  • Nonphotic Entrainment and Phase-Shifting of Circadian Rhythms by Novelty-Induced Wheel Running in Female Mice.

    English, Scientific journal, Environmental light-dark (LD) cycles serve as the primary zeitgeber for the mammalian circadian system, but scheduled physical activity such as voluntary wheel running can act as a potent nonphotic cue. Previous studies on nonphotic entrainment have predominantly used male animals, leaving sex-specific mechanisms largely unexplored. In this study, we investigated circadian behavioral entrainment and phase-shifting responses to scheduled wheel-running activity in female C57BL/6J mice under constant darkness (DD). Mice underwent scheduled daily exposure to a novel cage with a running wheel (NCRW) for 3 hours, and spontaneous locomotor rhythms were analyzed before, during, and after exposure. Fifteen of 16 female mice achieved steady-state entrainment when the NCRW schedule coincided with activity onset. However, they lacked both anticipatory behavior and behavioral after-effects of entrainment-defined as persistent changes in circadian period and activity timing following entrainment-unlike what has been previously reported in males. To interpret entrainment dynamics, we constructed phase response curve (PRC) for a single 3-hour pulse of exposure to NCRW in both sexes. Female PRC displayed prominent delay portions in the late subjective day and early subjective night, contrasting with male profiles showing minor advances in the same windows. These findings reveal a sex difference in nonphotic circadian entrainment and suggest that underlying mechanisms, including oscillator coupling or differential sensitivity to nonphotic stimuli, may vary between males and females. The results emphasize the importance of considering biological sex in circadian research and provide a basis for further investigation into sex-specific regulation of behavioral rhythms.
  • Circadian Phase Assessment of Core Body Temperature Using a Wearable Temperature Sensor Under the Real World.

    English, Scientific journal, OBJECTIVE: To evaluate whether a patch-type wearable temperature sensor (CALERA Research) could determine the circadian phase of core body temperature (CBT) in a manner like a rectal probe. OBJECTIVE: Sixteen participants (27 ± 11 years, 8 males and 8 females) wore an actigraph and CALERA Research sensor on the chest region for 3-5 days in a real-world setting. Simultaneous rectal temperature measurements were performed during the nocturnal sleep period. The midpoints of the nocturnal decrease in CBT (CBT trough ) were used as the circadian phase marker. We analyzed 60 pairs of CBT trough . The reliability and agreement of the CBT trough from the two devices were analyzed using the intraclass correlation coefficient (ICC) and the concordance correlation coefficient (CCC). The Bland-Altman analysis was used to quantify the limit of agreement of CBT trough between the devices. OBJECTIVE: The ICC of 0.96 (95%CI: 0.93-0.98) and CCC of 0.96 (95%CI: 0.93-0.97) values indicated excellent reliability and substantial agreement, respectively. The mean bias was 0.16 hours (95%LoA: -0.76-1.07 hours). The mean CBT trough comparison was 5.9 ± 1.6 hours in the CALERA Research sensor and 5.8 ± 1.7 hours in the rectal probe. CONCLUSION: The difference in the CBT trough between the two devices was about ± 1.0 hour which would be an acceptable range for determining the CBT trough . We suggest that the CALERA Research sensor could be a useful tool for reasonably estimating the circadian phase of CBT trough and providing a surrogate for a rectal probe.

  • Variety of sleep and circadian rhythms of elderly dementia patients living in group home residences.

    English, Scientific journal, In the present study, we investigated 70 elderly individuals with dementia residing in a group home in Japan (88.1 ± 6.6 SD years; 12 males and 58 females). Sleep measurements were collected via an accelerometer placed under the mattress from March 2021 to January 2024. The accelerometer-derived activity counts were transformed and plotted as actograms. On the basis of the actograms, sleep patterns were categorized into three groups: regular nighttime sleep (Type 1, n = 38, 54.3%), regular nighttime sleep with daytime naps (Type 2, n = 29, 41.4%), and irregular sleep (Type 3, n = 3, 4.3%). We compared accelerometer-derived sleep measures between the Type 1 and Type 2 groups due to the small number of participants in Type 3. Interestingly, Type 2 individuals had a significantly longer bed in time and total sleep time. However, their sleep onset latency, time of wakefulness after sleep onset and sleep efficiency did not differ from those of Type 1 individuals. Additionally, we assessed the relationships between sleep patterns and dementia characteristics, such as independence in daily living and level of care needed. No significant differences were observed in sleep measurements across these characteristics. To evaluate endogenous circadian rhythms, seven dementia patients (Type 1, n = 1; Type 2, n = 6) wore a patch-type wearable temperature sensor for approximately 24 h to estimate core body temperature (CBT). Although the zero-amplitude test revealed a significant rhythmicity in all patients, the CBT rhythms showed lower amplitude and unstable phases. In summary, our findings indicate that dementia patients display varying sleep patterns that are independent of dementia characteristics. The dysfunction of sleep homeostasis and entrainment of circadian pacemakers might be due to the light environment in the group home residence or impairment of the circadian clock itself in dementia patients.
  • Two oscillatory components detected by forced splitting of the sleep-wake cycle in humans.

