English, Scientific journal, Localized plasmonic heating by metallic nanoparticles offers a promising strategy to destroy cancer cells through controlled thermal stress. However, how cells sense and respond to microscale temperature variations within complex tissue environments remains unclear. Here, we investigate how plasmon-induced local heating reshapes intercellular signaling and cell fate within tumor spheroids, focusing on the extracellular signal-regulated kinase (ERK) pathway. HeLa spheroids expressing a FRET-based ERK biosensor were subjected to defined photothermal stimuli using gold nanostars as nanoscale heat sources, while ERK activity was tracked with a deep-learning algorithm (3DeeCellTracker). Local heating triggered marked alterations in ERK signaling dynamics compared to spontaneous activity, including changes in activation frequency, timing, and duration. Remarkably, heat-induced ERK activation spread across neighboring cells, revealing thermally mediated intercellular propagation. Quantitative analysis further showed that temperature elevation modulates cell death and division in a power-dependent manner. Near-infrared light activation enhances phototherapeutic efficacy due to deep-tissue penetration and minimal invasion. While near-infrared-phototherapeutic agents are widely studied, conventional designs often increase molecular size, affecting pharmacokinetics. This study elucidates the photo-excited states of sulfone-containing aromatic compounds as efficient near-infrared-activated photothermal agents for cancer therapy. We synthesized sulfone-rosamine compounds (3 and 4) exhibiting strong optical absorption around 700 nm, attributed to a unique orbital in the LUMO. Compound 4, 35% smaller than conventional near-infrared-dye molecules, such as Cy5.5 (0.86 nm3/molecule), enhances intracellular mobility. Upon photoexcitation, 4 undergoes rapid non-radiative relaxation, resulting in remarkable photothermal effects and photostability. Transient absorption spectroscopy and density functional theory calculations revealed a mechanism involving rapid non-radiative deactivation after illumination, efficiently converting light to heat. The near-infrared-induced photothermal effect effectively killed various cancer cell lines, including HeLa, MDA-MB-231, and PCI-55. This study provides guidelines for designing novel photodrugs with improved intracellular mobility, near-infrared light responsiveness, and photostability, and introduces the advancement of cancer treatment. - Carbon nanohorn coating on titanium promotes M2 macrophage polarization

English, Scientific journal, This study investigated the effects of carbon nanohorns (CNHs) coated on titanium surfaces on macrophage responses. The CNHs were electrodeposited on titanium discs to create a uniform coating with a thickness of 0.2-0.3 µm. Analysis of macrophages cultured on CNH-modified titanium (CNH/Ti) revealed decreased production of pro-inflammatory cytokines (TNFα and IL6) and increased IL10 gene expression compared to unmodified titanium. Immunofluorescence analysis showed higher expression of the M2 marker CD206 and lower expression of the M1 marker CD86 on the CNH/Ti surfaces. Gene expression analysis indicated downregulation of genes involved in DNA transcription, repair, and replication, correlating with controlled cell proliferation on CNH/Ti. These findings demonstrate that CNH modification on titanium surfaces influences macrophage cytokine production and phenotype marker expression, suggesting its potential application in dental implant development. - Near-infrared light-boosted antimicrobial activity of minocycline/hyaluronan/carbon nanohorn composite toward peri-implantitis treatments

