept can be applied to all human hard tissues with varying HAp crystallinity.     Objective: A new type of so-called 'packable', 'condensable' or 'mouldable' composite has been developed and aims at replacing amalgam for posterior restorations. The purpose of the present investigation was to study the dynamic elastic modulus of 12 packable composites, and to follow the evolution of this property following prolonged water absorption. Methods: Of each material ten rectangular samples (1.5 x 5 x 35 mm) were prepared. The elastic modulus (GPa) of each sample was determined with a non-destructive dynamic method using a Grindo-Sonic after 24 h of dry storage at room temperature, and after 24 h, 1, 3 and 6 months of wet storage at 37 degreesC. All data were analyzed using two-way ANOVA, Bonferroni/Dunn's test for multiple comparisons and paired t-test with a significance level of p < 0.05. In addition, inorganic filler volume percentages were derived from the phenomenological model introduced by Braem et al. [11]. Results: The studied materials varied widely in terms of elastic modulus, ranging between composites classified as Compact-Filled Densified (elastic modulus of 23.4 +/- 2.4 GPa) and as Microfine (elastic modulus of 8.5 +/- 2.1 GPa). Significance: The great diversity observed in the elastic modulus of this type of composites necessitates clear specifications with regard to 'first' the definition of marketing terms such as packable and so on, and 'second' the justified use in posterior teeth.     Intracanal exudates in persistent endodontic cases were sampled for detecting bacteria using a preculture method, and the antibiotic susceptibility of the bacteria identified was examined, Twelve species were recovered from 15 cases. The species most commonly detected were alpha-Streptococcus and Enterococcus, The results of the antibiotic susceptibility tests revealed that Enterococcus was highly resistant to antibiotics tested, especially to cephalosporin products, Enterococcus is known as one of the bacteria that show multidrug resistance. In cases of prolonged endodontic treatment, a specific antibiotic prescription would play an important role for efficient treatment. Collecting data about antibiotic susceptibility could be helpful. Identification, however, take similar to 2 wk and a further study to develop the method that rapidly informs us about the antibiotic susceptibility that might be needed.

    The purpose of this study was to evaluate the bleaching effect of sodium percarbonate on artificially stained pulpless teeth. Twenty extracted human mandibular premolers were stained by immersing them in human blood mixed with iron sulfide, and divided into three test groups and one control group. The following bleaching agents were used: sodium perborate mixed with 30% hydrogen peroxide (WB), sodium percarbonate mixed with distilled water (PW) or 30% hydrogen peroxide (PH), On days 5, 10, and 15, each pulp chamber was refilled with fresh bleaching paste. The L* (average lightness) of the midbuccal area of specimens was measured before and after staining, and on days 5, 10, 15, and 20 by colorimeter. The bleaching effect was evaluated by the lightness recovery rate (LRR) calculated from L*. WB showed the highest LRR among all groups, and PW and PH showed significantly higher LRR when compared with the control group, PW and PH were not significantly different. Results showed that sodium percarbonate had an obvious bleaching effect without hydrogen peroxide and indicated that it could be a safe intracoronal bleaching agent.

    The structures and electronic states of alkyl-radical-functionalized C-20 fullerenes (denoted by C-20-R) have been investigated using density functional theory (DFT). The different alkyl radicals investigated were methyl, ethyl, propyl, and butyl radicals. The DFT calculation indicated that the alkyl radical binds to the carbon atom of C-20 in the on-top site, thus forming a strong C-C single bond. The binding energies of the alkyl radicals to C-20 were calculated to be 83.9-86.6 kcal/mol at the CAM-B3LYP/6-311G(d,p) level. The electronic states of the C-20-R complex are discussed on the basis of the theoretical results. (C) 2016 The Japan Society of Applied Physics