    English, Scientific journal, The sleep-wake cycle of human subjects was artificially split into two episodes by imposing an 8-h light and 4-h dark cycle (LD 8:4) twice a day for 7 days, which was followed by a 3-day free-running session. Sleep was permitted only in the dark period. The subjects in the ordinary group were exposed to ordinary light (ca. 500 lx) in the 8-h light period, and those in the bright light group to bright (ca. 5,000 lx) and ordinary light alternatively with bright light after the first dark period (2400-400). Split sleeps persisted in the free-running session with the major episode around the first dark period and the minor episode around the second dark period. By contrast, circadian melatonin rhythm in the free-running session significantly phase delayed in the ordinary light group, but phase advanced in the bright light group, keeping the melatonin rhythm unsplit. The length of nocturnal melatonin secretion (NMS) was significantly shortened in the bright light group. Interestingly, the falling phase of NMS advanced significantly further than the rising phase. Such a difference was not detected in the ordinary light group. Similar differences were observed in the body temperature rhythm. These findings indicated oscillatory mechanisms underlying split sleeps distinct from the circadian pacemaker and suggested an involvement of different circadian oscillators in the rising and falling phases of NMS, which is consistent with the dual oscillator model proposed for the circadian system of nocturnal rodents.NEW & NOTEWORTHY The present study demonstrated that human sleep was separated into two essentially identical components, which persisted under constant conditions, suggesting circadian oscillator underlying split-sleep episodes. The study also indicated differential light sensitivities in the rising and falling phases of circadian melatonin rhythm, indicating the involvement of two different oscillators. These results consisted of the evening and morning dual-oscillator hypothesis for the circadian pacemaker and the hierarchical model for the pacemaker and sleep-wake cycle.

    English, Scientific journal, Circadian rhythms and sleep-wake cycles were measured in volunteers staying singly in temporal isolation unit where they were exposed to artificial short and long light-dark (LD) cycles for 7 days. The long day consisted of 16-h light and 8-h dark (LD 16:8) and the short day consisted of 8-h light and 16-h dark (LD 8:16). During the light period, bright light of approximately 5,000 lux was given from the ceiling and during the dark period there was no illumination. Sleep was monitored by bed sensors, wrist actiwatch, and polysomnography (PSG) on the first and last nights of the schedule. Sleep length was significantly longer under LD 8:16 than under LD 16:8 and the sleep quality estimated by PSG was worse under LD 8:16 than under LD 16:8, which were comparable to natural seasonality in sleep. The circadian rhythm in plasma melatonin was measured in dim light (10 lux) before and after the LD exposures. The nocturnal melatonin secretion (NMS) was significantly longer after LD 8:16 than after LD 16:8 due to differential phase shifts of the rising and falling phases of NMS. After LD 8:16, the falling phase was much advanced than the rising phase, whereas after LD 16:8 the rising phase was much delayed than the falling phase, resulting in the NMS compression. These results indicate that the light sensitivity in terms of phase shifting is different in the two circadian phases, supporting a dual oscillator hypothesis with different phase-response curves for light in the human circadian system.NEW & NOTEWORTHY The present study demonstrated differential light responsiveness of the rising and falling phases of nocturnal melatonin secretion in human subjects exposed to artificial long (LD 16:8) and short days (LD 8:16) and suggested the involvement of different oscillators under these phases. The findings well mimicked the seasonality of the circadian rhythms in nature and consisted with the evening/morning dual oscillator hypothesis proposed originally for nocturnal rodents, providing a new concept for the human circadian system.
  • Nonphotic entrainment of central and peripheral circadian clocks in mice by scheduled voluntary exercise under constant darkness.

