    Scientific journal, Pulpotomy is performed when tooth decay reaches the dental pulp or when the crown is fractured due to trauma. Mineral trioxide aggregate (MTA) is commonly used in pulpotomy, but its prognosis can be variable. N-acetyl-L-cysteine (NAC), an antioxidant amino acid, has garnered attention due to its potential benefits. This study aimed to investigate the effects of MTA and NAC on pulpotomy outcomes. We used Sprague Dawley rat maxillary molars to perform pulpotomy and employed Superbond C&B, MTA, and MTA mixed with NAC (MTA–NAC) for pulp capping. We obtained tissue sections 3 and 7 days postpulpotomy, conducting histological analysis by examining the morphology of pulp tissue and assessing dentin sialophosphoprotein (DSPP) and osteopontin expression levels. At 3 days postpulpotomy, MTA and MTA–NAC reduced the inflammatory response. At 7 days postpulpotomy, dentin bridge formation was observed following MTA–NAC application, and although MTA resulted in DSPP- and osteopontin-positive areas, these areas were more extensive following MTA–NAC application. Given that adding NAC to MTA enhanced dentin bridge formation, MTA–NAC appears to be a superior option for pulp capping.

    English, Scientific journal, BACKGROUND: A brain abscess is a focal infection in which abscesses form in the brain. A brain abscess is a rare but fatal disease when rupture occurs into the ventricles. We report a case of multiple brain abscesses caused by a hematogenous infection from the apical periodontitis of deciduous teeth. CASE PRESENTATION: The patient was a 7-years and 8-months-old male with congenital heart disease. The patient sought medical attention due to fever and headache, for which he was started on three antibiotics with a diagnosis of multiple brain abscesses. Given that apical periodontitis of deciduous teeth was strongly suspected as the source of the brain abscess, the deciduous teeth were extracted. Immediately after deciduous teeth extraction, the patient's headache and neurological symptoms disappeared. CONCLUSIONS: After teeth extraction, a clear shrinkage of the brain abscess was observed, and the patient was discharged from the hospital.

    The surface topography of a material can influence osteoclast activity. However, the surface structural factors that promote osteoclast activity have not yet been investigated in detail. Therefore, we investigated osteoclastogenesis by testing various defined patterns with different dimensions and shapes. The systematic patterns, made of a cyclo-olefin polymer, were prepared at a micron-, submicron-, and nano-scale with a groove, hole, or pillar shape with a 1:1 pitch ratio. RAW264.7 cells were cultured on these patterns in the presence of the receptor activator of NF-κB ligand (RANKL). Osteoclast formation was induced in the order: pillar > groove ≥ hole. The two-dimensional factors also indicated that submicron-sized patterns strongly induced osteoclast formation. The optimal pillar dimension for osteoclast formation was 500 nm in diameter and 2 µm in height. Furthermore, we observed two types of characteristic actin structure, i.e., belt-like structures with small hollow circles and isolated ring-like structures, which formed on or around the pillars depending on size and height. Furthermore, resorption pits were observed mainly on the top of calcium phosphate-coated pillars. Thus, osteoclasts prefer convex shapes, such as pillars for differentiation and resorption. Our results indicate that osteoclastogenesis can be controlled by designing surfaces with specific morphologies.
  • Anti-RANKL Inhibits Thymic Function and Causes DRONJ in Mice

Denosumab, a human monoclonal antibody against receptor activator of nuclear factor-kappa B ligand (RANKL), is a novel bone antiresorptive agent used in patients with osteoporosis or metastatic bone cancer. Denosumab-related osteonecrosis of the jaw (DRONJ) has been recently reported in patients using denosumab. However, the mechanisms of DRONJ are not fully understood. Appropriate pathogenic mechanisms of DRONJ have yet to be established. Therefore, we investigated the pathogenesis of DRONJ in mice. Methods. Anti-mouse RANKL monoclonal antibody and melphalan were performed to create a mouse model of DRONJ-like lesions in female C57BL/6J mice. We examined the development of DRONJ-like lesions and immune function. Results. We showed that administration of anti-mouse RANKL monoclonal antibody and melphalan caused DRONJ-like lesions that recapitulated major clinical manifestations of the human disease, including the characteristic features of an open alveolar socket and exposed necrotic bone. In the analysis using a mouse model of DRONJ-like lesion, it was revealed that anti-mouse RANKL monoclonal antibody and melphalan suppress autoimmune regulator (AIRE) expression in the thymus and imbalanced T cell populations. Conclusion. This study suggests evidence of an immunity-based mechanism of DRONJ-like disease. This work may contribute to a better understanding of the pathogenesis of human DRONJ.

    English, Scientific journal, This study examined the effects of N-acetylcysteine (NAC) on the inflammatory reactions of murine osteoblastic cells cultured on the 4-methacryloxyethyl trimellitate anhydride/methyl methacrylate (4-META/MMA)-based resin. Superbond C&B (SB) was used as the 4-META/MMA-based resin and placed in a 48-well cell culture plate. The cells were cultured in αMEM (control) as well as on SB and SB in αMEM with NAC (SB+NAC). They were examined using the WST-1 proliferation assay, real-time PCR, enzyme-linked immunosorbent assay (ELISA), intracellular reactive oxygen species (ROS) measurements, and cellular glutathione (GSH) detection. COX-2 and IL-6 gene expressions were upregulated in SB; however, they were suppressed by NAC. Furthermore, PGE2 production in the culture medium was increased in SB, whereas NAC decreased the PGE2 production. NAC lowered the ROS level in the culture medium and significantly increased the intracellular GSH level. The present in vitro study demonstrated that NAC might be effective for dental material detoxification.

