English, Optic morphology (Om) mutations in Drosophila ananassae map to at least 22 loci scattered throughout the genome. They are semidominant, neomorphic, nonpleiotropic, and are associated with the insertion of a retrotransposon, tom. The Om(1A) gene, which is cytogenetically linked to the cut locus, was cloned using a DNA fragment of the cut locus of Drosophila melanogaster as a probe. Three of the eight alleles of Om(1A) examined have insertion of the tom element within a putative cut region. The gamma-ray-induced revertants of Om(1A) are accompanied with cut lethal mutations and rearrangements within the cut coding region. In the eye imaginal discs of the Om(1A) mutants, differentiation of photoreceptor clusters is suppressed, abnormal cell death occurs in the center and the cut protein is expressed ectopically. D. melanogaster flies transformed with a chimeric cut gene under the control of a heat-inducible promoter show excessive cell death in the region anterior to the morphogenetic furrow, suppressed differentiation to photoreceptor clusters and defect in the imaginal eye morphology when subjected to temperature elevation. These findings suggest that the tom element inserted within the Om(1A) region induces ectopic cut expression in the eye imaginal discs, thus resulting in the Om(1A) mutant phenotype. - A Novel Fluorescent Method for in situ Hybridization.

In 1996, genes involved in the biosynthesis of a long-chain polyunsaturated fatty acid (LCPUFA), eicosapentaenoic acid (EPA), were first cloned from a bacterium isolated from the intestines of marine fish. This was approximately 10 years after the discovery of deep-sea psychrophilic bacteria capable of producing EPA and docosahexaenoic acid (DHA). Homologous genes now designated as pfa genes, including those responsible for the biosynthesis of DHA have been found in marine bacteria. These pfa genes generally consisted of five pfaA−pfaE genes encoding multifunctional proteins and were similar to those required for polyketide biosynthesis. The recombinant production of EPA and DHA mostly in Escherichia coli has been achieved. Although comparable pfa genes sequences can be retrieved from DNA databases, including those established using bacteria not only from cold temperature environment but also from subtropical and tropical regions, the distribution of typical pfa genes responsible for LCPUFA biosynthesis was restricted in some genera of marine γ-proteobacteria. Recent studies demonstrated that pfa genes, together with four oleA−oleD genes, were involved in the biosynthesis of a very long-chain polyunsaturated hydrocarbon, hentriacontanonaene (C31:9). Interestingly a set of pfa and ole genes was presumed to be distributed in all LCPUFA-producing bacteria and even in some strictly anaerobic bacteria that contain C31:9. ナミテントウ(Harmonia axyridis)は,生態学,応用昆虫学の研究材料として古くから注目されてきた。本種の翅鞘斑紋は遺伝的多型の例として知られ、赤地型、二紋型、四紋型、多紋型などが知られている。遺伝学的研究によれば、斑紋を支配する遺伝子は30以上知られており、これらの遺伝子はすべて同じかもしくはきわめて近接した遺伝子座の複対立遺伝子であると考えられている。本研究では斑紋形式のメカニズムの解明の第一歩として、斑紋形成に関わる遺伝子の探求を以下の2つの方法で試みた。
鱗翅目昆虫では、Distal-less(Dll)遺伝子が斑紋を形成する翅始原組織の細胞群で発現していることが明らかになっている。そこでDll遺伝子が、ナミテントウにおいても斑紋状に発現している可能性について調査した。組織学的観察の結果、翅原基の形成は4齢虫期に開始することが明らかになったので、4齢幼虫、前蛹、蛹期の翅原基でのDll遺伝子の発現をチョウのDll蛋白に対する抗体を用いて調べた。その結果、4齢虫期の翅原基においてはこの遺伝子は翅の縁の沿って発現しており、斑紋状には発現していなかった。従ってナミテントウではDll遺伝子は、斑紋形成には直接は関与していないと考えられた。
次に、斑紋形成に関与する遺伝子を単離することを目的に、翅原基に特異的なcDNAライブラリーを作成することにした。まず、4齢、前蛹、蛹の各段階の翅原基からcDNAライブラリーを作成した。次に、前蛹のcDNAから4齢のcDNAと共通に存在するものをハイブリダイゼーションを利用して除去、前蛹期に発現が始まる遺伝子のcDNAとした。また、前蛹のcDNAと蛹のcDNAとに共通するものを除去し、蛹期に発現が減少する遺伝子のcDNAとした。以上の結果、10個の遺伝子が分離された。これらのcDNAの塩基配列を決定し、ホモロジー検索をした結果、これまで知られている遺伝子と高い相同性は示されなかった。今後、得られたcDNAを用いて本種翅始原組織における発現場所を調べ、これらの遺伝子が斑紋形成に関係しているかどうかを明らかにしたい。

Tandem insertions of defective P elements (1.15 kb KP and 0.6 kb core P) accelerate the transcription rate of the glucose-6-phosphate dehydrogenase (G6PD) gene in Drosophila melanogaster. In this resarch, we have analyzed the activation mechanism of G6PD promoter by in vitro transcription and gel retardation assays. Results showed that on cis-acting region in the core P and two such regions in the KP are associated with the activation of G6PD promoter, and that putative transcriptional regulatory protein(s) which specifically bind to each of the cis-acting regions are present in nuclear extracts of Canton S embryos. On the other hand, the P elements do not activate the normal actin 5C promoter, but activate the promoter when the 20 bp sequence around the G6PD transcription start site is placed in front of the promoter. It seems that the GC-rich region in the 20 bp sequence is required for the activation.

In recombinant DNA techniques, it is required to detect a very small amount of nucleic acids. In standard protocols this is accomplished by the use of radio-labeled nucleic acids as probes. However, the inherent drawbacks of radiolabeled probes, such as chemical lability due to radiolytic decomposition, personnel safety and disposal problems, short half-life and duration of autoradiographic exposure limit the usefulness of radio-labeled probes in conventional laboratories. It is therefore desirable to have sensitive, non-radioisotope methods for detecting nucleic acids. Recontly, efforts have been made in a number of laboratories to innovate a new non-radioisotopic methods and a variety of methods have been proposed. Among these, the Boehringer's Digoxigenin-alkaline phosphosphatase method utilizing either BCIP-Nitro BT or a chemiluminescent substrate, AMPPD as a substrate appears to be most reliable and sensitive method. However, the method is still lower in sensitivity than the RI method. To improve the sensitivity of thenon-RI method, we have attempted in this profect to synthesize fluorogenic substrates for alkalien phosphatase which upon hydrolysis yield insoluble, highly fluorescent products. As a result, HNPP (3-hydroxy-N-2-biphenylyl-2-naphthalenecarboxamide phosphate) was found to be the most useful substrate among a total of 86 fluorochromes synthesized. With this substrate, it is possible to detact as small as 10fg of DNA by spot tests, and 70fg of DNA by southern blots. The method ustilizing HNPP is superior to any available techniques with respect to resolution, speed, ease of reaction monitoring, data storage, reprobing and stability of reagents.