    English, Scientific journal, In mammals, the central circadian pacemaker in the suprachiasmatic nucleus (SCN) entrains to an environmental light-dark (LD) cycle and organizes the temporal order of circadian rhythms in physiology and behavior. Previously, some studies have demonstrated that scheduled exercise could entrain the free-running behavior rhythm in nocturnal rodents. However, it remains unknown whether entrainment by scheduled exercise alters the internal temporal order of the behavioral circadian rhythms or clock gene expression in the SCN, extra-SCN brain regions, and peripheral organs when mice are entrained to the scheduled exercise under constant darkness (DD). In the present study, we examined circadian rhythms in locomotor activity and Per1-luc rhythms in the SCN, arcuate nucleus (ARC), liver, and skeletal muscle of mice entrained to an LD, mice free-running under DD and mice entrained to daily exposure to a new cage with a running wheel (NCRW) under DD. All mice showed a steady-state entrainment of behavioral circadian rhythms to NCRW exposure under DD in parallel with shortening the alpha when compared to that under DD. The temporal order of behavioral circadian rhythms and the Per1-luc rhythms in the SCN and peripheral tissues but not in the ARC were maintained in the mice entrained to the NCRW and LD cycles; in contrast, the temporal order was altered in the mice under DD. The present findings reveal that the SCN entrains to daily exercise, and daily exercise reorganizes the internal temporal order of behavioral circadian rhythms and clock gene expression in the SCN and peripheral tissues.
  • [Impacts of mastication frequency on circadian rhythm of glucose metabolism].

    Japanese, Scientific journal, Postprandial glucose concentration is dependent on the time of day and its concentration in the morning is lower than in the evening. The circadian rhythm of glucose metabolism is regulated by the central circadian pacemaker in the suprachiasmatic nucleus (SCN). Both the SCN circadian clock and the pancreatic clock play important role in generating and maintaining the circadian rhythm of glucose metabolism. Also, short sleep duration and circadian misalignment are closely associated with a decrease in insulin sensitivity and an increase in type2 diabetes. Increased frequency of mastication and/or thorough chewing has been reported to alter the secretion of hormones related to appetite and energy metabolism. Furthermore, we have reported that the effect of mastication on postprandial glucose metabolism is dependent on the time of day and frequency of mastication. Morning mastication but not evening decreases postprandial blood glucose concentrations and increases insulin secretion at 30 min and so-called the insulinogenic index as a marker of early-phase β-cell function. This novel finding may aid in reducing the incidence of obesity and type 2 diabetes mellitus. This review covers the basic concept of the mammalian human circadian system, the underlying mechanism causing phase adjustment of the circadian rhythms in the SCN and peripheral organs, and the effect of eating behavior (e.g., chewing frequency) on the circadian rhythm of glucose metabolism.
  • Time of Day of Vaccination Does Not Associate With SARS-CoV-2 Antibody Titer Following First Dose of mRNA COVID-19 Vaccine.

    English, The immune system exhibits circadian rhythms, and its response to viral infection is influenced by the circadian clock system. Previous studies have reported associations between the time of day of vaccination against COVID-19 and production of anti-SARS-CoV-2 antibody titer. We examined the effect of vaccination time of day on anti-SARS-CoV-2 antibody titer after the first dose of vaccination with the mRNA-1273 (Moderna) COVID-19 vaccine in an adult population. A total of 332 Japanese adults participated in the present study. All participants were not infected with SARS-CoV-2 and had already received the first dose of mRNA-1273 2 to 4 weeks prior to participating in the study. The participants were asked to provide basic demographic characteristics (age, sex, medical history, allergy, medication, and mean sleep duration), the number of days after the first dose of vaccination, and the time of day of vaccination. Blood was collected from the participants, and SARS-CoV-2 antibody titers were measured. Ordinary least square regression was used for assessing the relationship between basic demographic characteristics, number of days after vaccination, time of day of vaccination, and the log10-transformed normalized antibody titer. The least square mean of antibody titers was not associated with the vaccination time and sleep durations. The least square means of antibody titers was associated with age; the antibody titers decreased in people aged 50 to 59 years and 60 to 64 years. The present findings demonstrate that the vaccination time with mRNA-1273 was not associated with the SARS-CoV-2 antibody titer in an adult population, suggesting that these results do not support restricting vaccination to a particular time of day. The present findings may be useful in optimizing SARS-CoV-2 vaccination strategies.

