  • Low-CO2-inducible bestrophins outside the pyrenoid sustain high photosynthetic efficacy in diatoms

    Anion transporters sustain a variety of physiological states in cells. Bestrophins belong to a Cl− and/or HCO3− transporter family conserved in bacteria, animals, algae, and plants. Recently, putative bestrophins were found in the green alga Chlamydomonas reinhardtii, where they are up-regulated under low CO2 conditions and play an essential role in the CO2-concentrating mechanism (CCM). The putative bestrophin orthologs are also conserved in diatoms, secondary endosymbiotic algae harboring red-type plastids, but their physiological functions are unknown. Here, we characterized the subcellular localization and expression profile of bestrophins (BSTs) in the marine diatoms Phaeodactylum tricornutum (PtBST1−4) and Thalassiosira pseudonana (TpBST1 and 2). PtBST1, PtBST2, and PtBST4 localized at the stroma thylakoid membrane outside of the pyrenoid, and PtBST3 localized in the pyrenoid. Contrarily, TpBST1 and TpBST2 both localized in the pyrenoid. These bestrophin proteins accumulated in cells grown in atmospheric CO2 (LC) but not in 1% CO2 (HC)-grown cells. To assess the physiological functions, we generated knock-out mutants for the PtBST1gene by genome editing. The lack of PtBST1 decreased photosynthetic affinity for dissolved inorganic carbon to the level comparable to the HC-grown wild type. Furthermore, non-photochemical quenching in LC-grown cells was 1.5–2.0 times higher in the mutants than in the wild type. These data suggest that HCO3− transport at the stroma thylakoid membranes by PtBST1 is a critical part of the CO2-evolving machinery of the pyrenoid in the fully induced CCM and that PtBST1 may modulate photoprotection under CO2-limited environments in P. tricornutum.
  • Pyrenoid proteomics reveals independent evolution of the CO2-concentrating organelle in chlorarachniophytes.

    English, Scientific journal, Pyrenoids are microcompartments that are universally found in the photosynthetic plastids of various eukaryotic algae. They contain ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and play a pivotal role in facilitating CO2 assimilation via CO2-concentrating mechanisms (CCMs). Recent investigations involving model algae have revealed that pyrenoid-associated proteins participate in pyrenoid biogenesis and CCMs. However, these organisms represent only a small part of algal lineages, which limits our comprehensive understanding of the diversity and evolution of pyrenoid-based CCMs. Here we report a pyrenoid proteome of the chlorarachniophyte alga Amorphochlora amoebiformis, which possesses complex plastids acquired through secondary endosymbiosis with green algae. Proteomic analysis using mass spectrometry resulted in the identification of 154 potential pyrenoid components. Subsequent localization experiments demonstrated the specific targeting of eight proteins to pyrenoids. These included a putative Rubisco-binding linker, carbonic anhydrase, membrane transporter, and uncharacterized GTPase proteins. Notably, most of these proteins were unique to this algal lineage. We suggest a plausible scenario in which pyrenoids in chlorarachniophytes have evolved independently, as their components are not inherited from green algal pyrenoids.

    English, Scientific journal, Spermatogenesis is one of the most dramatic changes in cell differentiation. Remarkable chromatin condensation of the nucleus is observed in animal, plant, and algal sperm. Sperm nuclear basic proteins (SNBPs), such as protamine and sperm-specific histone, are involved in chromatin condensation of the sperm nucleus. Among brown algae, sperm of the oogamous Fucales algae have a condensed nucleus. However, the existence of sperm-specific SNBPs in Fucales algae was unclear. Here, we identified linker histone (histone H1) proteins in the sperm and analyzed changes in their gene expression pattern during spermatogenesis in Sargassum horneri. A search of transcriptomic data for histone H1 genes in showed six histone H1 genes, which we named ShH1.1a, ShH1b, ShH1.2, ShH1.3, ShH1.4, and ShH1.5. Analysis of SNBPs using SDS-PAGE and LC-MS/MS showed that sperm nuclei contain histone ShH1.2, ShH1.3, and ShH1.4 in addition to core histones. Both ShH1.2 and ShH1.3 genes were expressed in the vegetative thallus and the male and female receptacles (the organs producing antheridium or oogonium). Meanwhile, the ShH1.4 gene was expressed in the male receptacle but not in the vegetative thallus and female receptacles. From these results, ShH1.4 may be a sperm-specific histone H1 of S. horneri.