English, Scientific journal, Luminescent nanomaterials such as semiconductor nanocrystals (NCs) and quantum dots (QDs) attract much attention to optical detectors, LEDs, photovoltaics, displays, biosensing, and bioimaging. These materials include metal chalcogenide QDs and metal halide perovskite NCs. Since the introduction of cadmium chalcogenide QDs to biolabeling and bioimaging, various metal nanoparticles (NPs), atomically precise metal nanoclusters, carbon QDs, graphene QDs, silicon QDs, and other chalcogenide QDs have been infiltrating the nano-bio interface as imaging and therapeutic agents. Nanobioconjugates prepared from luminescent QDs form a new class of imaging probes for cellular and in vivo imaging with single-molecule, super-resolution, and 3D resolutions. Surface modified and bioconjugated core-only and core-shell QDs of metal chalcogenides (MX; M = Cd/Pb/Hg/Ag, and X = S/Se/Te,), binary metal chalcogenides (MInX2; M = Cu/Ag, and X = S/Se/Te), indium compounds (InAs and InP), metal NPs (Ag, Au, and Pt), pure or mixed precision nanoclusters (Ag, Au, Pt), carbon nanomaterials (graphene QDs, graphene nanosheets, carbon NPs, and nanodiamond), silica NPs, silicon QDs, etc. have become prevalent in biosensing, bioimaging, and phototherapy. While heavy metal-based QDs are limited to in vitro bioanalysis or clinical testing due to their potential metal ion-induced toxicity, carbon (nanodiamond and graphene) and silicon QDs, gold and silica nanoparticles, and metal nanoclusters continue their in vivo voyage towards clinical imaging and therapeutic applications. This review summarizes the synthesis, chemical modifications, optical properties, and bioimaging applications of semiconductor QDs with particular references to metal chalcogenide QDs and bimetallic chalcogenide QDs. English, Scientific journal, Photodynamic therapy (PDT) is a cancer therapy that uses a photosensitizer (PS) in the presence of oxygen molecules. Since singlet oxygen is highly reactive, it is important to deliver it to the target site. Thus, an efficient drug delivery system (DDS) is essential for enhancing the efficacy of such a treatment and protecting against the side effects of PDT. Here, we report on attempts to increase the therapeutic effect of PDT by using a DDS, a lipid nanoparticle (LNP). We prepared a porphyrin analog, rTPA (PS) that was encapsulated in LNPs using a microfluidic device. The findings indicated that the internal structure of the prepared particles changed depending on the amount of rTPA in LNPs. The photoactivity and cell-killing effect of PS in LNPs also changed when the amount of the cargo increased. These results suggest that the internal structure of LNPs is important factors that affect drug efficacy. This article is protected by copyright. English, Scientific journal, As structure defined cutouts of the graphene lattice, nanographene molecules have gained plenty of attention because of their high potential for versatile applications in organic electronics and energy conversion devices and as ideal model systems for the better understanding of intrinsic structure-property correlations of graphenes. In this study, well-defined nanographenes with sp2 carbon networks of different sizes, hexa-peri-hexabenzocoronene (HBC) and its rectangularly π-extended version, a short graphene nanoribbon (GNR), have been covalently functionalized with photoactive porphyrin molecules. On the basis of their spectroscopic studies, the photodynamics of the porphyrin-linked nanographenes was found to be influenced substantially by the size of the nanographenes. Photoexcitation of the porphyrin-HBC linked system led to exclusive energy transfer (EnT) from the first singlet excited state (S1) of the nanographene to the porphyrin, whereas opposite selective EnT occurred from the first and second singlet excited states (S1 and S2) of the porphyrin to the nanographene in the porphyrin-GNR linked system. In particular, ultrafast efficient EnTs from both the S2 and S1 states of the porphyrin to GNR mimic the corresponding ultrafast EnTs from the S2 and S1 states of carotenoids to chlorophylls in light-harvesting systems of natural photosynthesis. Japanese, Derivatives of Ce@C82 (C2v) have been synthesized and fully characterized, and their anisotropic magnetism has been observed as paramagnetic shifts in NMR measurements. Carbene addition by photochemical reaction afforded two isomers of Ce@C82(C2v)Ad (Ad = adamantylidene), 2a and 2b, demonstrating high regioselectivity. The two isomers were characterized using MALDI-TOF mass spectrometry, vis-NIR absorption spectroscopy, 1H and 13C NMR spectroscopy, and electrochemistry. The structure of the minor isomer (2b) was elucidated by single-crystal X-ray structural analysis. 13C and 1H NMR measurements revealed the characteristic anisotropic interaction between the f electron on the Ce atom and nuclear spins of the carbon atoms of the cage and the protons of the Ad group, respectively. - Radical Coupling Reaction of Paramagnetic Endohedral Metallofullerene La@C-82

Japanese, La@C82 is one of the endohedral metallofullerenes which have attracted special attention because they engender new spherical molecules with unique electronic properties. We have found that four isomers of benzyl mono-adducts, La@C82(CH2C6H5), are obtained by the radical coupling reaction in a toluene solution of La@C82. Thermolysis of 3-triphenylmethyloxazolidin-5-one (1) may generate benzyl radical by hydrogen abstraction from toluene, which affords the radical coupling product with La@C82. The same mono-adducts are also obtained by photoirradiation of La@C82 in toluene in the absence of 1. These reactions are applicable to paramagnetic metallofullerenes, such as La@C82 and Ce@C82. Photoirradiation of La@C82 in 1,2-dichlorobenzene in the presence of tetrachlorotoluene also affords the mono-adducts(3a-3d). Single-crystal X-ray structure analysis for 3d reveals its unique structure. Theoretical calculation confirms that the cage carbons having high spin densities are selectively attacked by radical species to form the singly bonded mono-adducts linked by a carbon-carbon single bond. - Nanorods of endohedral metallofullerene derivative