    English, Scientific journal, Effects of a fixed single meal per day were examined on the circadian pacemaker and sleep-wake cycle in subjects under temporal isolation. When the time of single meal was allowed to take at any time of day (Ad-lib meal), the sleep-wake cycle as well as the circadian rhythms in plasma melatonin, cortisol and core body temperature were significantly phase-delayed in 8 days. On the other hand, when the time of meal was fixed at 1800 h in local time (RF meal), the phase-shift of sleep-wake cycle was not significant, while those of the circadian rhythms were significant. The differential effects of a fixed single meal schedule were confirmed in most individual subjects. There was no evidence for the prefeeding increase in plasma cortisol and leptin levels under the fixed single meal schedule. The plasma ghrelin level was apparently high before meal in both Ad-lib and RF meal groups, which was, however, likely sculptured by a non-specific prandial drop and gradual increase after meal intake. Single meal augmented the prandial increase of plasma insulin levels by 4-5 times. These findings indicate that a single meal at fixed time of day during the subjective day failed to prevent the human circadian pacemaker but prevented the sleep-wake cycle to free-run at least 8 days under temporal isolation, suggesting that meal time was a potent non-photic time cue for the human sleep-wake cycle.

    Background
    Lactococcus lactis subsp. cremoris (YRC3780), which is isolated from kefir, has been associated with anti-allergic effects in humans. However, it remains unknown whether daily intake of YRC3780 attenuates the response to psychological stress in humans in parallel with changes to the gut microbiome. We examined the fundamental role of YRC3780 in the gut microbiome, stress response, sleep, and mental health in humans.



    Methods
    Effects of daily intake of YRC3780 on the hypothalamic-pituitary-adrenal (HPA) axis response to acute psychological stress were investigated in a double-blind, placebo-controlled clinical trial involving 27 healthy young men (mean age and body mass index: 23.5 years and 21.5 kg/m2) who were randomly assigned to placebo (n = 13) or YRC3780 (n = 14) groups. The HPA axis response to acute psychological stress, the diurnal rhythm of HPA axis activity, and gut microbiome were assessed and compared between the two groups.



    Results
    The results showed that daily intake of YRC3780 significantly lowered morning salivary cortisol levels compared with placebo. In addition, salivary cortisol levels following a social stress test significantly decreased +40 min after beginning the TSST in the YRC3780-treated group compared to placebo. There were no significant differences between the two groups in terms of actigraphy-based sleep quality, but the subjective sleep quality and mental health were significantly improved in the YRC3780-treated group compared to placebo.



    Conclusions
    Our study suggests that daily intake of YRC3780 improves the HPA axis response to acute psychological stress, which might be associated with a decrease in morning cortisol levels.


  • Basic concepts and unique features of human circadian rhythms: implications for human health.

    English, Scientific journal, Most physiological functions and behaviors exhibit a robust approximately 24-hour rhythmicity (circadian rhythm) in the real world. These rhythms persist under constant conditions, but the period is slightly longer than 24 hours, suggesting that circadian rhythms are endogenously driven by an internal, self-sustained oscillator. In mammals, including humans, the central circadian pacemaker is located in the hypothalamic suprachiasmatic nucleus. The primary zeitgeber for this pacemaker is bright sunlight, but nonphotic time cues also affect circadian rhythms. The human circadian system uniquely exhibits spontaneous internal desynchronization between the sleep-wake cycle and core body temperature rhythm under constant conditions and partial entrainment of the sleep-wake cycle in response to nonphotic time cues. Experimental and clinical studies of human circadian rhythms must take into account these unique features. This review covers the basic concepts and unique features of the human circadian system, the mechanisms underlying phase adjustment of the circadian rhythms by light and nonphotic time cues (eg, physical exercise), and the effects of eating behavior (eg, chewing frequency) on the circadian rhythm of glucose metabolism.