    English, Scientific journal, The first mitotic division of the initial cell is a key event in all multicellular organisms and is associated with the establishment of major developmental axes and cell fates. The brown alga Ectocarpus has a haploid-diploid life cycle that involves the development of two multicellular generations: the sporophyte and the gametophyte. Each generation deploys a distinct developmental programme autonomously from an initial cell, the first cell division of which sets up the future body pattern. Here, we show that mutations in the BASELESS (BAS) gene result in multiple cellular defects during the first cell division and subsequent failure to produce basal structures during both generations. BAS encodes a type B″ regulatory subunit of protein phosphatase 2A (PP2A), and transcriptomic analysis identified potential effector genes that may be involved in determining basal cell fate. The bas mutant phenotype is very similar to that observed in distag (dis) mutants, which lack a functional Tubulin-binding co-factor Cd1 (TBCCd1) protein, indicating that TBCCd1 and PP2A are two essential components of the cellular machinery that regulates the first cell division and mediates basal cell fate determination.

    English, Scientific journal, In brown algae, the extracellular matrix (ECM) and its constitutive polymers play crucial roles in specialized functions, including algal growth and development. In this review we offer an integrative view of ECM construction in brown algae. We briefly report the chemical composition of its main constituents, and how these are interlinked in a structural model. We examine the ECM assembly at the tissue and cell level, with consideration on its structure in vivo and on the putative subcellular sites for the synthesis of its main constituents. We further discuss the biosynthetic pathways of two major polysaccharides, alginates and sulfated fucans, and the progress made beyond the candidate genes with the biochemical validation of encoded proteins. Key enzymes involved in the elongation of the glycan chains are still unknown and predictions have been made at the gene level. Here, we offer a re-examination of some glycosyltransferases and sulfotransferases from published genomes. Overall, our analysis suggests novel investigations to be performed at both the cellular and biochemical levels. First, to depict the location of polysaccharide structures in tissues. Secondly, to identify putative actors in the ECM synthesis to be functionally studied in the future.
  • Ultrastructural Observation of Cytokinesis and Plasmodesmata Formation in Brown Algae.

    English, Scientific journal, Similar to land plant cells, brown algal cells possess plasmodesmata with minute cytoplasmic tunnels, which enable the direct connection between adjacent cells. Plasmodesmata are distributed depending on the association of their formation with cytokinesis. Primary plasmodesmata are formed during cytokinesis, while secondary plasmodesmata appear on the cell wall septum following cytokinesis. Typically, the plasmodesmata of brown algae are cylindrical without the penetration of desmotubules from the endoplasmic reticulum, and there are no morphological differences between primary and secondary plasmodesmata. This present chapter describes the observation of cytokinesis and primary plasmodesmata formation in brown algae using electron microscopy as well as the examination of polysaccharide distribution using antibodies and enzyme-gold probes.
  • Targeted CRISPR-Cas9-based gene knockouts in the model brown alga Ectocarpus.

    English, Scientific journal, Brown algae are an important group of multicellular eukaryotes, phylogenetically distinct from both the animal and land plant lineages. Ectocarpus has emerged as a model organism to study diverse aspects of brown algal biology, but this system currently lacks an effective reverse genetics methodology to analyse the functions of selected target genes. Here, we report that mutations at specific target sites are generated following the introduction of CRISPR-Cas9 ribonucleoproteins into Ectocarpus cells, using either biolistics or microinjection as the delivery method. Individuals with mutations affecting the ADENINE PHOSPHORIBOSYL TRANSFERASE (APT) gene were isolated following treatment with 2-fluoroadenine, and this selection system was used to isolate individuals in which mutations had been introduced simultaneously at APT and at a second gene. This double mutation approach could potentially be used to isolate mutants affecting any Ectocarpus gene, providing an effective reverse genetics tool for this model organism. The availability of this tool will significantly enhance the utility of Ectocarpus as a model organism for this ecologically and economically important group of marine organisms. Moreover, the methodology described here should be readily transferable to other brown algal species.
  • Changes in Cell Wall Structure During Rhizoid Formation of Silvetia babingtonii (Fucales, Phaeophyceae) Zygotes.

