Scientific journal, Marine ecosystems in the northwest Pacific are known for their high biodiversity, with many marine species, from tiny plankton to huge macroalgae. Marine biodiversity is affected by irreversible global warming, mainly the increase of greenhouse gas emissions from anthropogenic activities. For the past several decades, marine ecosystems in East Asia have been changing due to the increase in sea surface temperature (SST), which has changed some important environmental factors, including rainfall patterns, extreme weather, and ocean circulation. Such modifications in the environmental parameters have altered the physiology, phenology, and distribution pattern of marine organisms. The SST increase has also altered population and community structure, and the functioning of the ecosystem. Certain subtropical and tropical fauna and flora are now extending their range of distribution from the warm southern area to the temperate regions, disrupting or modifying the biotic interactions in the temperate ecosystem. Collaborative conservation projects and responsible policies are crucial to safeguard the environmental value of East Asia's marine ecosystems for future generations and mitigate the negative effects of global warming on marine biodiversity. Scientific journal, To understand the impact of the melting of late summer Arctic brash ice on the surface waters of the Chukchi Sea, we collected sea-ice samples during 2021. Floating sea ice was collected by a wire mesh pallet cage from the side of the R/V Mirai. We measured physical and biogeochemical parameters such as salinity, oxygen stable isotopic ratios, turbidity, and concentrations of chlorophyll-a and nutrients. The samples of brash ice were multiyear ice based on satellite back-trajectory analysis. Comparison of nutrient concentrations in brash ice with those of seawater samples from the temperature minimum layer similar to the water in the sea ice originated suggested that the characteristics of the brash ice were greatly affected by biogeochemical processes such as remineralization. The extremely high turbidity and concentrations of chlorophyll-a observed in the brown/green ice samples reflected the impact of sediment as well as the influence of biological activities. The N:P ratios were less than 1 because of the high phosphate concentrations, even though the ammonium concentrations were high. We hypothesized that this low N:P ratio reflected the combined effects of the accumulation of nutrients due to remineralization in the biofilm and differences of remineralization rate and adsorption features of nitrogen and phosphorus. Based on the high nitrate and ammonium concentrations in the sea-ice samples, we postulated a marked impact of sea-ice meltwater on the nitrogen cycle in the nitrate-depleted surface waters of the Chukchi Sea during late summer. We estimated that meltwater nitrogen could support 0.3%–2.6% of primary production in the northern Chukchi Sea. Our results suggest that high-turbidity ice will play an important role as a source of nutrients to the ocean during melting of sea ice, and understanding its distribution, amount, and geochemical characteristics is vital. - Linkage between spatiotemporal distribution of environmental

Environmental DNA (eDNA) is a promising tool for the continuous monitoring of fish ecology and diversity. However, its potential for describing the phenological activity of fish has rarely been examined. This study aimed to elucidate a linkage between the spatiotemporal distribution of eDNA and the phenology of an amphidromous fish, ayu Plecoglossus altivelis altivelis, in a river in Hokkaido, Japan, which is its northernmost distributional area. A significant positive correlation between eDNA concentration and catch per unit effort of P. a. altivelis in the river confirmed the use of eDNA as a surrogate for the abundance of P. a. altivelis. eDNA of P. a. altivelis was first detected in late April on a sandy beach adjacent to the river mouth. Subsequent to its first detection at the lowest site in the river in early May, eDNA spread throughout the river, indicating the upstream migration of P. a. altivelis. Spawning activity was also represented by a rapid increase in eDNA concentration and its surge at night in the lowest reaches of the river during September and October. These results suggest that upstream migration and spawning primarily commenced when the water temperature reached 10°C and decreased below 20°C, respectively. This observation is consistent with the behavioral responses observed in P. a. altivelis populations from other regions of Japan. Scientific journal, Ongoing warming and sea-ice reductions in the Arctic can seriously impact cold-water species, such as polar cod (Boreogadus saida), necessitating biomonitoring to reveal the ecological consequences. Recent methodological advancements in environmental DNA (eDNA) techniques have increased our ability to conduct ecological monitoring at various locations, including the Arctic. This study aimed to provide an overview of the distribution of polar cod across the Bering and Chukchi Seas by employing species-specific detection of eDNA. First, we successfully developed novel species-specific qPCR assay targeting the mitochondrial D-loop region, which exclusively amplifies eDNA derived from polar cod. Subsequently, polar cod eDNA was detected using the assay from the samples that we collected latitudinally across the study area during the open-water season. Polar cod eDNA was primarily detected in the surface water from the central Chukchi Sea shelf and the northernmost observation line (75°N), which was located on the shelf slope, off the Point Barrow, and in the marginal ice zone. In contrast, only trace amounts of eDNA were detected in the Bering Sea. This pattern corresponded well with the distribution of water masses classified based on environmental conditions. The detection of eDNA in surface water was clearly limited to cold (-1 to 5°C) and low salinity (25–32) water, whereas it was detected in a higher salinity range (32–34) in the middle and bottom layers. These findings are consistent with current knowledge about the distribution and habitat of the polar cod, suggesting that eDNA can be regarded as a reliable tool to replace or supplement conventional methods. Incorporating eDNA techniques into large-scale oceanographic surveys can improve the spatial and temporal resolution of fish species detection with a reasonable sampling effort and will facilitate the continuous monitoring of Arctic ecosystems. - Evaluating the sampling effort for the metabarcoding‐based detection of fish environmental DNA in the open ocean

本年度は、浜名湖に生息する魚類の採集およびこれまでに採集した魚類のDNA解析と、浜名湖内の水流1の観測およびネットサンプリングによる浮遊性魚卵の採集をおこなった。
(1)本研究では、まず、浜名湖産魚類を網羅したDNAデータベースを作成することを目的とした。前年度に採集した魚類と本年度に採集した魚類、合わせて15目63科119種類に属する212個体を材料に、ミトコンドリアDNA(mtDNA)の16SrRNA領域の約1000塩基対の配列を決定した。その結果、新たに23種の魚類についてmtDNAの16SrRNA領域のシークエンスデータが得られた。
(2)2007年1月22日にAcoustic Doppler Current Profiler(ADCP)による浜名湖本湖内の表層流を観測した。その結果、湖内の東側では北向きの流れが卓越することが明らかとなった。
(3)2007年1月23日〜3月9日に浜名湖内に設けた6定点(水深2.2〜11.5m)において係留計を設置し、長期的な湖水の流れを観測した。その結果、湖口に近い定点では、潮汐に同期した流れが観測され、湖口部においては外海との水の行き来があると考えられた。また、湖中央部においては全体として反時計回りの流れがあることが明らかとなった。
(4)2007年3月9日にADCPによる海流観測と、浮遊性魚卵の採集を同時におこなった。ADCPによる観測結果は現在解析中である。浮遊性魚卵は湖内に設けた5定点で、目合い0.33mmのプランクトンネットを表層曳きし採集をおこなった。採集したネットサンプルはホルマリンサンプルとして保存した。このサンプルについても現在解析中である。
以上、平成18年度の研究結果から、湿地からのメタン放出量を適切に見積もるためには、噴出による大きなフラックスの変動をカバーする高頻度での観測を行う必要があること(特に気圧が低下局面にある悪天候時に観測を行うこと)、大きな放出が生じる融雪期にも観測を行う必要性があること、が明らかとなった。