    歯科治療における象牙質の接着には、樹脂含浸層の形成による機械的嵌合に加え、化学的な結合が関与する。特に化学的な結合に関しては、機能性モノマーの構造や純度が影響する。また、HEMAなど他のモノマーが共存する場合では、歯質との反応性が異なることも報告されている。異なるスペーサーの長さを持つ機能性モノマーの化学的反応性、機能性モノマーの純度が接着性・接着耐久性に与える影響、フッ素を含有した機能性モノマーについて述べた。

    Adhesive technology has evolved rapidly since it was introduced more than fifty years ago. Today, decayed or fractured teeth can be reconstructed minimally invasively and nearly invisibly using adhesive technology. However, the clinical longevity is sometimes still too short. Although water sorption is thought to be the main destabilizer of the biomaterial-tooth bond, the actual interfacial degradation mechanisms are far from understood. The fundamental mechanism of bonding to enamel and dentin is essentially based on an exchange process, in which minerals removed from the dental hard tissues are replaced by resin monomers that upon polymerization become micro-mechanically interlocked in the created porosities. In addition to micro-mechanical interlocking through hybridization, the potential benefit of additional chemical interaction between functional monomers/polymers and tooth substrate components has recently regained attention. In this review paper, we focus on how chemical interaction at the biomaterial-hard tissue interface can improve bond durability. © 2012 Japanese Association for Dental Science.

    Purpose: To evaluate the hydrolytic stability of three one-step self-etching adhesives (1-SEAs) bonded to dentin through bond strength testing and ultra-morphological interfacial analysis before and after long-term thermocycling. Materials and Methods: Eighteen flattened mid-coronal dentin surfaces of extracted human molars were subjected to bonding treatment using three 1-SEAs (Clearfil S(3) Bond, Kuraray (S3), G-Bond, GC (GB), Absolute, Dentsply-Sankin (AB)), after which the bonded surfaces were built up with composite. After storage overnight at 37 C, the specimens were sectioned into slabs and further trimmed into an hourglass shape with an interface area of approximately 1 mm(2). The specimens were left untouched (control) or were thermocycled for 100,000 cycles. The microtensile bond strength (mu TBS) was measured and the ultrastructure of the adhesive/dentin interface characterized using transmission electron microscopy (TEM). Results: Long-term thermocycling significantly decreased the mu TBS of all one-step adhesives tested (p < 0.05, one-way ANOVA and Games-Howell test). TEM revealed a similar interfacial ultrastructure before and after thermocycling for S3. For GB, many voids were observed at the interface after 100,000 thermocycles. Regarding AB, collagen fibrils could no longer be clearly observed upon staining, while the adjacent unaffected dentin was rich in voids. Conclusion: The bond strength and ultramorphological data demonstrated that the bond of 1-SEAs to dentin degrades with time, although the degree of degradation is obviously material dependent.

