English, Scientific journal, Conditioned taste aversion (CTA) is established by pairing a taste solution as a conditioned stimulus (CS) with visceral malaise as an unconditioned stimulus (US). CTA decreases the taste palatability of a CS. The bed nucleus of the stria terminalis (BNST) receives taste inputs from the brainstem. However, the involvement of the BNST in CTA remains unclear. Thus, this study examined the effects of chemogenetic inhibition of the BNST neurons on CS intake after CTA acquisition. An adeno-associated virus was microinjected into the BNST of male C57/BL6 mice to induce the inhibitory designer receptor hM4Di. The mice received a pairing of 0.2% saccharin solution (CS) with 0.3 M lithium chloride (2% BW, intraperitoneal). After conditioning, the administration of clozapine-N-oxide (CNO, 1 mg/kg) significantly enhanced the suppression of CS intake on the retrieval of CTA compared with its intake following saline administration (p < 0.01). We further assessed the effect of BNST neuron inhibition on the intake of water and taste solutions (saccharin, sucralose, sodium chloride, monosodium glutamate, quinine hydrochloride, and citric acid) using naïve (not learned CTA) mice. CNO administration significantly decreased the intake of saccharin and sucralose (p < 0.05). Our results indicate that BNST neurons mediate sweet taste and regulate sweet intake, regardless of whether sweets should be ingested or rejected. BNST neurons may be inhibited in the retrieval of CTA, thereby suppressing CS intake. - Effects of area postrema lesions and bilateral subdiaphragmatic afferent vagotomy on emetine-induced conditioned taste avoidance in rats.

English, Scientific journal, We investigated the effect of area postrema lesions and selective vagotomy of afferent fibers on emetine-induced nausea in rats. We evaluated the acquisition of the conditioned taste avoidance (CTA) to 0.1% saccharin solution after conditioning with emetine dihydrochloride (5.54 mg/kg, i.p., 1% BW). The CTA was measured in three groups of rats: a bilateral subdiaphragmatic afferent vagotomy group, an area postrema lesion group, and a sham lesion group. The bilateral vagotomy and sham groups of rats showed acquisition of CTA within 2 days of the test date. Taste avoidance was never conditioned in the area postrema lesion group. English, Scientific journal, OBJECTIVES: The aim of the present study is to demonstrate the effects of inhaled methyl methacrylate (MMA) on the excitability of neurons in the area postrema (AP). We also investigated the relation between vagal afferent inputs and responding cells in the AP. METHODS: We set up two groups of experimental animals, such as rats inhaling MMA and rats inhaling room air. To visualize the changes of AP neuron excitability after inhalation of MMA for 90 min, c-Fos protein expression was identified and quantified by immunohistochemical analysis. Some rats receiving ventral gastric branch vagotomy were also subjected to the abovementioned experiment. RESULTS: The number of c-Fos-immunoreactive (Fos-ir) cells in the MMA group was more than six times greater than that of the control group (statistically significant, p < 0.01). In vagotomized rats inhaling MMA, markedly smaller number of Fos-ir cells was identified in the AP compared to that of rats inhaling MMA without vagotomy. CONCLUSIONS: These results indicate that inhalation of MMA increases neuronal excitability in the AP, suggesting that vagal afferent inputs are involved in the induction mechanism of Fos-ir cells by MMA. - 経管栄養時の血糖調節に関する基礎的研究—味覚,嗅覚,視覚が血糖動態に与える影響―