    Japanese, Scientific journal, 各臓器由来のアルカリ性ホスファターゼ(ALP)の生体における真の基質と機能に関して未だに不明な点が多い.そこで,ヒトの骨型,小腸型,胎盤型及び肝臓由来のALPを購入して使用し,それらの基質選択性及び阻害剤に対する感受性を検索した.基質として,生体内には存在しないがALP活性の測定に汎用されるパラニトロフェニルリン酸(p -NPP)と,生体内に存在するナトリウムピロリン酸(Na-PPi),アデノシン三リン酸(ATP)及びピリドキサルリン酸(PLP)を使用して,各ALP活性のpH依存性と基質濃度依存性、及び阻害剤であるlevamisole及びvanadateによる阻害を検討した.活性は各基質が加水分解されて生ずる無機リンをChifflet法によって定量して測定した.骨型ALPの生体内基質とされているピロリン酸分解の至適pHはどのALPも8.8から9.1であり,また50%活性化濃度(K0.5)もALPの種類によらず3から5 mMであった.またピロリン酸を基質とした際の阻害剤の50%活性阻害濃度(Ki0.5)も類似しており,ALPの種類による違いは見いだせなかった.小腸型ALPの基質とされるPLP分解の至適pHはどのALPも9.9から10.4であり,またPLP分解のK0.5もALPの種類によらず1.1から1.6 mMであった. PLPを基質とした際の阻害剤のKi0.5は,vanadateではALPの種類によらず類似していたが,levamisoleでは異なっていた.ATPあるいはp -NPPを基質として同様の実験を行った結果,基質に対する至適pHと基質に対するK0.5はALPの由来には関係なく基質によって類似していた.また,levamisoleなどの阻害剤に対するKi0.5は基質の種類には関係なく,ALPの種類によってほぼ一定であった.ヒトに存在する各臓器由来のALPはその機能と生体内基質に対応して変化してきた可能性があると考えて本研究を行ったが,少なくとも今回検討した基質に関してはALPの種類による相違は見いだせず,むしろ阻害剤に対する感受性にALPの種類による相違が見られた.The human alkaline phosphatase (ALP), is homologous to not only other mammal ALP but also bacterial ALP. However, the substrate and function of ALP in vivo have not been cleared. Recently, we obtained human ALP as a chemical agent. In this study, we examined the differences in substrate selectivity and inhibitor sensitivity among several types of human ALP. We used 4 types of human ALP from bone, intestine, placenta, and liver. Also used were p -nitrophenyl phosphate (p -NPP), sodium pyrophosphate (Na-PPi), adenosine triphosphate (ATP) or pyridoxal phosphate (PLP) for substrate. The amount of inorganic phosphate as a result of hydrolysis by ALP was measured by Chifflet methods under different pH or substrate concentration. To examine the inhibitory effect on activity, levamisole or vanadate were used. The substrate concentration exhibiting 50% activity and the concentration exhibiting 50% inhibition were proposed as K0.5 and Ki0.5 respectively. When Na-PPi, the substrate of bone ALP, was used, the optical pH and K0.5 did not show differences among each of the ALP types (optical pH : 8.8 ~ 9.1, K0.5 : 3 ~ 5 mM). In the case of PLP, the substrate of intestine ALP, the optical pH and K0.5 did not show differences among each of the ALP types (optical pH : 9.9 ~ 10.4, K0.5 : 1.1 ~ 1.6 mM), although the optical pH shifted to the alkaline side. Additionally, the optical pH and K0.5 on p -NPP, which is not in the human body, but many researchers use it, did not show differences among any of the ALP types. However, in the case of PLP, Ki0.5 of levamisole was different among each of the ALP types, although Ki0.5 of levamisole or vanadate did not show any differences among the ALP types. We found that human ALP showed different inhibitor sensitivity among several ALP types, although the substrate selectivity was not determined.
  • Change of Na, K-ATPase and ouabain sensitivity in oral cancer cells accompanied by acquisition of cisplatin resistance