    睡眠・覚醒や体温調節など、ほぼすべての生理機能には24時間を1サイクルとする“概日リズム”が存在し、“概日時計”がその時間的調節を担う。我々の身体を構成する すべての細胞に、概日時計は備わり、時計遺伝子の転写・翻訳を介したフィードバックループがその中心的役割を示す。概日時計の中でも、中枢時計である「視交叉上核」はペースメーカーとして機能し、睡眠・覚醒のリズムを調節する。


    近年の光遺伝学の技術開発により、様々な生命現象を調節する神経回路が同定されてきている。哺乳類の睡眠研究においても、実験室の飼育環境下における睡眠・覚醒調節に関わる神経細胞が同定されてきた。一方、自然界における動物の睡眠研究はほとんどなされていない。しかし、最近になって自然界における睡眠・覚醒パターンが、実験室の結果とは大きく異なる事が示唆されてきた。国際生物学賞を受賞したDr. S. Daan(故)は、自然条件下(野外)でマウスを飼育すると、夜行性から昼行性にシフトする事を発見した。本研究では、自然環境下における動物行動の調節を担う神経メカニズムの解明を行う。

  • Mechanism of exercise effect on the central circadian pacemaker

    本研究では雄性のC57BL/6-Tg(Period1-luc)マウスを用いて、マウスの自発活動リズムが①LD12:12明暗サイクルに同調、②恒常暗下でフリーラン、③恒常暗下で1日3時間の回転輪のついた新しいケージに移す新奇環境暴露スケジュールに同調、の3条件下で視交叉上核、弓状核、肝臓、骨格筋を採取し、生物発光測定装置を用いてPer1-lucリズムを測定した。各組織における培養翌日のPer1-lucリズムのピーク位相をリズム指標として、行動リズムの活動開始・終了位相および中枢・末梢時計間の位相関係を解析した。その結果、明暗サイクル同調時と新奇環境暴露スケジュール同調時におけるSCNのPer1-lucリズムのピーク位相はいずれも主観的明期(マウスの休息期)にみられた。肝臓および骨格筋のピーク位相は、主観的暗期(マウスの活動期)にみられた。行動リズム位相とSCN、肝臓、骨格筋のPer1-lucリズムの位相関係は明暗サイクル下と新規環境暴露スケジュール同調時で有意な差は認められなかった。一方、弓状核のPer1-lucリズム、明暗サイクル下および恒常暗でのフリーラン条件下では主観的暗期にピーク位相がみられたが、新奇環境暴露同調時に明暗サイクルおよび恒常暗条件に比べ、有意に位相前進し主観的明期にピーク位相がみられた。本研究により、恒常暗下での習慣的な運動スケジュールがマウスSCNの同調因子として作用し、明暗サイクル同調時におけるSCNと行動リズム、肝臓、骨格筋の末梢時計間の位相関係を維持した状態で同調することを明らかにした。

    Brain microdialysis is a powerful technique for measuring a neurotransmitter in extracellular fluid from free moving animals. By using this technique with some modification for measuring a large molecule (neuropeptide), we examined circadian rhythm of neuropeptide released from a central circadian pacemaker of suprachiasmatic nucleus in free moving mice. The mice were placed under a normal light-dark cycle and constant darkness conditions for 2-4 days. During this period, artificial cerebrospinal fluid was perfused at a flow rate of 1 μl/min and the dialysate were collected every 1 h. As a result, circadian rhythm of AVP in dialysate was detected in some mice that the dialysis probe was inserted close to the SCN. The highest value was observed in early subjective day (ZT4). However, in other mice, circadian rhythms of AVP in dialysate were not detected. Technical improvement would be needed in future studies.

    Appearance of premeal anticipatory hormone peak and entrainability of restricted meal schedule were examined in humans who stayed in a temporal isolation facility without knowing the time of day. Meal was supplied once a day for 10 days. As a result, the premeal peak was not detected in plasma melatonin, cortisol, insulin, ghrelin, and leptin. On the other hand, the sleep-wake cycle was entrained by the meal schedule, whereas the circadian melatonin rhythm was not entrained showing phase-delay shifts. These findings indicated that the so-called food entrainable circadian rhythm failed to appear in humans under the present conditions, and that the meal schedule was the time cue for the sleep-wake cycle but not for the circadian pacemaker.