    本研究の目的は,種々の研削器具を用いて生成されたスミヤー層の超微細構造を検討し,2種の市販1ステップ接着システムの象牙質への短長期的接着性能に対して,スミヤー層が与える影響を評価することである.健全ヒト抜去大臼歯歯冠部から,2種のダイヤモンドポイント(レギュラー,スーパーファイン)および2種のSiCペーパー(#600,#2,000)を用いて,可及的平坦な研削象牙質面を得た.各研削器具の表面,研削象牙質表面および割断面をSEM観察し,スミヤー層の厚さ,および各研削器具表面粒子の大きさを測定した.また,各研削象牙質面のTEM観察を行った.次に,2種の研削象牙質面(レギュラー,#2,000)を作製し,Clearfil S3Bond(クラレメディカル,S3)およびAbsolute(デンツプライ三金,AB)を用いて,指示書どおりに処理した後,光照射をせず,ただちにアセトン洗浄し,脱水乾燥を行った.それらの試料の処理面および割断面をSEM観察した.さらに,レギュラーおよび#2,000研削象牙質面に,S3およびABを用いてコンポジットレジンを築盛し,24時間水中保存後,接着界面の面積が約1mm2となるよう試料をダンベル型に調整した.それらの試料をサーマルサイクリング(TC)負荷試験を行うものと行わないものに分け,試料製作後ただちに,あるいは100,000回TC負荷試験に供した後に,微小引張り接着強さの測定および接着界面のTEM観察を行った.スミヤー層は研削器具の表面粒子が大きくなると厚くなったが,形態的な差異は認められなかった.S3は,スミヤー層の厚さが異なる研削面によりスミヤー層の除去程度は異なった.ABでは,研削面の違いによらずスミヤー層が除去された.S3は#2,000研削面に比較し,レギュラー研削面への接着強さが有意に低かったが,ABでは研削面の違いによる接着強さに有意差はなかった.また,TC負荷後,いずれの接着システムも接着強さは有意に減少し,接着界面の劣化を示唆する像が観察された.以上の結果より,1)スミヤー層の厚さは研削器具に左右されること,2)スミヤー層の除去能が低い接着システムでは,厚いスミヤー層の場合,スミヤー層が残存し,接着性能に影響を及ぼすこと,3)接着システムによっては,その厚さにかかわらずスミヤー層を除去でき,接着性能に影響しないものもあること,4)1ステップ接着システムは長期的に接着性能が低下することが示唆された.(著者抄録)

    Objectives. The objectives of this study were (1) to clarify the relationship between the duration of air-drying of one-step self-etch adhesives (1-SEAs) and the evaporation degree (ED) of solvents, and (2) to evaluate the effect of ED on the ultimate micro-tensile strength (mu TS) of the adhesives. Methods. The ED of one HEMA-rich I-SEA, Clearfil S' Bond (Kuraray), and two HEMA-free 1-SEAs, iBond (Heraeus-Kulzer) and G-Bond (GC), was determined without air-drying and after, respectively, 5- and 10-s air-drying using a gravimetric method. Next, the mu TS of the adhesives at ED equivalent to 0-, 5- and 10-s air-drying was measured. Results. The ED increased with extension of air-drying time. Among the adhesives tested, iBond showed the largest ED, followed by G-Bond and Clearfil S-3 Bond in this order. A longer air-drying time for 10-s resulted in a statistically significantly higher mu TS for the HEMA-rich Clearfil S3 Bond. The mu TS of the latter was higher than that of the other two HEMA-free adhesives for each air-drying time. Significance. Air-drying of 1-SEAs had a significant effect on the degree of solvent evaporation (ED) and also on the mechanical properties (mu TS) of the 1-SEAs upon setting. It is therefore beneficial to remove solvents of the 1-SEAs as much as possible by thorough, strong air-drying in order to achieve a strong adhesive layer at the interface.     シングルステップボンディングシステムIMB-3(IM)およびG-Bond(GB)の象牙質に対する接着強さを、2種のコンポジットレジンAP-X(AP)およびEsteliteΣ(ES)を充填材料として、微小引張り接着試験を用いて比較検討した。ヒト抜去大臼歯の歯軸に垂直な象牙質平坦面を、レギュラータイプのダイヤモンドポイントにより研削、各接着材料を取扱説明書どおりに塗布し、それぞれのコンポジットレジンを接着した。37℃水中24時間保管後、これら4グループの接着試料は1×1mmのスティック状にトリミングされ、クロスヘッドスピード1.0mm/minにて微小引張り接着試験に供された。データはANOVAおよびGames/Howell法(p<0.05)により統計処理された。本実験では、IMを異なるコンポジットレジンを用いて象牙質に接着した場合、ESを用いたグループがAPグループより有意に高い接着強さを示した。GBではコンポジットレジンESとAPをそれぞれ用いた場合、接着強さに有意な差はみられなかった。これらの相違を引き起こす要因としては、コンポジットレジンの機械的性質の違いの影響が示唆された。(著者抄録)