    English, Scientific journal, We examined the ultrastructure of the cell wall and immunolocalization of alginates using specific antibodies against M-rich alginates and MG blocks during rhizoid formation in fucoid zygotes, Silvetia babingtonii. The thallus region of 24-h-old zygotes had a cell wall made of three layers with different fiber distribution. In the 12-h-old zygotes, three layers in the thallus were observed before rhizoid formation, namely the inner, middle, and outer layers. During rhizoid elongation, only the inner layer was apparent close to the rhizoid tip area. Immunoelectron microscopy detected M-rich blocks of alginate on the inner half of the cell wall, irrespective of the number of layers in the thallus and rhizoid regions. The MG blocks were seen to cover a slightly wider area than M-rich alginate blocks. It was suggested that parts of M in mannuronan would be rapidly converted to G, and MG-blocks are generated. Transcriptome analysis was performed using 3 -, 10 -, and 24-h-old zygotes after fertilization to examine the relationship between gene expression and alginate synthesis over time. The expression of two mannuronan C5-epimerase homologs that convert mannuronic acid into guluronic acid in alginates was upregulated or downregulated over the course of the examination.
  • Ultrastructural observations of mitochondrial morphology through the life cycle of the brown alga, Mutiomo cylindricus (Cutleriaceae, Tilopteridales).

    English, Scientific journal, Mitochondrial morphology varies according to development and the physiological conditions of the cell. Here, we performed electron tomography using serial sections to analyze the number, individual volume, and morphological complexity of mitochondria in the cells across two generations in the life cycle of the brown alga Mutimo cylindricus. This species shows a heteromorphic alternation of generations between the macroscopic gametophyte and the crustose sporophyte during its life cycle and displays anisogamous sexual reproduction. We observed the mitochondria in the vegetative cells of gametophytes and sporophytes to mainly show tubular or discoidal shapes with high morphological complexity. The morphology of the mitochondria in the male and female gametes changed to a nearly spherical or oval shape from a tubular or discoidal shape before release. In this species, degradation of the paternal mitochondria was observed in the zygote 2 h after fertilization. Morphological changes in the mitochondria were not observed until 6 h after fertilization. Twenty-four-hour-old zygotes before and after cytokinesis showed a similar number of mitochondria as 6-h-old zygotes; however, the volume and morphological complexity increased. The results indicated that the maternal mitochondria did not undergo fission or fusion until this stage. Based on the analysis results of the number and total volume of mitochondria before and after the release of the gametes, it is possible that the mitochondria in the female gametes fuse immediately before release.

    English, Scientific journal, Based on the morphology of gametes, sexual reproduction in brown algae is usually classified into three types: isogamy, anisogamy, and oogamy. In isogamy, chloroplasts and chloroplast DNA (chlDNA) in the sporophyte cells are inherited biparentally, while mitochondria (or mitochondrial DNA, mtDNA) is inherited maternally. In oogamy, chloroplasts and mitochondria are inherited maternally. However, the patterns of mitochondrial and chloroplast inheritance in anisogamy have not been clarified. Here, we examined derivation of mtDNA and chlDNA in the zygotes through strain-specific PCR analysis using primers based on single nucleotide polymorphism in the anisogamous brown alga Mutimo cylindricus. In 20-day-old sporophytes after fertilization, mtDNA and chlDNA derived from female gametes were detected, thus confirming the maternal inheritance of both organelles. Additionally, the behavior of mitochondria and chloroplasts in the zygotes was analyzed by examining the consecutive serial sections using transmission electron microscopy. Male mitochondria were isolated or compartmentalized by a double-membrane and then completely digested into a multivesicular structure 2 h after fertilization. Meanwhile, male chloroplasts with eyespots were observed even in 4-day-old, seven-celled sporophytes. The final fate of male chloroplasts could not be traced. Organelle DNA copy number was also examined in female and male gametes. The DNA copy number per chloroplast and mitochondria in male gametes was lower compared with female organelles. The degree of difference is bigger in mtDNA. Thus, changes in different morphology and DNA amount indicate that maternal inheritance of mitochondria and chloroplasts in this species may be based on different processes and timing after fertilization.
  • Organelle inheritance and genome architecture variation in isogamous brown algae.

