English, Scientific journal, Studies of postglacial range shifts could enhance our understanding of seaweed species' responses to climate change and hence facilitate the conservation of natural resources. However, the distribution dynamics and phylogeographic diversification of the commercially and ecologically important kelp Saccharina japonica in the Northwest Pacific (NWP) are still poorly surveyed. In this study, we analyzed the evolutionary history of S. japonica using two mitochondrial markers and 24 nuclear microsatellites. A STRUCTURE analysis revealed two partially isolated lineages: lineage H, which is scattered along the coast of Japan; and lineage P, which occurs along the west coast of the Japan Sea. Ecological niche modeling projections to the Last Glacial Maximum (LGM) revealed that the southern coasts of the Japan Sea and the Pacific side of the Oshima and Honshu Peninsulas provided the most suitable habitats for S. japonica, implying that these regions served as ancient refugia during the LGM. Ancient isolation in different refugia may explain the observed divergence between lineages P and H. An approximate Bayesian computation analysis indicated that the two lineages experienced post-LGM range expansion and that postglacial secondary contact occurred in Sakhalin. Model projections into the year 2,100 predicted that S. japonica will shift northwards and lose its genetic diversity center on the Oshima Peninsula in Hokkaido and Shimokita Peninsula in Honshu. The range shifts and evolutionary history of S. japonica improve our understanding of how climate change impacted the distribution range and diversity of this species and provide useful information for the conservation of natural resources under ongoing environmental change in the NWP. - Chocolate and tea made from kelp

English, The DNA fragment analysis can become an effective tool to study genetic differences between not only species but also individuals on saccharinan kelp from which the little genetic diversity was reported. This time, extraction methods of suitable DNA for use in the analysis with a capillary sequencer was examined on Saccharina japonica var. diabolica that contains polysaccharide abundantly. When AFLP was performed using genomic DNA extracted by seven different methods: (1) commercial kit, (2) original CTAB method, (3)-(5) three types of modified CTAB method, (6) modified SDS method, (7) combination of CTAB method and SDS method, high reproducible peak that was worth for the analysis was noticeable in the electropherogram in the experiment with the last combination method (7). It is considered that the pretreatment washing of polysaccharide and the subsequent purification for protein and RNA in SDS method and for polysaccharide in CTAB method are effective to obtain the high purity DNA. - A new name for Laminaria sachalinensis(Laminariales, Phaeophyceae)

The inhibition of germination and development of zoospores originating from Laminaria angustata originated on various kinds of hydrogels is studied. The effects of the water content of the gel (the degree of swelling), the electrical nature (neutral, positive charge, negative charge), the charge density, the counterions of hydrogels on the inhibition of zoospore germination and development of gametophytes are phenomenologically investigated. Among the gels investigated, poly(acrylic acid) (PAA) gel showed a dramatic inhibition to germination, and might serve as a novel, environmentally safe inhibitor of zoospore adhesion.

When maturation was induced by short-day conditions in naturally collected Saccharina japonica sporophytes as well as in the sporophytes cultivated, sorus formation began in approximately three weeks and spread to 0.4-16.8% on the upper side area and 23.2-41.1% on the underside area after about five weeks, with the highest rates, 23.2-59.5%, observed around the base part of the underside. Sorus formation began from the base and median parts in the margin-intact algae and from the marginal part in the margin-cut algae. Given that the released zoospore germinants grew well as seedlings for aquaculture, seedling production through the induction of sporophyte maturation is a feasible option for seedling production facilities and may be useful for stably producing seedlings without being influenced by the natural environment.

Based on Parker, Baker and Smith’s theory (PBS theory) for the evolution of anisogamy, we tested disruptive selection on gamete size. We can expect that more strongly anisogamous species produce larger female gametes. Also, the relationship between the size of male gametes and the degree of anisogamy remains untested. In anisogamous species, the PBS theory predicts that male gametes are soon minimized in size. Because the evolution of isogamy with a gamete of zero size is favoured by selection there must be the minimum size of gametes. In such cases, the degree of anisogamy would not depend on the size of male gametes. We confirmed that, as the degree of anisogamy increases, the size of female gametes increases, and that in strongly anisogamous species, the degree of anisogamy does not depend on male gamete size. These results well support the PBS theory.