    Ionic bonding between polyalkenoic acid and hydroxyapatite may explain the excellent bonding retention of glass-ionomers in clinical trials. We have here investigated the extent to which the self-adhesiveness of resin-modified glass-ionomers (RMGIs) can be attributed to this chemical bonding capacity. Therefore, the interaction of 3 RMGIs with tooth substrates was comprehensively characterized, with electron and atomic force microscopy correlated with x-ray photoelectron spectroscopy (XPS). Interfacial ultrastructural analysis for 2 RMGIs disclosed a shallow hybridization of hydroxyapatite-coated collagen, on which a submicron gel phase was deposited through reaction of the polyalkenoic acid with calcium extracted from the dentin surface. One RMGI, however, bonded to dentin without hybrid layer or gel phase formation. XPS indicated that polycarboxylic acids included in the RMGIs electrostatically interacted with hydroxyapatite. We conclude that the self-adhesiveness of RMGIs should be attributed to ionic bonding to hydroxyapatite around collagen, and to micro-mechanical interlocking for those RMGIs that additionally hybridize dentin.

    Objectives. Flexure at the tooth cervix has been suggested to dislodge cervical restorations more rapidly The objective of this randomized controlled clinical trial was to test the hypothesis that a higher composite flexibility better withstands tooth flexure stress. Methods. One hundred and forty-two non-carious cervical lesions were restored with composites with contrasting stiffness. Seventy-one patients randomly received two cervical restorations placed following two out of the three experimental groups: (1) the three-step etch-and-rinse adhesive Permaquick applied with the stiff micro-hybrid composite Amelogen Hybrid (PMQ/A-Hy, Ultradent), (2) Permaquick applied with the more flexible micro-filled composite Amelogen Microfill (PMQ/A-Mi, Ultradent), or (3) the 'golden standard' three-step etch-and-rinse adhesive Optibond FL applied with the micro-hybrid composite Prodigy (O-FL/Pro, Kerr). Results. The recall rate at 7 years was 80%. No statistically significant difference was found for any of the parameters evaluated neither between both adhesives, nor between both composites (McNemar, p > 0.05). Eleven percent of the O-FL/Pro restorations were clinically unacceptable due to loss of retention (5.5%) and severe marginal discoloration (5.5%). In the PMQ-group, 22% of the PMQ/A-Mi restorations (8% loss of retention, 5% severe enamel margin defects, 3% severe dentin margin defects, 6% severe marginal discoloration, 3% extreme sensitivity) and 19% of the PMQ/A-Hy restorations (13% loss of retention, 3% severe enamel margin defects, 3% severe marginal discoloration) needed repair or replacement. Significance. The clinical performance of the three adhesive/composite combinations was good and reliable during the 7-year clinical trial. It was concluded that the composite stiffness does not affect the clinical longevity of cervical composite restorations. (C) 2006 Academy of Dental Materials. Published by Elsevier Ltd.     シングルステップボンディングシステムDCB-100(DC),Clearfil S3 Bond(TS)および2ステップボンディングシステムClearfil SE Bond(SE)の接着直後の接着強さを、微小せん断接着試験を用いて比較検討した。ヒト抜去大臼歯象牙質面に各接着材料を仕様説明書どおりに塗布し、そこにコンポジットレジンを接着、接着終了10分および24時間後に微小せん断接着強さを測定した。データはone-way ANOVAおよびDuncan's new multiple range testにて処理した。接着強さに関しては、時間経過による有意な差はみられなかった。接着から測定までの時間が同じであるグループ間では、接着10分後の場合、SEのほうがDCよりも有意に高い値であった。マクロな破断形態は、TS10分後のグループではボンディング中の破断および界面剥離が少数みられ、他グループではほぼ混合破壊であった。ただしミクロでは、同じ接着材料でも10分と24時間のグループ間で破断部位の異なる様子が観察され、触媒の組成を含めての重合系の違いが関与していると推測された。(著者抄録)