    English, Scientific journal, Among the brown algal lineages, Ectocarpales species have isogamous fertilization in which male and female gametes are morphologically similar. In contrast, female gametes are much larger than male gametes in the oogamous species found in many other brown algal lineages. It has been reported that the plastids of isogamous species are biparentally inherited whereas the plastids of oogamous species are maternally inherited. In contrast, in both isogamous and oogamous species, the mitochondria are usually inherited maternally. To investigate whether there is any relationship between the modes of inheritance and organellar genome architecture, we sequenced six plastid genomes (ptDNA) and two mitochondrial genomes (mtDNA) of isogamous species from the Ectocarpales and compared them with previously sequenced organellar genomes. We found that the biparentally inherited ptDNAs of isogamous species presented distinctive structural rearrangements whereas maternally inherited ptDNAs of oogamous species showed no rearrangements. Our analysis permits the hypothesis that structural rearrangements in ptDNAs may be a consequence of the mode of inheritance.

    English, Scientific journal, Studies of postglacial range shifts could enhance our understanding of seaweed species' responses to climate change and hence facilitate the conservation of natural resources. However, the distribution dynamics and phylogeographic diversification of the commercially and ecologically important kelp Saccharina japonica in the Northwest Pacific (NWP) are still poorly surveyed. In this study, we analyzed the evolutionary history of S. japonica using two mitochondrial markers and 24 nuclear microsatellites. A STRUCTURE analysis revealed two partially isolated lineages: lineage H, which is scattered along the coast of Japan; and lineage P, which occurs along the west coast of the Japan Sea. Ecological niche modeling projections to the Last Glacial Maximum (LGM) revealed that the southern coasts of the Japan Sea and the Pacific side of the Oshima and Honshu Peninsulas provided the most suitable habitats for S. japonica, implying that these regions served as ancient refugia during the LGM. Ancient isolation in different refugia may explain the observed divergence between lineages P and H. An approximate Bayesian computation analysis indicated that the two lineages experienced post-LGM range expansion and that postglacial secondary contact occurred in Sakhalin. Model projections into the year 2,100 predicted that S. japonica will shift northwards and lose its genetic diversity center on the Oshima Peninsula in Hokkaido and Shimokita Peninsula in Honshu. The range shifts and evolutionary history of S. japonica improve our understanding of how climate change impacted the distribution range and diversity of this species and provide useful information for the conservation of natural resources under ongoing environmental change in the NWP.

  • 珪藻のウイルス弱毒化因子は,珪藻個体群の維持システムとして機能するのか?

    「海の牧草」と称され、海洋資源生物として重要な『珪藻』は、常に強毒ウイルスによる死滅の危機に曝されている。ところが我々研究チームは、珪藻に感染する強毒ウイルスの働きを抑える『弱毒化因子』を発見した。この弱毒化因子は、珪藻 vs 強毒ウイルスの関係性に関与して、珪藻細胞内での強毒ウイルス複製を抑制すると考えられる。さらに、強毒ウイルス蔓延を抑え、珪藻個体群が生残する生態システムの一つとして機能している可能性も考えられる。本研究では、地球上で巨大生産力を誇る珪藻が、強毒ウイルス感染による死滅から回避する戦略の一端として、この弱毒化因子が関与しているのかを生理生態学的に理解することが目的である。
    令和3年度は、弱毒化因子の関与によって、珪藻 vs 強毒ウイルスの関係に生じる変化(課題1:形質・発現遺伝子の変化、課題2:構造・形態の変化)を室内実験で解明することに主に取り組んだ。強毒ウイルス株に混在する弱毒化因子を探索するため、DNAウイルス株に存在する弱毒化因子DNA配列を高感度nested PCRにて検出した。その結果、274株のDNAウイルス株中29株から未知DNA因子を検出した。これらの株について、弱毒化因子が存在するウイルス株の違いによって、弱毒化因子DNA配列がどのように異なるのかを解析した。その解析の結果、弱毒化因子は珪藻DNAウイルスと関連を持って存在しており、機能的に保存されるORF領域と、強毒ウイルスに依存するDNA領域の2つを持っていることが示唆された。また、今後研究で使用する弱毒化因子を特異的に認識する抗血清を作成した。
    なお、令和3年度では、成果に基づいて国内学会発表(日本藻類学会1件)を実施した。

