    Adults of tortoise beetles (Coleoptera: Chrysomelidae: Cassidinae) possess peculiar morphologies, such as explanate margins surrounding their body, tarsal pad for adhesion to the substrate, and long male genitalia that might be evolved via natural and sexual selection. Therefore, tortoise beetles are ideal materials for investigating the developmental mechanisms and evolution of many adaptive traits. This study attempts to evaluate the effectiveness of the RNA interference (RNAi) method for analyzing functions of genes related to the formation of adult morphologies using tortoise beetle species Thlaspida biramosa (Boheman) (Coleoptera: Chrysomelidae: Cassidinae). We inject dsRNA of the well-known appendage patterning gene, dachshund ( dac ), into the last-instar larvae of T. biramosa . dac RNAi adults exhibited a reduction in number of subsegments of antennae and legs as well as the formation of abnormal grooves on legs. Reduction of appendages has also been known in the previous studies using Drosophila and Tribolium when dac expression was suppressed, suggesting that the RNAi-based dac knockdown was likely to work successfully in T. biramosa . Additionally, this study reveals that the degree of abnormal phenotypes is the same between treatments with 0.1 and 1 µg of dac dsRNA, suggesting high RNAi potential in T. biramosa . Furthermore, there appears to be a specific dsRNA injection timing that maximized RNAi efficiency. Therefore, this study provides the first evidence of the potential for the RNAi-based gene silencing in tortoise beetles. Future studies will be able to analyze genes that cause the unique morphology of tortoise beetles by applying our larval RNAi method.

  • Soma–germ contact across the basement membrane in the ovary

    Scientific journal, Epithelial cells interact with other cells and environments at their apical side, while the basement membrane typically impedes such interaction at the basal surface. One notable instance is communication between soma and germ cells within the ovary across numerous bilaterian taxa. This contact underlies proper oogenesis and subsequent embryogenesis. Throughout the history of morphology and cell biology, there has been an emphasis on this heterocellular interaction primarily occurring at the apical side of epithelial cells. Contrary to this long-standing understanding, we uncover that ovarian follicle cells in two myriapod species belonging to phylogenetically basal myriapod clades extend their cytoplasmic processes, penetrating the basement membrane to establish direct contact with oocytes. These discoveries demonstrate that the ovarian soma–germ interaction transverses the basement membrane, suggesting that the basal matrix is not always a physical barrier to soma–germ communication. Furthermore, we find that the ovarian somatic cells in a myriapod directly connect with the oogonia or young oocyte before forming their basement membrane. These results encourage reconsidering the conventional view of soma–germ interaction and suggest an overlooked construction manner of heterocellular communication in epithelial cells.

    Alternative splicing underpins functional diversity in proteins and the complexity and diversity of eukaryotes. An example is the doublesex gene, the key transcriptional factor in arthropod sexual differentiation. doublesex is controlled by sex-specific splicing and promotes both male and female differentiation in holometabolan insects, whereas, in hemimetabolan species, doublesex has sex-specific isoforms but is not required for female differentiation. How doublesex evolved to be essential for female development remains largely unknown. Here, we investigate ancestral states of doublesex using Thermobia domestica belonging to Zygentoma, the sister group of Pterygota, i.e., winged insects. We find that, in T. domestica, the doublesex expresses sex-specific isoforms but is only necessary for male differentiation of sexual morphology. This result supports the hypothesis that doublesex initially promoted male differentiation during insect evolution. However, T. domestica doublesex has a short female-specific region and upregulates the expression of vitellogenin homologs in females, suggesting that doublesex may already play some role in female morphogenesis of the common ancestor of Pterygota. Reconstruction of the ancestral sequence and prediction of protein structures show that the female-specific isoform of doublesex has an extended C-terminal disordered region in holometabolan insects but not in non-holometabolan species. We propose that doublesex acquired its function in female morphogenesis through a change in the protein motif structure rather than the emergence of the female-specific exon.
  • Efficient production of long double-stranded RNAs applicable to agricultural pest control by Corynebacterium glutamicum equipped with coliphage T7-expression system

    English, Scientific journal, RNA-based pesticides exert their function by suppressing the expression of an essential gene in the target pest through RNA interference caused by double-stranded RNA (dsRNA). Here, we selected target genes for growth suppression of the solanaceous crop pests ladybird beetle (Henosepilachna vigintioctopunctata) and Colorado potato beetle (Leptinotarsa decemlineata)-the death-associated inhibitor of apoptosis protein 1 gene (diap1), and an orthologous gene of the COPI coatomer protein complex (copI), respectively. We constructed a cost-competitive overproduction system for dsRNA using Corynebacterium glutamicum as a host bacterium. The dsRNA expression unit was equipped with two sets of promoters and terminators derived from coliphage T7, and the convergent expression system was designed to be selectively transcribed by T7 RNA polymerase. This expression system efficiently overproduced both target dsRNAs. On culture in a jar fermentor, the yield of diap1-targeting dsRNA (approximately 360 bp) was > 1 g per liter of culture. Long-chain diap1-targeting dsRNAs (up to around 1 kbp) could be produced without a substantial loss of efficiency. dsRNA accumulated in C. glutamicum significantly suppressed larval growth of H. vigintioctopunctata. The dsRNA expression technology developed here is expected to substantially reduce dsRNA production costs. Our method can be applied for a wide range of industrial uses, including agricultural pest control. KEY POINTS: • Overexpression of dsRNA was achieved in C. glutamicum using a coliphage T7 system. • The best strain produced > 1 g/L of the target dsRNA species, for use as an insecticide. • The developed system efficiently produced long dsRNA species, up to ~ 1 kbp.

    English, Scientific journal, Apicomplexa mainly comprises parasitic species and some of them, which infect and cause severe diseases to humans and livestock, have been extensively studied due to the clinical and industrial importance. Besides, apicomplexans are a popular subject of the studies focusing on the evolution initiated by a secondary loss of photosynthesis. By interpreting the position in the tree of eukaryotes and lifestyles of the phylogenetic relatives parsimoniously, the extant apicomplexans are predicted to be the descendants of a parasite bearing a non-photosynthetic (cryptic) plastid. The plastid-bearing characteristic for the ancestral apicomplexan is further strengthened by non-photosynthetic plastids found in the extant apicomplexans. The research on apicomplexan members infecting invertebrates is much less advanced than that on the pathogens to humans and livestock. Gregarines are apicomplexans that infect diverse invertebrates and recent studies based on transcriptome data revealed the presence of cryptic plastids in a subset of the species investigated. In this study, we isolated gregarine-like organisms (GLOs) from three arthropod species and conducted transcriptome analyses on the isolated cells. A transcriptome-based, multi-gene phylogenetic analysis clearly indicated that all of the three GLOs are eugregarines. Significantly, the transcriptome data from the GLO in a centipede appeared to contain the transcripts encoding enzymes involved in the non-mevalonate pathway for isopentenyl diphosphate biosynthesis and C5 pathway for heme biosynthesis. The enzymes involved in the two plastid-localized metabolic pathways circumstantially but strongly suggest that the particular GLO possesses a cryptic plastid. The evolution of cryptic plastids in eugregarines is revised by incorporating the new data obtained from the three GLOs in this study.