English, Scientific journal, It remains unclear whether the rhythmic processes of chewing and gait synchronize during concurrent execution in humans. To evaluate the entrainment of chewing rhythm by gait speed, we measured electromyography from the masseter and tibialis anterior muscles during chewing at a habitual rhythm while walking on a linear treadmill in 12 healthy volunteers. Vertical movement of the head was also measured using an accelerometer. Each 5-min session included gait tasks using a treadmill at three speeds: Auto: the participant's self-selected gait speed, High: Auto × 1.3, and Low: Auto ÷ 1.3. Electromyography from the masseter muscles were also measured during chewing while stationary (Chew-Only). Chewing rhythm during walking was the same as that for head movement, occurring at twice the speed of the walking rhythm, in nine participants (Low), eight participants (Auto), and eight participants (High). For these participants, chewing rhythm in the Auto and High conditions differed significantly from that in the Chew-Only condition. Significant differences in chewing rhythm were also observed among gait speeds (Low vs. Auto vs. High). Our findings demonstrate that entrainment of habitual chewing rhythm to gait speed is a significant phenomenon, and that the dominant ratio of chewing-walking-head movement rhythms is 2:1:2. - エメチン誘発の悪心に対する迷走神経切除と最後野損傷の影響

English, Research institution, This mini-review highlights electrophysiological studies of neurons in the area postrema, including studies using anesthetized animals, neurons in the brain slices, and cultured cells. Electrophysiological studies have demonstrated chemosensitivities, intrinsic membrane properties, synaptic responses and the receptive mechanism of several peptides. For seven decades, electrophysiological research has produced much of what we know about the behavior of postrema neurons. English, Scientific journal, Cholecystokinin (CCK) is a well-known gut hormone that shows anorexigenic effects via action at peripheral and central receptors. CCK is also widely distributed throughout the mammalian brain and appears to function as a neurotransmitter and neuromodulator. The area postrema is one of the circumventricular organs, located on the dorsal surface of the medulla oblongata at the caudal end of the fourth ventricle. Blood vessels in the area postrema lack a blood brain barrier, offering specific central neural elements unique access to circulating substances. Immunohistochemical studies show CCK-A receptors in the area postrema, and we reported CCK-sensitive area postrema neurons. However, the receptive mechanism of CCK in area postrema neurons still remains unexplained. We investigated the responses of area postrema neurons to agonists and antagonists of CCK receptors using whole cell and perforated patch-clamp recordings in rat brain slices. The application of CCK-8 elicited excitatory responses, such as increases in the frequency of mEPSCs (miniature excitatory postsynaptic currents), a shift toward larger amplitude mEPSCs, and increases in the frequency of action potentials. These changes were found mostly in cells not displaying the hyperpolarization-activated cation current (Ih), except for small excitatory changes in a minority of Ih-positive neurons. Tonic inward currents or an inhibitory response to CCK-8 were never seen. Analysis of the amplitude of mEPSCs before and after the administration of CCK-8 indicated the responses mediated via the presynaptic receptors. The effect of CCK-8 was abolished in the presence of CNQX (AMPA type glutamate receptor antagonist). In the presence of lorglumide (a selective CCK-A receptor antagonist), CCK-8-induced excitatory responses were inhibited. No cells responded to the administration of non-sulfated CCK-8 (CCK-8NS, a selective CCK-B receptor agonist). We conclude that CCK-8 exerts its action via presynaptic CCK-A receptors to facilitate glutamate release onto Ih-negative area postrema cells. - Investigation of neuronal functions in the area postrema : functional differentiation between neurons related to the feeding regulation and neurons triggering nausea and/or emesis

English, Scientific journal, Studies have shown that exercise can enhance learning and memory. Conditioned taste aversion (CTA) is an avoidance behavior induced by associative memory of the taste sensation for something pleasant or neutral with a negative visceral reaction caused by the coincident action of a toxic substance that is tasteless or administered systemically. We sought to measure the effects of treadmill exercise on CTA in rats by investigating the effects of exercise on acquisition, extinction and spontaneous recovery of CTA. We made two groups of rats: an exercise group that ran on a treadmill, and a control group that did not have structured exercise periods. To condition rats to disfavor a sweet taste, consumption of a 0.1% saccharin solution in place of drinking water was paired with 0.15M LiCl (2% body weight, i.p.) to induce visceral discomfort. We measured changes of saccharin consumption during acquisition and extinction of CTA. The exercise and no-exercise groups both acquired CTA to similar levels and showed maximum extinction of CTA around 6 days after acquisition. This result indicates that exercise affects neither acquisition nor extinction of CTA. However, in testing for preservation of CTA after much longer extinction periods that included exercise or not during the intervening period, exercising animals showed a significantly lower saccharin intake, irrespective of having exercised or not during the conditioning phase of the trial. This result suggests that exercise may help to preserve aversive memory (taste aversion in this example) as evidence by the significant spontaneous recovery of aversion in exercising animals. - Somatosensory evoked magnetic fields following tongue and hard palate stimulation on the preferred chewing side