  • Analysis of membrane fusion process and related proteins on cytokinesis of brown algae

    In this study, a three dimensional analysis was performed by electron tomography in zygotes of Silvetia babingtonii, for understanding the transitional membrane configuration during cytokinesis. The analysis revealed that a complicated architecture with fringes (TA; tubule-like appendage) were formed by fusion of Golgi derived vesicles to flat cisternae (FCs). Each FC was connected by TAs, thus forming the membranous network (MN). At advanced cytokinesis, the MN grew into broad membranous sac (MS). To turn the 3D image to arbitrary angle indicated a stratification of membrane fusion process from the MN to the MSc. Moreover, proteins extracted from zygotes of Fucus distichus on cytokinesis and cytokinesis blocked zygotes treated with latrunculin B were separated by two-dimensional gel electrophoresis (2-DE). From a total of about 600 spots reproducibly separated on gels, 2-DE from cytokinesis blocked zygotes showed 31 under-expressed and 2 over-expressed proteins in comparison with control.

  • ヌクレオモルフの自律性から探る葉緑体二次共生藻類の成立過程

    葉緑体二次共生植物(藻類)であるクリプト藻とクロララクニオン藻では、葉緑体となった真核藻類の核の名残りであるヌクレオモルフが観察できる。昨年度は、海産クリプト藻Pyrenomonas helgolandiiについて葉緑体・ヌクレオモルフ・核のDNA合成時期・分裂時期を特定した。その結果は、ヌクレオモルフは1回だけのDNA合成をホストの核分裂直前の2-4時間の間に行うという極めて真核的なものであった。本年度はクリプト藻とは系統が異なるクロララクニオン藻において同様に調査する目的で、Partenskyella glossopodiaの核分裂について電子顕微鏡観察を行った。本種の直径は1.5-3μmと極めて小さく、核とピレノイドを持たない葉緑体が1個ずつという単純な構造であるために連続切片による観察が容易である。その結果、まず葉緑体が分裂し、次にヌクレオモルフが分裂し、細胞にホスト核が分裂することが明らかになった。また、今回の観察と今までの報告からクロララクニオン藻内での核分裂の多様性も示された。次にBrdUの取り込み実験を行ったが、クリプト藻Pyrenomonas helgolandiiと異なり本種ではBrdUの取り込みが確認できなかったためヌクレオモルフにおける正確なDNA合成時期の特定はできなかった。しかし、ホスト核とヌクレオモルフの分裂時期を考えるとクリプト藻の場合と同様に真核的なDNA合成制御を行っているものと推察できた。

  • Studies on recognition system of male and female gametes and cytoplasmic inheritance of organelles during fertilization and zygote development of algae

    Cytoplasmic inheritance of chloroplasts and mitochondria during fertilization of brown algae were studied by electron microscopy and molecular biological techniques using Scytosiphon lomentaria (isogamy), Unidaria pinnatifica and Cystoseira hakodatensis (oogamy). Chloroplasts were derived from both parents in S. lomentaria, while mitochondrial DNA derived from male gametes became selectively disappeared from four-celled sporophytes. In the case of oogamous U. pinnatifida, sperm chloroplasts and mitochondria were digested by lysosome soon after fertilization. In C. hakodatensis, differences of the organellar DNA methylation degrees could be clearly detected between eggs and sperm.