    Japanese, Scientific journal, cis-diamminedichloro-platinum (II) (シスプラチン,CDDP)による癌の化学療法においては癌細胞の耐性化が障害となるが,その機序には不明な点も多い。そこで,口腔癌細胞株(Sa3,H1,KB)及びそれらのCDDP耐性株(Sa3R,H1R,KBR)と正常歯肉から採取した細胞を用いて,口腔癌細胞のCDDP耐性化に伴うNa,KATPaseの変化を中心として検討した. CDDPによる 50%細胞生存濃度(IC50)は,H1とH1Rではそれぞれ1.5×10-4及び7.5×10-4 M,Sa3とSa3Rでは4.5×10-3及び5.5×10-3 M,KBとKBRでは1.0×10-4及び7.0×10-4 Mであり,正常歯肉細胞は5.0×10-5 Mであった.癌細胞は正常歯肉細胞に比べてCDDP感受性が低く、耐性株はそれぞれの感受性株より低い感受性を示した.Na,K-ATPase活性は,Sa3よりSa3Rが高く,H1よりH1Rがわずかに高値を示したが,KBとKBRで差はなかった.Ouabain存在下での各細胞のIC50は、H1とH1Rでは5及び20 nM,Sa3とSa3Rでは80及び180 nM,KBとKBRでは40及び130 nMであり,正常歯肉細胞は5 nMであった.この結果から,CDDP耐性化はouabainに対する耐性化も伴うことが示唆された.各親株と耐性株間のNa,K-ATPase活性阻害に対するouabain濃度依存性は細胞によって異なり,ouabainに対する耐性化はNa, K-ATPaseの変化によるものではないと示唆された.各細胞のP-糖タンパク質とATP7Aの発現を親株と耐性株間で比較したところ,発現量は細胞によって異なった.以上の結果から,CDDP耐性化のメカニズムは細胞の種類によって異なることと,CDDP耐性化はouabain耐性化を伴うことが示唆された.Purpose : Resistance of cancer cells is an obstacle in chemotherapy for cancer by cisplatin (CDDP). But it is not well elucidated in the mechanism. We examined the change of Na, K-ATPase accompanied by the acquisition of the CDDP resistance of oral cancer cells.Methods : Oral cancer cell lines (Sa3, H1, KB) and CDDP-resistant lines of (Sa3R, H1R, KBR) provided from Chiba University, Faculty of Medicine and cells collected from normal gingiva, were used. Na, K-ATPase activity and cell viability under treatment ouabain and CDDP for each cell was measured. Assessment of Na, K-ATPase α subunits, ATP7A and P-glycoprotein (P-gp) was done by western blotting.Results : According to CDDP, 50% inhibitory concentration (IC50) was, 7.5 × 10-4 M and 1.5 × 10-4 M respectively in H1R and H1, 6.0×10-3 M and 4.5×10-3 M respectively in Sa3R and Sa3, 7.0×10-4 M and 1.0×10-4 M respectively in KBR and KB. CDDP sensitivity was lower than the normal gingival cells to cancer cells. Resistant strains showed a lower sensitivity than the more sensitive strains of each. Measurement of Na, K-ATPase activity, Sa3R is higher than Sa3, H1R was higher than H1 slightly, but there was no significant difference in the KBR and KB. Activity measurements were consistent in Na, K-ATPase and protein expression levels. IC50 was examined for each cell in the presence of ouabain and were 80 nM and 60 nM in the H1R and H1, 180 nM and 80 nM in the Sa3R and Sa3, 130 nM and 40 nM in the KBR and KB. The normal gingival cells was 5 nM. From the results of these examinations, it was suggested the cancer cells resistant to CDDP get resistant to ouabain at the same time. As for the ouabain concentration dependence of Na, K-ATPase inhibition of the cells, there is no significant difference in the 50% inhibitory concentration between resistant and parent lines. Therefore, change of Na,KATPase sensitivity to ouabain was not related to resistance of the cells against ouabain. It was suggested that P-gp and Na, K-ATPase, ATP7A is involved in CDDP resistance. The mechanism depends on the type of cells, although we found no features that are common in the between the parent and resistant lines for the expression of ATP7A and P-gp.
  • Effects of intravenous anesthetics on the activities of various ATPases in rat brain

    Japanese, Scientific journal, 静脈麻酔薬のATPaseに対する作用の報告は多いが,作用機構の詳細については不明な点が多い.ATPaseにはF型ATPase(ミトコンドリアATP合成酵素),P型ATPase,V型ATPase等が存在し,活性発現にMg2+(Mg)を必要とするが,役割や性質の不明なMg-ATPaseも多数報告されている.本研究ではラット脳に存在するMg-ATPaseを分析するとともに,静脈麻酔薬の作用を検討した. ラット全脳から,Pottorfの方法に従って,形質膜(PII)とミクロソーム(PIII)分画を得た.Mg-ATPase活性の至適pHはPIIが約9.4,PIIIは約7.4であった.PIIのMg-ATPase活性はF型ATPaseの阻害剤であるNaN3によって約80%抑制され,PIIIのMg-ATPase活性はP型ATPase及びV型ATPaseの阻害剤であるNa3VO4及びbafilomycineA1によってそれぞれ約10%及び30%抑制された.ウエスタンブロッティングの結果からも,F型ATPaseはPIIに,V型ATPaseはPIIIに最も多く検出された.上記阻害剤で抑制されないMg-ATPase活性をBasal Mg-ATPaseとし,各阻害剤の存在下で各ATPaseを分別して静脈麻酔薬の影響を調べた.PropofolはV型ATPase活性,PII及びPIII Basal Mg-ATPase活性を濃度依存的に抑制したが,F型ATPase活性は80%程度活性化した.PentobarbitalはすべてのATPase活性を濃度依存的に抑制した.ThiopentalもすべてのATPase活性を抑制したが,その程度は各ATPaseによって異なった. 以上の結果から,静脈麻酔薬はラット脳Mg-ATPase活性を基本的に抑制するが,propofolはミトコンドリアに特異的な作用を及ぼす事が示唆された.This study examined whether the activity of magnesium-dependent ATPase (Mg2+-ATPase) in rat brains is a target for intravenous anesthetics. The effects of propofol, pentobarbital, and thiopental on magnesium-dependent ATPase in the plasma membrane fraction (PII) and microsomal fraction (PIII) isolated from rat brains homogenates by the method of Pottorf were examined. The optimal pH values for Mg2+-ATPase activities in PII and PIII were 9.4 and 7.4, respectively. The Mg2+-ATPase activity of PII was inhibited by about 80% by NaN3 (sodium azide), an inhibitor of F-type ATPase. The Mg2+-ATPase activity of PIII was inhibited by about 30% by the V-type ATPase inhibitor bafilomycin, and by about 10% by the P-type ATPase inhibitor vanadate. Western blotting analysis revealed that the largest amount of F-type ATPase was detected in PII, and the largest amount of V-type ATPase was detected in PIII. We defined all ATPase activity that was insensitive to the above inhibitors as basal Mg2+-ATPase activity. In the presence of these inhibitors, we examined the effects of intravenous anesthetics on the activities of various ATPases. Propofol inhibits V-type ATPase activity, PII basal Mg2+-ATPase activity, and PIII basal Mg2+-ATPase activity in a dose-dependent manner; however, F-type ATPase activity was uniquely stimulated about 80% by propofol at concentrations below 0.6 mM. Pentobarbital inhibited all types of ATPases in a dose-dependent manner. Thiopental inhibited all types of ATPases to varying degrees. These results suggest that intravenous anesthetics basically inhibit Mg2+-dependent ATPase activities in rat brains, but propofol shows specific effects on mitochondrial ATPase activity.