English, Scientific journal, Amylin, which is co-secreted together with insulin by pancreatic beta cells, is considered to be an important peptide hormone involved in the control of feeding behavior and energy homeostasis. Although the area postrema has been implicated to be a primary target of amylin, there are no studies of the mechanisms by which amylin may alter the excitability of area postrema neurons. To investigate the mechanism for amylinergic modulation of neuronal excitability, we performed perforated patch-clamp recordings from area postrema neurons in rat brainstem slices. Amylin-induced changes in excitatory responses, such as increases in the frequency of mEPSCs (miniature excitatory postsynaptic currents) and changes in the amplitude distribution of mEPSCs, were found in cells not displaying the hyperpolarization-activated cation current (I(h)). Area postrema cells displaying I(h) did not respond to amylin application. Inhibitory responses to amylin were never encountered. Bath application of CNQX (AMPA type glutamate receptor antagonist) abolished the effects of amylin. Depolarization of cells during amylin application was sufficient at 1 μM to cause action potential discharge by responding cells. We conclude that amylin receptors are located mostly on presynaptic glutamatergic terminals connecting to the area postrema neurons not displaying I(h) and amylin concentrations can increase glutamate release enough to cause cell firing. Japanese, ラットの新鮮脳海馬スライスにGABA A受容体拮抗薬ピクロトキシンを投与しててんかん様の自然発火を惹起させ,これにプロポフォールを投与したところ,濃度依存的に自然発火の頻度,持続時間が減少した.又,Schaffer側枝の電気刺激で惹起したてんかん様バースト発火に対しても,プロポフォールは濃度依存的に持続時間を減少させた.更にピクロトキシン及びAMPA型グルタミン酸受容体拮抗薬CNQX存在下でSchaffer側枝の電気刺激により誘発した興奮性シナプス後電位に対し,プロポフォール投与で振幅,持続時間は減少した.ピクロトキシン及びNMDA型グルタミン酸受容体拮抗薬CPP存在下での誘発電位に対してもプロポフォール投与は電位の持続時間を減少させた.次に細胞内パルス通電により生ずる海馬CA1ニューロンの膜電位変化に対し,プロポフォールを投与したところ,これらの膜電位変化は著明に減少した.プロポフォールが過分極作動性内向き電流(H電流)を抑制し,膜電位の過分極,膜抵抗の増加を引き起こすことが示された - ラット上唾液核細胞における抑制性シナプス伝達はGABAB受容体を介するシナプス前抑制を受けている