酵素分解を利用した革新的な微粒子生成法により、環境調和型メゾスコピックナノ粒子材料(ms粒子)を創製する手法の確立を目指します。特に、非可食性バイオマスや廃棄物由来の原料を用いて金属ナノ粒子集合体や分子触媒ナノ粒子などを開発し、新規なms粒子の機能発現を実現します。本手法で生成されるms粒子は酵素分解性の連結部位を有するため、使用後の分解・リサイクルが容易な環境調和型ナノ材料として期待できます。

本年度は、当初計画通り「(II):マイクロ組織内の分子動態解明を利用した高効果光がん治療薬の開発」および「(III):マイクロ組織を活用した光治療薬開発プロセスの一般化」を中心に研究遂行した。また、本研究の遂行から発明された新規ナノ粒子作成方法について「(I):3D培養系で高い浸透性・発光性・安定性をもつ機能化量子ドットの合成と培養組織内動態の解明」に関連して新規展開し、その理解の深化と基盤構築を並行して行った。
(II)に関して、各種光増感剤の改良・新規開発を行った。我々がこれまでも用いているπ拡張型ポルフィリン分子(rTPA)を利用した系の開発に成功した【成果論文2報】。ここではrTPAについて包括的な開発を行い、その液中挙動及び光励起状態での光増感剤としての性能評価をおこない、用途に応じて至適なrTPA誘導体を選択する基盤を確立した。この知見をもとに、ナノ薬剤やナノカーボンとの複合化による光治療薬向け化合物開発が加速した。
(III)に関して、歯科医療応用に向けた近赤外光治療向けの化合物開発を行った。カーボンナノホーンを担体としてrTPAと複合化することで、効果的な近赤外光増感作用を発現して殺菌効果を示す材料開発を達成した【成果論文1報】。
新規展開の(I)について、量子ドット凝集体を利用することでより長時間露光や環境変化に対しても安定に発光をつづける発光ラベルが得られることを見出した(特願2023- 79183、論文準備中)。この知見は、長時間観察が必要になることの多い3次元培養細胞系において、有用な発光ラベル調製法となりため、これを利用した薬物輸送キャリアのプロトタイプ開発も行った。これらは、3次元細胞系を基軸にした展開を行っていた本研究ゆえに実現した物質開発といえる。
以上のように、研究は申請書記載の当初計画案にそっておおむね順調に進行している。

本年度は、当初計画通り「(I):3D培養系で高い浸透性・発光性・安定性をもつ機能化量子ドットの合成と培養組織内動態の解明(研究1~2年目計画)」のために細胞標的指向性分子-量子ドット複合体の開発に着手した。量子ドットに対してポリエチレングリコール鎖を介して水酸基(中性)およびカルボン酸(アニオン性)を接合した量子ドットを合成した。また、量子ドットに対して癌細胞マーカーとして良く知られているCD44抗体およびEpCAM抗体を接合した複合体も作成し、血液モデルのPBMC中から癌細胞を選択的に癌細胞認識できることを確認した。当該量子ドット-抗体複合体は現在、膵がん細胞における取り込み挙動を、2次元培養および3次元培養にて検討中である。また、量子ドット自体の光安定性についても検討を行い、光照射強度に応じて表面エッチングが起きる一方、光強度の制御や量子ドット表面組成によって細胞毒性を抑え得ることを見出した。
また「(II):マイクロ組織内の分子動態解明を利用した高効果光がん治療薬の開発(研究2~3年目計画)」に関して、その前段階として光増感剤の開発検討を行った。我々がこれまでも研究に用いているπ拡張型ポルフィリン分子(rTPA)について、量子ドット表面への担持やナノカーボン表面への担持を行い、その光増感性能(=一重項酸素発生能)の検証を行った。その結果、量子ドット表面への直接不可は、分子凝集などが原因となり十分な一重項酸素発生能を発揮しない一方、ナノカーボンの1種であるカーボンナノホーンに担持したrTPAは十分な一重項酸素発生能と殺がん効果を発揮し得ることを見出した。