    不等分裂を行う褐藻類の受精卵の最初の細胞質分裂がどのように進行していくか,アクチンはどのような働きをもっているのかについて微細構造レベルで明らかにするために,急速凍結置換法という固定法を用いて透過型電子顕微鏡で詳細に観察を行った。その結果,アクチンは細胞質分裂面に供給される平板小嚢と呼ばれる構造の形成に関与していることが明らかになった一方,細胞質分裂面の決定には直接関与していないことが示唆された。

  • 雄性配偶子由来セントリオールの鞭毛基底小体から中心体への機能転換に関する研究

    本年度は、昨年度に引き続き、同形配偶子接合を行なう褐藻カヤモノリの配偶子、接合子を用いて、鞭毛基底小体から中心体へと機能転換する際の中心体構成既知タンパク質の局在変化(ex.セントリン、γチューブリン)について、抗体を用いた蛍光顕微鏡、及び、免疫電子顕微鏡法での観察を行なった。セントリンに関しては、鞭毛基底小体と中心体として機能する際の各々で、セントリオールへの局在箇所に変化が生じている可能性が十分に示唆された。しかしながら、γチューブリンに関しては、蛍光顕微鏡、および、電子顕微鏡での局在観察に関して、市販の抗γチューブリン抗体(SIGMA社、ポリクローナル抗体)では、ウェスタンブロットでの解析、および間接蛍光顕微鏡においては陽性反応を示すものの、電子顕微鏡用の試料上では、非特異的な反応が著しかったこともあり、局在変化を微細構造レベルでとらえることは困難であった。
    そのような理由もあり、褐藻カヤモノリのγチューブリンに由来する抗体を作製するため、遺伝子のクローニングを試みることとした。また、同時に、セントリオールを構築するトリプレット微小管に存在し、その構造維持に関係していることが知られている新規チューブリンと呼ばれるε(イプシロン)チューブリン遺伝子のクローニングも試みた。γチューブリンに関しては、生物間で保存性の高いアミノ酸配列部位をもとにプライマーを設計し、εチューブリンに関しては、ゲノムプロジェクトの一貫として進められている褐藻シオミドロのEST解析の情報を利用することとしてRT-PCRを行なった。εチューブリンに関しては、700bpの断片配列を得ることができ、現在、全長配列のクローニングを進めている。γチューブリンに関しては、その局在性の解析からも、細胞周期特異性が非常に強いと予想され、発現に偏りがあるため、継続して遺伝子の単離を行なっている。

  • Molecular and cell biological study on the heterogeneity of flagella in heterokont algae

    Heterokont algae usually have a long flagellum attached with mastigonemes and a short one with the basal swelling part, and the structure and the function of these flagella are quite different. In this study, we studied the heterogeneity of two flagella in heterokont algae, especially focusing on the process of their elongation and differentiation using the chrysophycean alga Ochromonas danica and the brown alga Ectocarpus siliculosus. Flagellar duplication in Ochromonas was observed in detail by the time-lapse video microscopy. Moreover, mitosis was observed by electron and immunofluorescence microscopy. Mitosis of Ochromonas is the open spindle type, and the rhizoplast, which is the striated structure from the flagellar basal bodies, is located at both poles. From studies using various antibodies, we found that the rhizoplast of Ochromonas was composed of gannma-tubulin, not centrin. This result shows that the rhizoplast, which can be seen in motile cells in various eukaryotic algal groups, is not homologous, especially on its constituted proteins. In zoosporogenesis in the plurilocular zoidangia of Ectocarpus siliculosus, we examined the process of the flagellar elongation and differentiation using cryofixation under electron microscopy. Both flagella began to elongate simultaneously, while we could detect on which of the flagella is long or short by the difference of the diameter and the associated flagellar rootlets. Interestingly, addition of small vesicles to the flagellar membrane from the plasma membrane was frequently observed. Attachment of mastigonemes to the long flagellum could not be observed, although we were able to clarify the development of the basal swelling part of the short flagellum at where was composed of different materials.