    Objective: Phase-separation within HEMA-free all-in-one dental adhesives may result in the entrapment of droplets within the adhesive resin. Strongly air-blowing prior to polymerization, can remove most of these droplets. The objective of this study was to evaluate the effect these droplets may have on the resistance of the adhesive-tooth interface to NaOCl degradation. Methods: The micro-tensile bond strength (mu TBS) to enamel and dentin was determined when a HEMA-free all-in-one adhesive was applied either following a mild or strong air-blowing technique. The bonds were also exposed to an aqueous sodium hypochlorite (NaOCl) solution for 1 h, following a recently introduced methodology to mimic in vivo bond degradation. Results: This study revealed that strong air-blowing of the adhesive only resulted in a significantly higher micro-tensile bond strength (mu TBS) to dentin, but not to enamel. Likewise, NaOCl only reduced the mu TBS to dentin for both the mild and strong air-blowing technique, but again not the mu TBS to enamel. Failure analysis by SEM clearly revealed that strong air-blowing is less effective in droplet removal when the adhesive was applied in small and narrow class-I cavities, as compared to when it was applied to flat surfaces. Conclusions: NaOCl did preferentially dissolve the hybrid layer at dentin, and more for the mild than for the strong air-blowing technique. A strong air-blowing procedure resulted in a more NaOCl-resistant hybrid layer, so that it can be concluded that a HEMA-free one-step adhesive definitely benefits from a strong air-blowing technique.     Adhesive-dentin interfaces degrade with time. This study determined the effect water storage may have on the bonding effectiveness of adhesives to occlusal Class I cavity-bottom dentin. Six adhesives, all representing contemporary classes of adhesives, were applied: a 3-step (OptiBond FL, Kerr) and 2-step (Scotchbond 1*, 3M ESPE) etch-and-rinse adhesive, a 2-step (Clearfil SE, Kuraray) and 1-step (Adper prompt, 3M ESPE) self-etch adhesive and a 2-step (FujiBond LC, GC) and 1-step (Reactmer, Shofu) resin-modified glass-ionomer adhesive. Bonding effectiveness was assessed by microtensile bond strength testing (mu TBS) and electron microscopy (Feg-SEM and TEM). The mu TBS was determined after 1 day and 1 year water storage of the entire restored cavity (indirect exposure of the adhesive-dentin interface to water) and prepared mu TBS-beams (direct exposure of the adhesive-dentin interface to water). The hypotheses tested were: (1) resin-dentin bonds formed at the bottom of Class I cavities resist 1-year water storage and (2) an adjacent composite-enamel bond protects the composite-dentin bond against degradation. Non-parametric Kruskal-Wallis analysis statistically analyzed the mu TBSs. The first hypothesis was rejected, as only the mu TBS of OptiBond FL and Clearfil SE did not significantly decrease after 1-year direct and/or indirect water storage. The second hypothesis was corroborated, as the bonding effectiveness of most simplified adhesives (Scotchbond 1, Adper Prompt, FujiBond LC and Reactmer) approached 0 (because of the frequent pre-testing failures) after 1-year direct water exposure. The second hypothesis concluded that the 3-step etch-and-rinse adhesive must still be regarded the "gold standard." Though mu TBS decreased significantly, Clearfil SE, as a 2-step self-etch adhesive, was the only simplified adhesive to perform reliably after 1-year direct water exposure.