    Japanese, Scientific journal, 全身麻酔薬の作用を受ける機能性タンパク質としてGABA受容体,アセチルコリン受容体などの報告があるが,これらに対する作用のみで全身麻酔作用のすべてを説明することはできないと考えられている.本研究はラット脳に存在するカルシウム(Ca)で活性化されるATPaseが,静脈麻酔薬の直接の作用点となり得るかを検討することを目的に行なった.ラット脳のホモジェネートから調整した膜分画(PⅠ)及び,Pottorfの方法に従って調整した形質膜(PⅡ)及びミクロソーム(PⅢ)分画のCaで活性化されるATPaseに対するpropofol,pentobarbitalとthiopentalの作用を調べた.PⅠにはCaの濃度に依存して活性化されるATPaseが存在し1mM以上のCaでほぼ飽和した(Ca-ATPase活性).PⅠのCa-ATPase活性はpropofol濃度に依存して抑制され50%阻害濃度(Ki0.5)は0.25 mMであった.Propofol存在下でCa-ATPase活性のCa濃度依存性を調べると,存在するpropofol濃度に依存して最大活性とCaに対するKmは低下した.PⅡ及びPⅢには,ウェスタンブロッティング及び酵素学的性質からCa,Mg-ATPaseであるPMCA(plasma membrane Ca-ATPase)及びSERCA(sarcoendoplasmic reticulum Ca-ATPase)の存在が明らかになった.PⅡ 及びPⅢ のCa,Mg-ATPase活性はいずれも類似した濃度依存性でpropofol,pentobarbitalとthiopentalによって阻害され,そのKi0.5はそれぞれ0.35,3及び1.5 mMであった.本研究で調べたCa-ATPase及びCa,Mg-ATPase活性のpropofol,pentobarbitalとthiopentalによる活性阻害濃度は,同じ生体膜に存在する酵素であるNa,K-ATPase活性の阻害濃度と類似していた.以上の結果から,ラット脳に存在するCaで活性化されるATPase活性の静脈麻酔薬による阻害は特異的なものではなく,麻酔薬の生体膜を構成する脂質とタンパク質にに対する作用の結果として,ATPaseタンパク質の機能が影響を受け抑制されたものと推測した.

    Japanese, Scientific journal, GST融合タンパク質として作製したGlutathione-S-transferase融合タンパク質(sRANKL)はマクロファージ様細胞株RAW264.7(RAW)細胞を破骨細胞に分化させ,その濃度に依存して破骨細胞の数は増加した.sRANKLを加えて培養すると破骨細胞は一定期間は増加するが,その後にピークを迎えてから減少した.sRANKLはRAW細胞を破骨細胞に分化させると共に,破骨細胞にアポトーシスを誘導した

    Japanese, Scientific journal, Cytotoxic effects of unpolimerized composite resin which was given through the dentin tubules to the human pulp tissue were evaluated in vitro.
    Human fibroblast-Like cells(pulp cells) were isolated from extracted primary and permanent tooth pulp. The Bovine permanent tooth was cut into a dentin slice(thickness: 2 mm), and a cavity(diameter: 2 mm, depth: 1.7 m) was prepared in the slice. The cavity was filled with a composite resin (AP-X®), B-2, Kuraray Co. ) which was either light-polymerized or unpolimerized. Pulp cells on which the dentine slice containing composite resin was placed by using the cell-culture insert®(Falcon)were cultured for 24 hours.
    The number of surviving cells was counted after Trypan blue staining. In order to determine whether or not apoptosis occured in the cultured cells, fragmented DNA was extracted and electrophoresis was performed using 2% agarose gel. Concentration of IL-1β in the conditioned medium was determined by enzyme-linked immunosorbent assay(ELISA).
    The number of surviving cells decreased in both primary and permanent pulp cultures when the dentin slices filled with unpolimerized resin were applied. Polymerized resin had no cytotoxic effect on the cultured cells in the same culture condition. DNA fragmentation was not observed even when the unpolymerized resin affected the viability of the cells. IL-1β, which is often produced by inflamatory tissues, was detected in neither of the resin-treated cell cultures.
    These results indicated that unlike polymerized resin, unpolymerized resin components could pass through the dentin tubules and had cytotoxic effects on cultured cells. We conclude that the unpolymerized resin-induced cell death was apoptosis, as revealed null DNA fragmentation, but necrosis.

