    英語, 研究論文(学術雑誌), Neonicotinoids (NNs) are commonly used insecticides; however, their biotransformation in wild mammals remains poorly understood. Asian elephants, which are declining in numbers, may be vulnerable to these chemicals. In this study, we examined the liver metabolism of four major NNs: acetamiprid, imidacloprid, thiamethoxam, and clothianidin, using elephant liver microsomes. Metabolite profiling and targeted analysis were conducted with liquid chromatography quadrupole time-of-flight mass spectrometry and liquid chromatography tandem mass spectrometer, respectively. Results indicated that elephants mainly metabolize these NNs via reductive pathways, suggesting the involvement of nitroreductase-like enzymes. Conversely, cytochrome P450-mediated oxidation may be limited; however, the roles of other enzymatic or microbial systems cannot be ruled out. These findings provide insights into xenobiotic metabolism in elephants, helping evaluate their toxicological vulnerability and supporting conservation efforts and risk assessments.
  • Intergenerational sex-specific transcriptomic alterations in gonads of mice after parental lead exposure.

    英語, 研究論文(学術雑誌), Lead (Pb) exposure is known to influence gene expression across multiple tissues, potentially affecting germline biology and offspring development. This study investigated transcriptomic alterations in the gonads of F1 mice following four weeks of parental exposure to Pb‑contaminated soil. After exposure, breeding generated four experimental groups: ① exposed dams with unexposed sires, ② exposed sires with unexposed dams, ③both parents exposed, and ④ unexposed controls. Testes and ovaries were collected from 9‑week‑old offspring, and RNA sequencing was performed using Qiagen CLC Genomics Workbench to characterise transcriptional changes. Principal component analysis revealed distinct transcriptomic profiles in Groups 1-3 relative to controls, indicating that parental Pb exposure, whether maternal, paternal, or combined, shaped offspring gonadal gene expression. Heatmap clustering showed partial overlap between Male Group 3 and controls, although unique expression features remained evident. All exposed groups exhibited reduced overall transcript abundance when compared to controls. Males with maternal Pb exposure displayed the highest number of differentially expressed genes (DEGs), with gene ontology analysis highlighting alterations in immune‑related and enzymatic pathways. In contrast, males with both maternal and paternal exposure showed the fewest DEGs, though a greater proportion were located on the Y chromosome. These sex‑specific transcriptional patterns, particularly those observed in the offspring of exposed sires, suggest that Pb‑related molecular effects may arise independently of direct maternal transfer. While the underlying mechanisms remain uncertain, the findings raise the possibility that some effects could be transmitted through heritable pathways.

    英語, 研究論文(学術雑誌), The impact of various human activities on protected environments remains a significant concern. Mining for ores is crucial in many aspects of society. To achieve global decarbonisation, more metals are required, which will lead to increased worldwide metal exploration and mining. Zambia is primarily a mining country and has expanded exploration into protected lands (national parks). The effects of this exploration on these protected areas is ongoing. This study aimed to establish baseline data for monitoring metal pollution in the Lower Zambezi National Park before mining commenced. It also evaluated whether tourism influenced metal pollution in national parks and how these metals and metalloids correlate within tree bark and soils. To accomplish this, soil and Acacia tree bark samples were collected from Lower Zambezi National Park and Mosi-Oa-Tunya National Park. Arsenic (As), cadmium (Cd), chromium (Cr), cobalt (Co), copper (Cu), mercury (Hg), manganese (Mn), nickel (Ni), lead (Pb), and zinc (Zn) were analysed in this study. The concentrations of metals and metalloids were higher in areas with vehicular traffic and mining activity in the national parks, which may directly impact the biosystems in those areas. Maximum levels of metals in soil in these areas were as follows: As 40.8 mg/kg, Cd 0.879 mg/kg, Cr 241 mg/kg, Hg 1.1 mg/kg, Pb 211 mg/kg, and Zn 1285 mg/kg. When compared to areas of lower anthropogenic activities, differences in metal concentrations were up to 1000-fold. These changes can potentially affect the adaptation of wildlife to various stressors, especially as we are facing global climate change.
  • A Comparative Genomic and Phylogenetic Investigation of the Xenobiotic Metabolism Enzymes of Cytochrome P450 in Elephants Shows Loss in CYP2E and CYP4A.