    The purpose of this study was to evaluate the morphological appearance of resin-dentin interface produced by 1-step adhesive systems in order to classify them, using TEM. It was hypothesized that all interfaces produced by each 1-step adhesive show characteristics similar to that produced by the conventional self-etching system rather than that produced by etch-and-rinse system. The flattened dentin surfaces were subjected to bonding treatment using either a 2-step adhesive: single bond, mega bond, unifil bond; or a 1-step adhesive: Prompt L-Pop, absolute, xeno CF, AQ bond, one-up bond F, G bond, and SSB-200. A flowable resin was then applied to these surfaces. The results revealed that resin-dentin interfaces produced by 1-step systems were classified into three categories. Additionally, the interface produced by G bond and SSB-200 showed a different morphological appearance from that of the others. The interface produced by these adhesives was thinner and less demineralized, with the presence of hydroxyapatite-coated collagen fibrils. It is proposed that "Nanointeraction Zone (NIZ)" is a new complementary classification for partially demineralized resin-dentin interdiffusion zones. Thus, one-step self-etching adhesive systems can be classified into three categories, etch-and-rinse type, mild self-etching type, and NIZ type, by their morphological appearance of the interface.     The favorable self-adhesiveness of resin-modified glass ionomers (RMGIs) might be even further improved if the time-consuming and technically sensitive etch-and-rinse pre-treatment step with polyalkenoic acids could be avoided. We undertook this study to assess the effectiveness of an experimental self-etch adhesive for RMGIs that does not need to be rinsed off. Ultrastructural analysis and micro-tensile bond strength testing to enamel and dentin of a RMGI restorative material and a RMGI adhesive were performed after 4 different surface pre-treatments: no conditioning; 25% polyalkenoic acid; an experimental self-etch adhesive; and 37.5% phosphoric acid followed by the experimental self-etch adhesive. The use of an experimental self-etch adhesive increased the bond strength of RMGIs, especially after an additional conditioning step. Interfacial analysis showed the formation of a thin hydroxyapatite-containing hybrid layer. The self-etch technique enhances the user-friendliness of RMGIs and lowers their technique-sensitivity, while maintaining desirable characteristics of the conventional etch-and-rinse approach with polyalkenoic acids.

    歯質接着システムにおいて、ボンディング材の機械的強度が高い場合に高い接着性能が発揮されることが報告されてきた。臨床上、接着操作直後の接着性能も重要であり、この時期のボンディング材の強度を知る必要がある。本研究の目的は、ボンディング材の光照射直後の引張り強さを評価することである。3種の2ステップセルフエッチングシステム、クリアフィルメガボンド(MB)、フルオロボンド(FB)およびユニフィルボンド(UB)のボンディング材を使用した。引張り強さは、ボンディング材への光照射直後、コンポジットレジンへの光照射直後および24時間経過後の3つの段階で評価した。その結果、ボンディング材の引張り強さは、MBが最大で、以下FB,UBの順であった。3つの段階での比較では、ボンディング材への光照射後、コンポジットレジンへの光照射後、24時間経過後の順に大きくなった。ボンディング材への光照射直後では、重合が十分に進行しておらず、コンポジットレジンへの光照射やpost irradiation hardeningにより重合が進行し、引張り強さが大きくなったと考えられる。(著者抄録)

    Besides micro-mechanical interlocking through hybrid-layer formation, self-etch adhesives may benefit from additional chemical interaction between the functional monomer and residual hydroxyapatite. One-step adhesives are commonly associated with lower bonding effectiveness, which must be attributed in part to the dissolution of hydrophilic and hydrophobic monomers in a relatively highly concentrated solvent. In this 'difficult' mixture, also water is essential as ionization medium to enable self-etching activity. Due to the high hydrophilicity, one-step self-etch adhesives have been reported to behave as semi-permeable membranes, allowing fluids to pass through and seriously jeopardizing bond durability. Recent research has also revealed that HEMA-free one-step adhesives are prone to phase-separation, which may also account for their lower bonding effectiveness. Employing an appropriate air-drying technique may, however, improve the bonding effectiveness of such phase-separating adhesives by getting rid of substantially more interfacial water than HEMA-containing adhesives, of which water might be more difficult to remove from.