本年度は当初計画通り、「[段階-I] 捕食誘引物質の粗分離、観察、構造決定による同定」にとりかかり、膵がん細胞の死細胞を調製した上で捕食誘引物質の分離を開始した。まず、死細胞について大きく「アポトーシス様死細胞」と「ネクローシス様死細胞」に分けて調製し、それぞれ膵がん細胞より再構築したミクロ組織3次元と共培養しながらリアルタイム顕微鏡観察を行い、取り込まれる挙動の観察を行った。この際、コントロール条件として1μm(大体アポトーシス死細胞と同サイズ)のCOOH修飾蛍光ビーズを用いた。結果、COOH修飾蛍光ビーズでは細胞への取り込み、特に細胞集団が触手を伸ばすなどの能動的な取り込みは起きないことを確認した。そして「アポトーシス様死細胞」と「ネクローシス様死細胞」は両者とも取り込まれることを確認したが、ネクローシス様死細胞では取り込みがやや少ない傾向にあった。これはネクローシスでは死細胞構造がランダムであるとともに、細胞内容物が維持できないことが一因であると考えられた。よって、今後の検討は主にアポトーシス様死細胞で行うこととした。現在は、アポトーシス様死細胞の大量調製と、粗分離を開始したところである。
また、「[段階-II]捕食誘引物質を複合化した高薬効性の光がん治療薬の合成開発」の準備段階として、量子ドットに近赤外光殺がん分子(rTPA)を接合した複合体や、ドラッグデリバリキャリアとrTPAの複合化も行い、その一重項酸素発生能および光殺がん効果を確認した。本研究の遂行によってドラッグデリバリキャリアとrTPAの相性により、内容分子の機能性(本研究の場合は光殺がん性能)が変化することが分かった。今後、最適化を進め最大効率で殺がん効果を示す複合体プロトタイプを作成し、本研究の目的とするトロイの木馬型殺がん化合物を完成させる。

本研究の目的は,カーボンナノ物質の光線力学作用を用いて,近赤外光を照射した際に抗菌作用を発揮させ,インプラント周囲炎に応用することである.本年度は,さらに高い殺菌効果を得るため,光増感剤をカーボンナノホーン(CNHs)に修飾し,その効果を検証した.
π拡張型ポルフィリン分子(rTPA)は700nm近傍の光を高い効率で吸収し,約50%の高い量子収率で一重項酸素を発生させる.このrTPAをアミノ化したCNHsとの間でアミド結合形成し,rTPA-CNHを得た.CNH-rTPAの熱重量分析を行ったところ,CNH-rTPA中のCNHとrTPAの割合がおよそ1:1であることが示唆された.CNH-rTPAの吸収スペクトルを測定したところ,可視光領域から近赤外光領域に渡るCNHの特徴的な吸収と,仕込み量の比率に比例した712nm近傍に特徴的なrTPAの吸収帯が観測された.次に,一重項酸素発生能を評価したところ,CNHs:rTPA=1:1のものが一番大きな一重項酸素産生能を示したため,以後の実験ではこの条件で作成したCNH-rTPAを用いた.また,rTPAはPBSなどの高いイオン濃度溶液中に単独で存在すると凝集形成し一重項酸素が抑制されるところ,CNHと複合化することで十分な一重項酸素発生能を保持することも明らかとなった.
Streptococcus mutans,Aggregatibacter actinomycetemcomitanceの分散液にrTPA-CNHを添加して10分間730nmの光を照射したところ,どちらの菌も半分以上の菌数減少がみられた.また,細胞毒性を調べるために,線維芽細胞の培養実験を行った.細胞播種4時間後にCNH-rTPAを添加し,10分間光照射した結果,CNH-rTPA添加および光照射による細胞数の減少はみられなかった.

In this study, application of carbon nanomaterials (CNMs) for drug carrier and controlling release of drug, surface modification of Ti with CNMs, and production of CNMs based membrane were done to develop the intelligent carbon nanomaterials (i-CNMs) with high function capable of controlling its expression. Antibiotics was loaded to CNMs. It was able to control the expression of its function by photoirradiation. Also, acceleration of bone formation was observed around Ti by surface modification of Ti with CNMs. CNMs based membrane showed excellent shielding property and maintaining the space for GBR membrane. These results suggested the invention of i-CNMs and its possibility of application for biomaterials.

In this study, we found that i) the early stage of podocyte injury is accompanied by a decrease in glycosphingolipid GM3, ii) enhancement of GM3 expression via valproic acid has preventive and therapeutic effects on an appearance of proteinuria and glomerulosclerotic lesions, iii) it is effective in preventing F-actin bundle assembly collapse and normalizing nephrin phosphorylation function, iv) nephrin and GM3 behave with each other in cooperatively.
This suggests that nephrin, which constitutes the slit membrane, may be regulated by glycolipids around the transmembrane region of nephrin, resulting in the maintenance of healthy podocytes and normal slit membrane functions. Indeed, the study suggested the possibility that GM3, a glycosphingolipid expressed in podocytes, may also be a potential target for proteinuria therapy.