    (研究の目的)本研究は、ヒト生物時計に対する高照度光と身体運動の相互作用を時間隔離実験室を使用し、行動(睡眠覚醒)、ホルモン(メラトニン)、深部体温のリズムを測定し検証する。(実施計画)本研究は、健康な20代の成人男性を対象に時間隔離実験室を使用して行う。被験者は、時間隔離実験室内で12泊13日生活する。実験は、通常のリズム位相を測定するベースライン期間(3日)、通常の睡眠時間帯を8時間位相前進する強制スケジュール期間(4日)、そして、睡眠時間や食事のタイミングを制限しないフリーラン期間(6日)、3つのセッションからなる。ベースラインおよび強制スケジュール期間中は8時間睡眠、16時間覚醒とし、覚醒時の実験室内の照度は約3000ルクスの高照度光とし、睡眠中は約0ルクスである。メラトニンリズムを計測するための採血時およびフリーラン期間中は約10ルクスの低照度に設定する。(研究実績)平成23年度の研究計画に従い、3回の隔離実験を行い、対照群4名、運動群4名より以下の結果を得た。[睡眠覚醒リズム]対照群(4名中3名)、運動群(4名)ともにフリーラン第1日目の睡眠開始位相は8時間前進した就寝時刻付近にみられ睡眠覚醒リズムは身体運動を負荷しなくとも高照度光のみで再同調が完了していた。[メラトニリズム]対照群、運動群ともに8時間前進スケジュール最終日ではベースライン期間から5~6時間の位相前進が認められた。メラトニンリズムの位相前進量は、対照群と運動群間では同程度であり、運動によるメラトニンリズムの再同調促進効果は認められなかった。

  • Synchronization effects of physical exercise on human peripheral clocks regulating behavior rhythms

    To clarify whether physical exercise is a time cue for the human circadian clock and the mechanism by which physical exercise entrains the clock, we performed a series of experiment under temporal isolation using an 8h phase-advanced desynchrony protocol. Adult female subjects were required to perfome physical exercise of 2 hour duration twice a day using bicycle ergometer. The circadian rhythms in plasma melatonin and deep body temperature, which are believed to be regulated by the central circadian pacemaker, were measured simultaneously with the sleep-wake cycle which is thought to be driven by the peripheral clock. As a result, the re-entrainment of the sleep-wake cycle to a 8 h phase-advanced schedule was accelerated by timed physical exercise under dim light of ca. 10 lx, whereas the plasma melatonin did not phase-advance regardless of timed physical exercise. On the other hand, the re-entrainment of the sleep-wake cycle was accomplished regardless of timed physical exercise under bright light of ca. 5,000 lx, whereas the plasma melatonin rhythm phase-advanced but did not fully entrain but phase-advanced with or without timed physical exercise. The findings indicate that time physical exercise has a potency to entrain the sleep-wake cycle, but the effect is masked under bright light conditions.

  • Acceleration mechanism of re-entrainment of circadian rhythms by physical exercise in mammals

    The aim of present study was to clarify acceleration mechanism of re-entrainment of circadian behavioral and clock gene expression rhythm in peripheral tissues (peripheral clocks) to a large shift of light-dark cycle (e.g. jet-lag travel in humans) by physical exercise in mammals. We could demonstrate that the effect of physical exercise, such as exposure to new cage with a running-wheel, on the re-entrainment of peripheral clock was dependent on the timing of exposure. During the transient days in behavior after shift of light cycle, circadian clock gene expression rhythm in the central clock in the suprachiasmatic nucleus (SCN) showed a regional difference in the waveforms and phases. These finding addresses a new hypothesis that the scheduled physical exercise directly affect on the regional oscillator in the SCN. If so, it leads to increase a coupling strength between the SCN and peripheral clock and to accelerate re-entrainment of peripheral clocks by physical exercise.

    Oscillating neurons in the suprachiasmatic nucleus (SCN), the center of biological clock, constructs more than one regional pacemakers of different characteristics through mutual synchronization. The roles of regional pacemakers depend on the area of SCN, and their interaction determines the length of activity period, showing seasonal changes in behavioral rhythms. The function of biological clock is disrupted by restricted daily feeding or methamphetamine treatment, which is supposed to be due to desynchronization between the central and peripheral clocks. The peripheral clock(s) regulating behavioral rhythms is suggested to reside in the midbrain.