    Objectives: The purpose of this study is to investigate the effect of remaining dentin thickness and the use of a 20% polyalkenoic acid conditioner on the micro-tensile bond strength of a glass-ionomer adhesive to dentin. Methods: Resin composite was bonded to flat dentin surfaces from 14 extracted human teeth using Fuji BOND LC (GQ with or without a polyalkenoic acid conditioner, then sectioned to thin slabs, trimmed into an hourglass shape with the area of the interface being approximately 1 mm(2), and subjected to micro-tensile testing at a cross-head speed of 1 mm/min. Micro-tensile bond strengths were determined at three depth levels with a remaining dentin thickness of more than 3 mm, between 2 and 3 mm, and less than 2 mm. Failure modes of the broken interfaces were determined using field-emission scanning electron microscopy. Results: The micro-tensile bond strength to dentin significantly improved when the remaining dentin thickness increased and the conditioner was used. When the conditioner was used (irrespective of remaining dentin thickness), failures mainly occurred adhesively at the interface between the adhesive and resin composite. When no conditioner was used, no adhesive failures between the adhesive and resin composite occurred, but failures occurred mainly adhesively between dentin and the adhesive, or mixed adhesive-cohesively. Significance: The bonding effectiveness of the glass-ionomer adhesive tested was affected by the area of intertubular dentin available for micro-mechanical retention through hybrid-layer formation.     The effect of configuration factor (C-factor) on cavity adaptation was investigated in three compomer and one resin composite restorations. Eighty-four cylindrical dentin cavities (C-factor: approximately 2.5, 3.0 or 4.0) prepared on flat coronal dentin surfaces were filled with the materials in combination with their proprietary adhesive systems. Cavity adaptation was microscopically examined after 15 minutes storage in water at the top surface and at other four sites along the cavity walls. Additionally, indentation testing was performed for each material at 20 minutes and 24 hours after irradiation. Regression analysis revealed no relationship between C-factor and gap dimension in compomer restorations at any of the measuring sites, while a logarithmic relation was found only at the cavity floor of the composite fillings. All materials showed maturation of mechanical properties. The elastic component of the indentation was smaller in compomers than in the composite. It was concluded that C-factor had no influence on the cavity adaptation for compomer restorations. This might be due to reduced stress generation at the bonding interface caused by relatively low mechanical properties immediately after curing, less elasticity, and water absorption in compomers.

    H24年度は、各種1ステップ接着システムの微小引張り接着強さを再測定し、またその前の23年度から引き続き行っていた臨床的スミア層溶解能の透過型電子顕微鏡(TEM)による観察を行った。さらに、予定より少し遅れたものの、100,000回のサーマルサイクリング(TC)負荷試験を開始しH25年6月に終了予定であるので、H25年度に負荷後試料の微小引張り接着試験とTEM観察を行う予定である。1.接着強さの再測定TC試験は当初岡山大学で行う予定であったが、本学の設備で行うこととしたため、試料の形態を変える必要性が生じた。そのため、その形態に合わせたコントロールの接着強さを再測定した。ヒト抜去健全大臼歯歯冠部を水平に切断後、2種の臨床的スミア層を産生するためダイヤモンドポイント(レギュラー)および耐水研磨紙(#2000)を用いて可及的平坦な被着象牙質面を得た。この面に対し、Clearfil Tri-S Bond(S3), Hybrid Coat II(HC), BeautiBond(BB), Bond Force(BF), Absolute2(AB),あるいはG-Bond Plusの接着システムを用いて処理し、通法に従って微小引張り試験を行ったが、試料の形態はダンベル型ではなくスティック(ビーム)型とした。その結果、前年度とほぼ同様の値が得られた。2.TEM観察スミア層の溶解能を検討するためにH22年度に表面をSEM観察した試料を用い、H23年度から継続して行っていたTEM観察の結果、ABではレギュラー、#2000によらず、スミア層は脱灰され、コラーゲン線維網と思われる像が観察されたが、そのほかのシステムではレギュラー研削面のほうがスミア層が残存している像が観察された。