2019年度の研究実績の概要について,実施した研究計画,結果,考察,新たな問題点について下記に述べる.
ラットを用いた実験において甘味摂取直後に催吐物質を腹腔内注射すると,甘味(条件刺激)と悪心(無条件刺激)との連合学習が成立して甘味摂取を忌避するようになる.これを条件付け味覚嫌悪(CTA:conditioned taste aversion)と呼び,本研究ではこれを薬物投与による悪心誘発の指標として用いた.催吐性のある薬物としてシスプラチン(抗がん剤),エメチン(吐根の成分),トラネキサム酸を比較した.こららの薬物をコントロール群のラットに投与すると,著明なCTAの獲得を認めた.実験群として最後野切除群,迷走神経求心路切除群,最後野と迷走神経求心路の両方切除群を作成し,シスプラチン,エメチン,トラネキサム酸の腹腔投与を無条件刺激にした場合のCAT獲得を調べたところ,迷走神経求心路切除群はエメチン投与によってCTAを獲得したが,最後野切除群においてはエメチン投与によるCTA獲得は抑制された.この結果はエメチン誘発の悪心が最後野を介して惹起されることを示唆するものであった.シスプラチンについては,最後野切除群,迷走神経求心路切除群,最後野と迷走神経求心路の両方切除群のいずれにおいてもCTAの獲得を認めたため,エメチンとは異なり迷走神経や最後野を介さない経路が悪心誘発に関与していることが示唆された.また,トラネキサム酸の腹腔投与による悪心誘発は迷走神経求心路切除群で条件付けテスト日2日目におけるCTA獲得が抑制され,最後野切除群においては条件付けテスト日1日目からCTA獲得が強く抑制されたことから,迷走神経求心路と最後野の両方を介して悪心が誘発されていることが示唆された.橋と延髄より上位脳における悪心誘発機序を考慮することが全貌解明に必要であると考えられた.

As an intracellular mechanism for glucose responsiveness of the area postrema neurons, it was found that the ATP-dependent potassium channel (KATP channel) was closed by an increase in the extracellular glucose concentration, leading to the generation of depolarization and action-potentials. The glucose-responsive neurons were suggested to be H-channel non-expressing cells. The neuronal activity of the area postrema is crucial for the inducing mechanism of emetine-induced nausea, and it was also found that depolarization occurs in the central neurons including the nucleus tractus solitarius, the central nucleus of amygdala, the bed nucleus. It became clear that the memory recall of the taste aversion did not involve the activity of the nervous system related to a nausea induction.

Glucagon-like peptide-1 (GLP-1) is released postprandially from the gut and known as a satiety peptide. However, it’s unclear whether peripheral GLP-1 can directly reach receptors in the brain. To examine how peripheral GLP-1 reaches into the brain, we analyzed the c-Fos, a marker of neuronal activation, expression in the brainstem neurons after intraportal injection of GLP-1. Physiological concentration of GLP-1 induced c-Fos expression of nucleus tract solitarius neurons, but not of area postrema neurons. These results suggest the GLP-1 secreted from the gut might reach the brain as a signal via vagal nerve rather than as a hormone via the bloodstream. These experiments were used transgenic rat expressing green fluorescent protein (GFP) under control of a c-fos promoter. Therefore, c-Fos-expressing neurons were identifiable by GFP. This experimental design might be applicable to the identification of secondary neurons for satiety- or hunger-signal of peripheral peptides.

We investigated how neurons in the area postrema known as a chemoreceptor trigger zone are related to the induction of nausea and/or emesis and to the control of food intake. We found that the neurons expressing H channels are closely related to the induction of nausea but they do not respond to amylin that is known as a satiety peptide. Responses to amylin were found only in the neurons not expressing H channels. We concluded that these date suggested the presence of functional differentiation between the neurons expressing H channels and the neurons not expressing H channels.

Salivary glands of rats fed a liquid diet for 3 weeks were investigated microscopically to clarify the effects of soft food on salivary glands. Parotid glands of experimental rats became atrophic after liquid diet feeding, and cell shrinkage, cell death, and reduction of speed of cell-cycle were identified in the atrophic parotid glands. However, such atrophic alterations were not observed in submandibular and sublingual glands following liquid diet feeding. These observations suggest that it depends on the kind of salivary glands whether liquid diet feeding induces atrophy to the salivary glands.

Postoperative nausea and vomiting is one of the most common complications, but its mechanism is unknown. In the present study, we studied the c-Fos expression in the area postrema for the purpose of investigating whether intraoperative hypotension causes postoperative nausea and vomiting. Hypotension significantly increased c-Fos expression in the area postrema of the rats received isoflurane or propofol anesthesia. Although we could not confirm the evidence by behavioral experiments, it is possible that nervous excitement in the area postrema by intraoperative hypotension causes nausea and vomiting.