In this study, to establish new strategy on tissue and bone regeneration with the biological characteristics of CNHs, titanium was coated with CNHs with using electrodeposition. CNH/Ti was implanted in rat bone. Inflammatory cytokines and gene expression of macrophages cultured on CNH/Ti and Ti were analyzed and also fluorescent immunostaining was carried out.
CNH/Ti promoted bone formation in early stage.The amounts of TNFα and IL-6 on CNH / Ti were significantly lower than those on Ti, and there was no significant difference in IL-10.Gene Ontology analysis by microarray showed genes related to transcription, repair and replication of macrophage DNA in CNH / Ti were down-regulated compared to Ti. More macrophages with high intensity for CD206 was observed on CNH/T compared with Ti. Therefore, it was suggested that CNHs on the Ti surface regulate transcription, repair and replication of DNA of macrophage, and effect on M2 macrophage polarization.

In this study, novel functional molecules utilizing the photoinduced charge separation were developed for a fundamental technology to develope photocontrol methods of cellular activities. We investigated intracellular optical functionality mainly for amphipathic ferrocene-porphyrin-fullerene linked molecules and aryl-acridinium molecules.
As research outcomes, we succeeded in photoinduced depolarization of the cell membrane by high charge separation efficiency due to development of molecules with enhanced cell membrane fluidity. In addition, we succeeded in mitochondria selective molecular transport in living cells by utilization of drug transport carriers or optimization of molecular structure. The molecules demonstrated successful method for triggering photooxidation/reduction reactions in cells by irradiation with light.

Our comprehensive researches revealed that the linker structures in the porphyrin-nanocarbon covalent composites have a large impact on their optical and photophysical properties. Specifically, moderate length-spacers between donor and acceptor were found to be essential to produce a long-lived charge-separated state efficiently. Fast decay of photoinduced charge-transfer states (i.e., exciplex state) to the ground state was observed in not only simple donor-acceptor linked systems, but also more complex dye-semiconducting electrode interface for dye-sensitized solar cells and donor-acceptor mixed interface for organic thin film solar cells. Integrated views and interpretation on the undesirable unique phenomena have been developed to give us a universal guideline on efficient photoinduced charge separation.

本研究では自己組織化と光機能性分子/ナノ材料を組み合わせ、高効率エネルギー変換や新規生体機能を発現する次世代人工光合成を創成していくことを目指した。すなわち、ナノ領域での光機能性分子/ナノ材料、メゾ-マイクロ領域の自己組織化の鍵となる相互作用を理解し、制御することを目的とした。特に次のような学際的な手法により、特異な巨視的特性-機能を発現させることを行った。(1) 種々の分子/ナノ材料を階層的に自己組織化するための構造ー機能相関の学理を解明することを目指した。特にフラーレン・カーボンナノチューブ・グラフェンを半導体電極上に同時に自己組織化し、光電変換特性を発現することに成功した。 (2) ポルフィリン、共役ポリマー、フラーレン誘導体などの光機能性分子/ナノ材料を合理的に電極上に自己組織化させることで高効率太陽光エネルギー変換を実現することを目指した。特に 浸漬溶媒を変化することで酸化チタン上の色素分子の吸着状態を変化させ、結果として色素増感太陽電池性能を制御できることを見いだした。一方、有機薄膜太陽電池では共役ポリマーの主鎖構造にハロゲン原子を導入することでフラーレン誘導体との混合膜状態を変化させ、太陽電池特性を制御することに成功した。(3) 光電荷分離分子と生きた細胞を自己組織化を通じて融合することで細胞機能を光で制御することを行った。具体的には光電荷分離分子を生きた細胞膜に導入し光照射を行うと、細胞膜電位の脱分極が起こること、また光電荷分離寿命と脱分極の大きさに相関があることを見いだした。

To establish methodology for light-driven water splitting, we have focused on the creation of highly efficient water splitting catalyst driven by light as well as the fabrication of the corresponding photoelectrochemical cell for material conversion. First, we synthesized a series of porphyrins with high oxidizing power for extracting multi-electron from the water splitting catalyst with water. Second, ruthenium complexes as the water splitting catalyst were prepared. Finally, they were assembled onto a semiconducting electrode, which was found to exhibit the capability of light-driven water splitting.

Cell membrane potential plays a crucial role in cellular functions in various cells such as nerve cells. Therefore, artificial control on the membrane potential is promising technology for cellular engineering and medical applications. For the control, we have studied on organic molecules which can generate charge separated state by irradiation in cellular environment. As a fruit, we found a molecular system which display efficient charge separation (quantum yield = 41%). In a series of study revealed that the molecules can induce depolarization in membrane of PC12 and rat hippocampal neurons. In addition, the degrees parallel the charge separation efficiency.