    簡便で短時間に処置が終了し、化学的に歯質と接着し、さらに長期にわたり安定した接着性能を発揮できる接着システムの開発を最終目的とし、その基礎的知見を得るために、臨床的な条件下で作製したスミア層を形態学的に解析するとともに、各種スミア層を有する歯面に対する簡略化ステップ接着システムの短長期的接着性能を検討した。その結果、スミア層の厚さは切削器具に左右されること、スミア層の除去能が低い接着システムでは接着性能に影響があること、1ステップ接着システムは長期的に接着性能が低下することが判明した。

    1.短期的接着性能に関する研究(1)研削面の違いが接着性能へ及ぼす影響2種のダイヤモンドポイントを用いて形成した歯質研削面に対する各種接着システムの短期的接着性能を、微小引張り接着強さとその破断面のSEM観察の結果、1ステップ型接着システムでは、象牙質接着に対して有利な研削面を形成するためのバー選択が必要であると考えられた。(2)簡略化ステップ型接着システムの接着界面分類1ステップ型接着システムを、#600SiC研磨象牙質面へ接着した場合、その接着界面は(1)樹脂含浸層形成型、(2)機能性傾斜材料型、(3)ナノ相互作用層形成型に分類された。特に(3)は脱灰される歯質の量が少なく、レジンの浸透が充分に期待できるため、劣化の少ない長期に安定な材料であることが期待された。2.長期的接着性能に関する研究(1)2ステップ型セルフエッチングシステム3種の2ステップ型セルフエッチングシステムの加水分解に対する安定性を、100000回サーマルサイクリング試験後の接着強さならびにTEMによる接着界面像を指標に検討した。その結果、接着界面の長期安定性は、機能性モノマー自体の加水分解安定性や機能性モノマーと象牙質との化学的相互作用に依存していることが示唆された。(2)1ステップ型接着システム3種の1ステップ型接着システムについて100000回サーマルサイクリング試験前後の接着強さ測定と接着界面をTEM観察した結果、材料によりサーマルサイクリングの影響の程度は異なるものの、何らかの変化(劣化)が象牙質接着界面に起こっていることが示唆された。3.各種機能性モノマーの歯質に対する化学的相互作用の分析X線光電子分光法分析により、機能性モノマーは歯質と化学的に相互作用することが示された。10-MDPはハイドロキシアパタイトと最も強く反応するだけではなく、Caとの結合は加水分解に対し最も安定であることが判明した。

    難治性感染根管の原因を明らかにするためには根管内滲出物の採取分析が重要であるが、採取できる試料が微量であるために細菌叢や病態の把握が困難である。そこで試料中の嫌気性菌を簡便・確実に分離同定する方法として、採取した滲出物を増菌用培地に接種して24時間嫌気的に培養した後に検査所に試料を送る方法(増菌法)で培養同定を行い良好な結果を得た。難治性感染根管22例について調べた所、18例で何らかの菌が検出された。その内訳はα‐Streptococcus、En‐terococcusなど通性嫌気性菌8種、偏性嫌気性菌4種、好気性菌2種が検出された。Ent‐erococcusは従来の報告よりも多数検出された。以上の結果より、増菌法は従来の方法より確実に滲出物中の細菌を検出できると思われた。次に、Enterococcusは長期症例や難治性感染根管から検出される頻度が高いとの報告が近年なされていることから、同菌種の宿主細胞への影響をより詳細に検討した。分離回収可能であった5株のEnterococcusについて分子生物学的手法により比較したところ、全ての株の由来が異なっていた。また、培養細胞系に感染させ、細胞内情報伝達計(チロシンリン酸化)に及ぼす影響を調べたが、同菌の直接接触ではチロシンリン酸化に変化は認められなかった。このことから、Enterococcusは細胞間情報伝達物質を介して宿主との反応に関与しているのではないかと考えられた。そこで、さらに免疫学的手法を用いて原因を明らかにする必要があると思われた。