    英語, 研究論文(学術雑誌), Cytochrome P450 is an important enzyme that metabolizes a variety of chemicals, including exogenous substances, such as drugs and environmental chemicals, and endogenous substances, such as steroids, fatty acids, and cholesterol. Some CYPs show interspecific differences in terms of genetic variation. As little is known about the mechanisms of elephant metabolism, we carried out a comparative genomic and phylogenetic analysis of CYP in elephants. Our results suggest that elephant CYP genes have undergone independent duplication, particularly in the CYP2A, CYP2C, and CYP3A genes, a unique cluster specific to elephant species. However, while CYP2E and CYP4A were conserved in other Afrotheria taxa, their decay in elephants resulted in genetic dysfunction (pseudogene). These findings outline several remarkable characteristics of elephant CYP1-4 genes and provide new insights into elephant xenobiotic metabolism. Further functional investigations are necessary to characterize elephant CYP, including expression patterns and interactions with drugs and sensitivities to other chemicals.

    英語, 研究論文(学術雑誌), Anticoagulant chemicals (ACCs) such as warfarin are widely used in medical applications as well as for their rodenticide properties. Their efficacy is greatly influenced by polymorphisms in the gene encoding vitamin K epoxide reductase (VKOR). Evaluation of the activity of ACCs toward VKOR variants is essential to determine their proper use. Presently, this is achieved by co-expressing VKOR of Rattus Norvegicus and human clotting factor IX in cultured cells and measuring inhibition of vitamin K-dependent gamma-glutamyl carboxylation of factor IX (glaFIX) activity. However, glaFIX has only been quantified using indirect methods like blood coagulation assays. We have developed a sandwich enzyme-linked immunosorbent assay using a glaFIX-specific antibody to quantify glaFIX and used this to analyze inhibition of VKOR activity by warfarin.

    本研究ではゾウの代謝機構に関する理解を深めるために、二つの観点に着目して調査を進めている。第一に、未解明の異物代謝酵素およびその機能の解明を目指し、ゾウに特異的な酵素群の発現と活性を明らかにすることで、新たな代謝経路の存在を検討している。第二に、腸内細菌叢が異物代謝に果たす役割に着目し、宿主の代謝機構と腸内微生物叢の相互作用を含めた包括的な代謝システムの理解を試みている。
    2023年度からは、タイの研究機関と連携し、死亡したゾウの個体から肝臓、腎臓、腸管などの新鮮な臓器を収集する体制を確立した。これらの臓器サンプルを用いてRNAの抽出および酵素活性測定を実施し、代謝酵素の発現解析と代謝活性試験を進めている。その結果、これまでin silico解析により予測されていた仮説を実証する新たなデータが得られつつあり、ゾウの代謝機構に関する理解が着実に進展している。
    また、ザンビア・ローワーザンベジ国立公園において2022年度から2023年度にかけて採取したアフリカゾウの糞便サンプルを用いた研究では、環境中の化学物質への曝露評価を目的に、農薬(殺虫剤・除草剤)および重金属の濃度分析を行った。分析にはGC-MS/MS、LC-MS/MS、LC-QTOF/MS、ICP-MSといった質量分析機器を用いた。加えて、同地域の植物および土壌サンプルにも同様の分析を行った結果、植物と糞便の両方から共通の農薬類が検出され、同地域におけるゾウが植物を介して農薬に曝露している可能性が示唆された。さらに、一部の重金属に関しては、採取年度や地点によって濃度に有意な差が見られ、人為的影響の強い地域において高濃度を示す傾向が明らかとなった。
    現在は、これらの化学物質がゾウの体内でどのように蓄積され、どのように排泄されるか、さらにそれらがゾウの健康に及ぼす影響について、より詳細な検討を進めている。