    細胞の分化に細胞外環境が重要であることはよく知られている。伸展力や圧迫力などの機械的刺激が骨芽細胞分化に影響を及ぼすことは多数報告されている。しかし、どの報告の力も生体内に換算すると大きな力で、わずかな力による報告は少ない。研究代表者らは間葉系幹細胞株UEF7-13に微小な圧迫力をかけることで、骨芽細胞に分化することを確認した。これは分化誘導因子を添加することなく石灰化を認めたことから、機械的刺激のみで成熟した骨芽細胞への誘導が可能なことを示唆している。本研究では、間葉系幹細胞の骨芽細胞への分化に関与するメカノセンサーの同定とその下流のメカノトランスダクションの解明、およびその臨床応用を目的とする。
    本年度の研究計画では、まず、間葉系幹細胞を分化させる至適圧迫力を探索することから開始した。予備実験で用いた圧迫力は0.005MPaであったが、圧迫力を0.0001MPa、0.0005MPa、0.001MPa、0.005MPa、0.01MPa、0.04MPaの幅で実験をする予定であった。しかし、0.0001MPa、0.0005MPa、0.001MPaのかなり微弱な圧迫力下で細胞を培養すると、細胞が死んでいく現象が続いた。培養液を頻回交換するなど検討を試みたが、圧迫力が弱いため、培養機内の空気交換が上手くできておらず細胞が酸欠状態に陥った可能性が考えられた。そこで、現在は培養機内の空気交換回数を上げるように設定しなおし、培養を継続している。現在のところ、細胞は死ぬことなく培養ができている。

  • Mechanism for Epithelial-Mesenchymal Transition in Bone

    ビスホスホネート(BP)は、骨粗鬆症治療や癌の骨転移治療に用いられており、近年乳がんの転移や再発率の抑制など抗がん効果を有することが大規模臨床試験において示された。しかしながらBPは投与後速やかに尿中排泄され、生体内においてはほとんどが骨に蓄積し、破骨細胞を特異的に抑制することで骨吸収阻害効果を発揮することから、軟組織臓器において直接的な抗がん効果を発揮することは考えづらい。このことから、BPによるヒト乳がん患者の予後改善効果は、骨環境を抑制することで発揮されている可能性を推察した。骨には様々なサイトカインが豊富に含まれ骨吸収が生じるとそのサイトカインは骨髄中に放出されることが明らかになっているが、そのなかでもTGF-βは転移促進プログラムである上皮間葉転換(EMT)を誘導することが示されている。本研究においては、BPは骨吸収抑制作用によって骨髄中におけるがんのEMTを抑制する可能性について検討を進めた。まずEMTが骨髄中で促進されるか否かについてマウスモデルを作製し検討した。間葉細胞様形態を示さないヒト乳がん細胞MCF7を乳腺部皮下ならびに脛骨骨髄中に同時接種し腫瘍増大後EMT促進因子SnailのmRNA発現量を乳腺部腫瘍と比較しながら測定した。その結果、乳腺部腫瘍に比較して骨髄内腫瘍においてSnail発現が増加し、それに並行して上皮マーカーE-cadherinの発現が著明に減少した。この結果より骨髄はがん細胞のEMTに好都合な環境であることが推察された。更に現在はBPの培養細胞への添加がSnailをユビキチンプロテアソーム系での分解を促進するといった直接的なEMT抑制効果を有することを見いだしておりそのメカニズムを解析している。骨におけるがん細胞のEMTは現在まで示されておらず、本研究の進展は新たな組織特異的EMT制御メカニズムの一端を報告できると考えている。

  • 閉経後の骨粗鬆症に対する乳歯由来歯髄幹細胞の培養上清の効果

    本研究は、ヒト乳歯由来歯髄幹細胞から分泌されるサイトカインや成長因子、ケモカイン、エクソソームなどの生理活性分子群が骨組織のリモデリングにおいて骨吸収に対して抑制効果を有することを、動物実験によって明らかにすることを目的にしている。本年度の研究実績として、卵巣摘出後の骨粗鬆症モデルマウスを用い、ヒト乳歯由来歯髄幹細胞の培養上清液を投与し、エストロゲン欠乏によって発症する骨粗鬆症に対して抑制効果を有していたことについて報告を行った(Maeda A, et al., Exp Ther Med. 2022 Apr 23(4):299.)。
    この研究において、骨粗鬆症モデルマウスにヒト乳歯由来歯髄幹細胞の培養上清液を投与することで、エストロゲン欠乏による骨吸収が抑制されたことから、ヒト乳歯由来歯髄幹細胞の培養上清液には、骨リモデリングを正常化する因子が含まれていることが判明した。その作用機序についての詳細について検索を行ったところ、骨粗鬆症モデルマウスにおいてエストロゲン欠乏により活性化していた炎症性免疫細胞が、ヒト乳歯由来歯髄幹細胞の培養上清液の投与によってその活性が改善されていることを明らかにした。
    本成果と関連するデータにおいて、ヒト乳歯由来歯髄幹細胞の培養上清液中の生理活性分子群が免疫調節に関わることが予想されることから、ヒト乳歯由来歯髄幹細胞による免疫調整機能と骨組織のリモデリングにおける相互機能の解析が今後重要になると考えられた。

    To enhance the bonding treatment of vertically fractured roots, we aimed to the newly cementum formation on a compound of resin and dentin powder, or β-TCP which may improve the binding between resin and cementum. In vitro, cytotoxicity was assessed. It had little cytotoxicity. In vivo, we observed the newly hard tissue formation on a resin containing β-TCP applied by rhBMP-2 and cultured bone marrow stromal cell sheets. This experiment might exhibit a possibility of the newly hard tissue formation on a resin.