This study elucidated the membrane properties, morphology and chemosensitivities of area postrema neurons using a patch-clamp technique in rat brain slices. 本研究は,健康成人ボランティアを対象に,静脈内鎮静法によって引き起こされると考えられる,嚥下に関連した中枢神経活動への影響を,機能的磁気共鳴画像(機能的MRI)を用いることによって評価することが,目的であった。平成14年度の研究結果として,大脳皮質運動野において嚥下によって賦活する部位の神経活動が,静脈内鎮静法によって抑制される傾向がみられた。さらに前年度(平成15年度)の研究結果から,静脈内鎮静法による高位の中枢神経における嚥下動作の抑制は,静脈麻酔薬の用量依存性であることが示唆された。これらの結果をもとに,本年度は静脈麻酔薬の血中濃度と神経活動との関連を調べること,さらに,大脳皮質運動野だけでなく,脳内の各賦活領域に対して,静脈麻酔薬がどのような影響を及ぼすかを検討した。方法はこれまでと同様に,嚥下動作課題として,遠隔操作によってシリンジから1回1.5mlの蒸留水を20秒間隔で10回投与し,MR画像撮影は前交連〜後交連平面を基準に頭頂部から脳底部までとし,嚥下動作中に2秒間隔で100回撮像した。神経活動の評価として,嚥下動作課題を与え,嚥下時の機能的MRI画像から非実行時のMRI画像を差し引き,MR信号強度を算出し,T1強調画像と重ね合わせることによって,解剖学的脳賦活部位を同定した。静脈麻酔薬であるプロポフォールを最初の5分間に6mg/kg/hまたは4mg/kg/hの速...

The superior salivatory (SS) nucleus contains parasympathetic preganglionic neurons innervating the submandibular and sublingual salivary glands and those innervating the anterior part of the tongue. 海馬体の内嗅野深層の電気刺激に誘発されるバースト発火の扁桃体外側核への伝播について,両部位を含む水平断ラット脳スライスを用いて,ガラス微小電極による細胞内記録法と金属電極によるフィールド電位記録により解析を行った。扁桃体の単一ニューロンのうち内嗅野深層ニューロンのバースト発火と同期して活動するものは主に錘体細胞に検出された。扁桃体の単一ニューロンが示した誘発電位は次の4タイプに分類された。1)内嗅野のバースト発火にほぼ相似形なバースト発火でガンマ波を含むもの,2)刺激後50msの間のみ持続する200ヘルツ程度の発火頻度を示すバースト発火,3)1発の活動電位のみ発火するもの,4)活動電位の発火は見られず興奮性シナプス後電位のみが観察されるものであった。こららの誘発電位は全て近傍のフィールド電位と同期して観察された。このことは2つの細胞から同時細胞内記録を行った実験データを用いた相関解析によっても,扁桃体外側核内の2ニューロンの顕著な活動同期性が明らかとなった。また,活動電位の発火を伴わない誘発電位を示した扁桃体外側核ニューロンはGABA-A受容体のアンタゴニストであるピクロトキシンの潅流液内への投与によってもその応答様式には変化が見られず,内嗅野からのシナプス入力自体に依存していることが示唆された。さらに,これらの誘発電位がAMPA型およびNMDA型の両方のグルタミン酸受容体を...