    In this study, we examined the pathological and/or unphysiologicalroot resorption mechanism focused on the two types of osteoclasts. Our results weresuggested that morphologic characteristics of the osteoclasts varied according to thedown-regulation of Treg activity, difference of the major histocompatibility complex(MHC) haplotype, tensile force in culture condition and ROS scavengers.

    Salivary glands of rats fed a liquid diet for 3 weeks were investigated microscopically to clarify the effects of soft food on salivary glands. Parotid glands of experimental rats became atrophic after liquid diet feeding, and cell shrinkage, cell death, and reduction of speed of cell-cycle were identified in the atrophic parotid glands. However, such atrophic alterations were not observed in submandibular and sublingual glands following liquid diet feeding.   • Study of evolution of osteoclasts regulating factors in teleost fish

    The origin of osteoclasts resorbing bone remains unknown. Bone tissue is seen on teleost fish in phylogeny, and there old osteoclasts would be seen. The present study established the culture system of isolated Japanese medaka and observed their structure by light-and electron microscopy. The cultured cells showed no resorptive lacuna but etched surfaces on sperm whale dentine. These results may be related to the evolution of regulating fac tors of osteoclasts.

    In this study, we investigated the effects of bisphosphonate on osteoclasts and their precursor cells to clarify one of the mechanisms of bisphosphonate-related osteonecrosis of the jaw(BRONJ). BRONJ-related bisphosphonates strongly inhibited osteoclastogenesis and bone resorption activity. On the other hand, they showed cytotoxicity and enhanced microbial pathogens-induced cell death.

    Stem cells from human exfoliated deciduous teeth(SHED) are stem cell that differentiate into osteocyte and adipocyte. In this study, we investigated whether the Wnt signaling inhibitor has a potency of the stemness of SHED after long-term cell culture or not. We demonstrated that SHED treated with the Wnt signaling inhibitor was capable of stemness in vitro experiment and generating bone structures at 8 weeks post-transplantation into nude mouse.     本研究は、歯の発生の仕組みを明らかにするために、転写調節因子のひとつであるカルシニューリンについて着目し、歯胚形成期におけるカルシニューリンの発現について検索を行った。歯胚の分化期である蕾状期から鐘状期のではカルシニューリンは、歯性上皮由来の細胞、外胚葉性間葉由来の細胞両方において発現が確認された。以上のことからカルシニューリンは、歯の発生・分化過程において必要なシグナル分子として作用していることが示唆された。

    本研究は、骨免疫学的観点からCD4 陽性T 細胞(Th1、Th2、Th17細胞)に着目し、病的あるいは非生理的な乳歯歯根吸収機構の解明を目的に検索を行った。Th1細胞の産生するインターフェロン-γ、Th2細胞の産生するインターロイキン(IL)-4は破歯細胞への分化を抑制し、Th17細胞の産生するIL-17は亢進した。これらの結果はCD4 陽性T 細胞が産生するサイトカインが歯根吸収機構に作用していることを示唆した。

    慢性骨髄炎などの骨硬化を伴う疾患においては病巣局所に対する薬物の移行性が低下しており、治療上大きな障害となっている。骨硬化の原因としては局所に存在する骨芽細胞の機能が亢進し、石灰化が促進していることが推測される。病原体の侵入を特異的に認識することが知られているToll様受容体(TLR)がマクロファージ等の破骨細胞前駆細胞に発現しており、細菌性抗原などを認識して破骨細胞に分化することが知られている。本研究において、われわれは骨芽細胞におけるTLRを介したシグナルに関して研究を行い、それらの関与する遺伝子発現を制御することにより、硬化性骨髄炎の治療に応用することを試みようとした。石灰化現象を観察可能な骨芽細胞としてMC3T3-E1細胞を用い、硬化性骨髄炎の原因の一つであると推測されるウイルスの2本鎖RNAの代用としてpoly(I):poly(C)を用いて実験を行った。RT-PCR法ならびにウエスタンブロット法によりMC3T3-E1細胞には2本鎖RNAを認識するTLR3が発現していた。MC3T3-E1細胞をpoly(I):poly(C)処理すると転写因子STAT1の活性化が生じ、インターフェロン(IFN)β、CXCL10、TLR3の発現が増加した。これらの発現の増加は抗IFN-β抗体を細胞に作用させることにより抑制された。これらの結果からウイルス感染を生ずるとTLR3の発現を増加さ...

  • 骨再生を目指した転写因子Osterixの機能調節機構の解明

    昨年度に引き続き、Osterix転写活性化領域の詳細を同定するために、GAL4-binding sitesと連接させたルシフェラーゼreporter plasmidを用いたGal4融合システムレポーター分析を行い、Osterixの転写活性を調べた。その結果、Osterixの転写活性領域がプロリンとグリシン残基に富んでおり、哺乳類とイーストの細胞における活性の特性を持っていることを認めた。GST-pull down分析および免疫沈降法では、TBPではなく、基本転写因子であるTF-IIBが転写活性領域に結合することを示した。さらに、Osterix相互作用因子を探索するために、FLAG-tagを付加したOsterix発現plasmidを培養細胞に導入し、αFLAG抗体を用いた免疫沈降法を行ったところ、in vivoとin vitroにおいて、OsterixがそのC末端Zinc-Finger領域を通して、クロマチンリモデリング因子Brg-1および癌抑制遺伝子p53と相互作用することを見つけた。Osterixにより発現制御される遺伝子群の検索するために、G418耐性遺伝子を有したOsterix発現plasmidを作製し、薬剤選択によりOsterix遺伝子をstableに過剰発現するSaOS2細胞を樹立し、その遺伝子発現について解析した。RT-PCR法により骨芽細胞特異的遺伝子群の発現に...

    本年度はreceptor activator of NF-kappa B (RANK) ligand (RANKL)の真核細胞発現ベクターをマウス骨髄由来細胞ならびにマウス由来株化細胞(RAW264.7細胞、J774-1細胞)への遺伝子導入を行った。インデューサ(テトラサイクリン)を添加時にのみ目的遺伝子を発現するベクターを用いることによって、目的タンパク質の発現がコントロール可能となり、適切な時に適切な発現を可能とした。しかしながら、リポフェクション法、エレクトロポレーション法では、ともに導入率が0.01%以下であった。また、レトロウイルスベクターを用いた方法では導入率が0.01%程度であった。以上のように、遺伝子導入を効果的・効率的に行うことができなかった。予備実験の結果から炎症性サイトカインの1つであるマクロファージ遊走阻止因子(MIF)はその濃度に依存して、TRAP陽性細胞の総核数を変化させることなく、マウス骨髄由来細胞から分化したTRAP陽性多核細胞の出現数を減少させることが判明した。そこで、MIFの真核細胞発現ベクターをマウス骨髄由来細胞ならびにマウス由来株化細胞への遺伝子導入を、上記と同様にリポフェクション法、エレクトロポレーション法、レトロウイルスベクターを用いた方法で行ったが、効果的・効率的に行うことができなかった。マウス由来株化細胞(RAW264.7細胞)...

    We investigated the effects of general anesthetics and the temperature on electron spin resonance (ESR) spectra of multilamellar liposome (MLV). Liposomes were composed of DMPC, DOPC or EYPC, labeled with 5-DSA or 16-DSA. The ESR spectra were measured at different temperatures from 10 to 50 ℃. The spectra of MLV labeled by 16-DSA showed three sharp signals compared with 5-DSA. The difference of order parameter S value of 5-DSA and 16-DSA suggested that the nitroxide radical location of 5-DSA and 16-DSA were different in the membrane bilayr.     骨形成・骨吸収の遺伝子治療が考えられている現在、病的あるいは非生理的な「歯根吸収の制御」を遺伝子治療という観点から考えることは、新しい研究分野の展開につながる将来性のある研究の第一歩と考える。破骨細胞は骨吸収を担う多核細胞であり、単球・マクロファージ系細胞の前駆細胞から分化することが知られている。予備実験の結果から、今回我々は、炎症性サイトカインの1つであるマクロファージ遊走阻止因子(MIF)が、破骨細胞分化過程にどのような影響を及ぼすかについて検討を行った。マウス骨芽細胞様細胞(MC3T3-E1細胞)とマウス骨髄細胞との共存培養系によって破骨細胞を誘導し、酒石酸耐性酸性ホスファターゼ(TRAP)活性陽性細胞の出現数、破骨細胞分化因子(RANKL)の発現量、破骨細胞抑制因子(OPG)の産生量および象牙質切片を用いた硬組織吸収能についてMIFの影響を調べた。その結果、TRAP陽性多核細胞の出現数は、MIF濃度に依存して減少することが判明した。一方、MC3T3-E1細胞におけるRANKLの発現およびOPGの産生に、MIFの影響は確認されなかった。TRAP陽性細胞1個中に含まれる核数の分布を調べたところ、MIF作用群ではコントロールと比較し、多核化が抑制されていることが明らかになった。また、象牙質切片上に形成された吸収窩を観察したところ、MIF作用群では形成された吸収窩の大きさが明...

    ウサギ脳Na^+,K^+-AIPaseを用いて、種々の全身麻酔薬およびその関連薬がNa^+,K^+-AIPaseに及ぼす影響と作用機構について調べた。その結果、フェンタニルを除くすべての麻酔薬および関連薬は、濃度依存性にNa^+,K^+-ATPase全活性と部分反応であるNa^+-ATPase活性(Na^+活性)およびK^+-pNPPase(K^+活性)を阻害した。揮発性麻酔薬においては50%阻害濃度(IC_<50>値)と一般に麻酔作用の強さの指標とされるMAC値の間に強い相関関係(r=0.938)がみられた。各麻酔薬の全活性、 Na^+活性およびK^+活性に対するIC_<50>値の順序を比較したところ、バルビタール類、ペンゾジアゼピン類、揮発性麻酔薬およびその他の麻酔薬で順序のパターンに差異がみられ、麻酔薬のグループごとに阻害機構が異なることが示唆された。そこで各麻酔薬のリン酸化反応中間体(EP)形成に対する作用を調べたところ、全活性が阻害される濃度でベンゾジアゼピン類はEP形成を抑制し、揮発麻酔薬とケタミンは抑制しなかった。ミダゾラム、イソフルラン、ケタミンの濃度を変えてEP形成に対する作用を調べたところペンゾジアゼピン類であるミダゾラムのみが濃度依存性にEP形成を抑制した。また、揮発性麻酔薬であるイソフルランにおいてEPのK^+に対する反応性は低下し、ケタミンでは増加し...