扁桃体の単一ニューロンの電気生理学的および形態学的特性を調べるとともに,海馬体のうち特に内嗅野の深層の電気刺激に誘発される鋭波(200Hz)およびガンマ波(40-100Hz)(Funahashi and Stewart,J.Physiol.510(1)191-208,1998)と扁桃体ニューロン活動の相互関係について検討を行った。扁桃体の諸核うち外側核,基底外側核および中心核の単一ニューロンについて細胞内記録を用いて脱分極性の細胞内通電(〜1mA,200ms)に対する発火パターンを調べた結果,殆どの外側核ニューロンが一般的に見られる連続発火を示し最初の数発のスパイク間隔が短いのが特徴的であった。また,基底外側核のニューロンでは強い後電位を伴うバースト発火が多く見られ,中等度の活動電位の順応が見られた。一方,中心核ニューロンでは大部分に著しい活動電位の順応を伴うバースト発火が観察された。内嗅野の電気刺激に対する応答は外側核および基底外側核のみに見られ,これらは内嗅野の鋭波およびガンマ波を含むバースト発火に同期するもので,誘発電位にIPSPが強く見られないニューロンに多く観察された。記録終了後にニューロバイオチントレーサーを注入しておいたニューロンについては記録部位の確認を行い,また細胞体および樹上突起の形態,シナプス突起の有無による形態学的分類を行った。外側核および基底外側核ニ...

脳幹レベルでの摂食飲水行動の調節について調べるために、当初、最後野のグルコース応答性ニューロンについてスライスパッチクランプ法を用いて調べる予定であったが、細胞の大きさや数などの点で有利な近接の孤束核ニューロンからの記録を行った。孤束核と最後野は密な線維連絡を持ち、共に内臓感覚の一次求心路が投射している部位としてよく知られている。本実験では孤束核ニューロンの浸透圧応答性について、最後野からの入力がある場合とない場合とで比較検討した。これによると、孤束核ニューロンは直接電気刺激によってタイプ1、2、3の三種類の発火パターンに分類できた。記録した42ニューロン中、10ニューロンがNaClによる浸透圧増加とsucroseによる浸透圧増加の両方に対して放電頻度の増加を示した。また1ニューロンが両方の浸透圧増加にたいして放電頻度の減少を示した。残りの31ニューロンは浸透圧刺激に対して何の応答も示さなかった。浸透圧応答性を示した11ニューロン中4ニューロンは[Ca^<2+>]free,high[Mg^<2+>]の条件下にて同様の実験を行った。いわゆるシナプスを遮断した状態で、3ニューロンは浸透圧応答性を保ち、1ニューロンは浸透圧応答性を失った。つまり、孤束核にはそれ自身が浸透圧受容性を持つものが存在することが明らかとなった。浸透圧応答性を失った1ニューロンはおそらく最後野からの入力を受け...

Responses of neurons within the nucleus taractus solitarius NTS) to gastric distension were analyzed. ラットの第IV脳室底を備品で購入したハ-バ-ド940シリンジポンプを用いてリンゲル液で表面灌流した。微小電極を用いて最後野中のニュ-ロンの電気活動を記録し、灌流液をグルコ-スを含む試験液に切り換えて、神経応答を記録した。その結果、グルコ-スに応答して放電頻度の増加するもの(グルコ-ス受容性ニュ-ロン)と、減少するもの(グルコ-ス感受性ニュ-ロン)と、2種類の異なったニュ-ロンが最後野中に存在した。両者はほぼ同数存在し、最後野中での部位的局在は認められず混在した。これらのニュ-ロンは同じ単糖類であるマンノ-スに対しては応答しないから、グルコ-スに特異的に応答することがわかった。つぎに受容機構を解明するため、2DG、ウアバイン、およびフロリジンを、それぞれ含む試験液を調製し、グルコ-ス受容性ニュ-ロン、およびグルコ-ス感受性ニュ-ロンがこれら試験液に対してどのように応答するかを検討した。グルコ-ス受容性ニュ-ロンは神経細胞膜を通過しないフロリジンに対して応答するから、ニュ-ロン膜にグルコ-スを受容する部位が存在するものと思われる。一方、グルコ-ス感受性ニュ-ロンは、グルコ-スに対しては放電頻度の減少を生じるが、2DGに対しては放電頻度が逆に増加することから考えて、従来、外側視床下野で明らかにされているグルコ-ス感受性ニュ-ロンと同様、グルコ-スが細胞内にとりこまれ、グルコ-ス代謝...