    癌抑制遺伝の一つであるRbタンパク質(pRb)は転写因子E2Fと結合することにより細胞分裂を抑制しており、pRbがサイクリン-サイクリン依存性キナーゼ複合体によってリン酸化されるとE2Fと離開し、細胞分裂が開始することが知られている。これまでにpRbの発現の低下および遺伝子の変異が種々の癌で報告されているが、そのリン酸化の程度を調べた研究はない。今回我々は口腔癌20症例を対象としてpRbの発現およびそのリン酸化の状態をリン酸化部位特異的抗Rb抗体を用いて検索した。対照群6例では全症例でpRbを発現していたのに対し、腫瘍群では20例中9例でのみpRbを発現していた。また、pRb発現症例に限定すると対照群のpRbはリン酸化されていなかったのに対し、腫瘍群ではThr373のリン酸化が5症例、Ser780は6症例、Ser807/Ser811は8症例で生じていた。リン酸化の程度はどの部位においてもT1からT4になるにつれて上昇する傾向が見られた。pRbを発現しかつ低リン酸化型の症例を活性化型、pRbを喪失しているか高リン酸化型の症例を不活性化型として分類すると、対照群は6症例とも活性化型として、腫瘍群はThr373では20症例中16症例(80%)、Ser780では17症例(85%)、Ser807/Ser811では19症例(95%)が不活性型として分類され、対照群と腫瘍群間で有意差が認め...

  • 骨形成細胞の石炭化におけるアルカリ性ホスファターゼの生理的役割の解明-異なる培養条件を用いて-

    ラット胎児頭蓋冠由来骨形成細胞をBGJ_b並びにαMEMを用い培養し、これら細胞のアルカリ性ホスファターゼ(ALP)活性を測定した。活性は、細胞が単層期(con fluence)では正常環境(対照)に比べ、低カルシウム濃度培養液(低Ca環境)で有意に高い値を示した。この活性は、細胞多層化期(multilayer)では両培養液の細胞共に対照では上昇したが、低Ca環境では変動せず、その結果として対照と低Ca環境下の細胞間で、有意な差は認められなくなった。活性は、その後では対照における活性はBGJ_bで上昇せず、αMEMで上昇を続けた。光学並びに電子顕微鏡に認められた石炭化部位は、BGJ_bの対照並びに両培養液の低Ca環境では認められず、αMEMの対照のみ認められた。αMEMの対照ではALP活性の上昇並びに細胞の多層化と共に結節を形成し、石炭化の開始が認められた。オステオカルシンはαMEMの対照のみに検出された。これらの結果は、多くの研究で行われているような培養液へのβ-glycerophosphateやdexamethasone等の添加をすることなしに得られた。以上の結果から、低Ca培養液において細胞のALP活性が亢進することは細胞の機能を考える上で興味深く、その一つとして環境の異常に対する細胞の反応が推測された。また、培養液の違いによる石炭化現象の有無は、BGJ_bとαMEMの...

    It was examined whether the gene function of mouse osteoblastic MC3T3 - E 1 Cells (E 1 cells) was affected by a low-calcium environment.In the E 1 cells with the treatment of FBS or EGF,c-fos mRNA expression increased with time in the control and low Ca groups, reached maximum at 30 minutes and then decreased at 60 minutes.     ラット胎児頭蓋冠由来骨形成細胞を、BGJb並びにalphaMEMを用い培養した。これら細胞のアルカリ性ホスファターゼ(ALP)活性は、細胞が単層の時期では正常環境(対照)に比べ、低カルシウム濃度培養液(低Ca環境)で有意に高い値が認められた。細胞が多層化する時期では、この活性は両培養液の細胞共に対照では上昇したが、低Ca環境では変動せず、その結果として対照と低Ca環境下の細胞間で、有意な差が認められなくなった。その後、対照における活性はBGJbでは上昇せず、alphaMEMでは上昇を続けた。石灰化部位は、BGJbの対照並びに両培養液の低Ca環境では認められず、dMEMの対照のみ認められた。dMEMの対照はALP活性の上昇並びに細胞の多層化と共に結節を形成し、石灰化の開始が認められた。引き続き、結節間の連結が生じることも認められた。走査型電子顕微鏡像では細胞膜には多数の長い突起が認められ、伸展した突起の一部は他の細胞と結合していた。また、透過型電子顕微鏡像ではコラーゲン線維と基質小胞が認められた。オステオカルシンはalphaMEMの対照のみに検出された。これらの結果は、beta-glycerophosphateやdexamethasone等を用いることなく得られた。一方、BGJbを用いた場合、本研究の初代培養細胞のみならず骨芽細胞様細胞株(MC3T3-E1細胞)でもALP活性の...
