English, Scientific journal, INTRODUCTION: Inducible bronchus-associated lymphoid tissue (iBALT) develops with different morphologies and functions depending on the type of antigen, in which various cytokines, such as interleukin (IL)-1 and IL-17, and the cells producing them, such as T helper 17 (Th17) and T follicular helper (Tfh) cells, play an important role. We recently observed that numerous inflammatory cells, mainly B cell like-cells forming peribronchial clusters, accumulate in the lungs of mice exposed to Asian sand dust (ASD), suggesting that ASD induced iBALT development. However, whether ASD induced iBALT formation, much less the mechanism by which ASD promotes iBALT formation, remains unknown. METHODS: B cell clusters were analyzed using the next generation serial section-three-dimensional (nSS3D) imaging method, in which we attempted to introduce batch image acquisition using a high-resolution slide scanner and AI-based image registration and target extraction. Furthermore, the mechanism underlying ASD-induced B-cell cluster formation was examined using CD4-Cre Bcl6f/f mice lacking Tfh cells. RESULTS: ASD induced B-cell cluster formation in mouse lung tissue, which was enhanced by allergen (Ovalbumin: OVA) exposure. Furthermore, the nSS3D images revealed that a part of the B cell clusters induced by OVA+ASD but not others exhibited common histological features of previously reported iBALTs. Moreover, OVA+ASD exposure failed to induce all of the B cell cluster formation including iBALTs in CD4-Cre Bcl6f/f mice. CONCLUSION: B cell clusters including iBALTs are induced by ASD; this process is enhanced by OVA, in which Tfh cells were suggested to play important roles. Characterization of the OVA+ASD-induced B cell clusters proved that the SS3D technique is useful for the analysis of mouse disease models. English, Scientific journal, The epithelial basal lamina of the small intestine has numerous fenestrations for intraepithelial migration of leukocytes. We have reported dynamic changes of fenestrations in dietary conditions. To investigate this phenomenon, we performed statistical analyses using scanning electron microscopy images of the epithelial basal lamina of rat intestinal villi after removal of the villous epithelium by osmium maceration. We examined structural changes in the number and size of fenestrations in the rat jejunum and ileum under fasted and fed states for 24 h. Our findings revealed that, in the jejunum, the number of free cells migrating into the epithelium through fenestrations increased from 2 h after feeding, resulting in an increase in the fenestration size of intestinal villi; the number of free cells then tended to decrease at 6 h after feeding, and the fenestration size also gradually decreased. By contrast, the increase in the fenestration size by feeding was not statistically significant in the ileum. These findings indicate that the number of migrating cells increases in the upper part of the small intestine under dietary conditions, which may influence the absorption efficiency of nutrients including lipids, as well as the induction of nutrient-induced inflammation. - Scanning ion-conductance microscopy with a double-barreled nanopipette for topographic imaging of charged chromosomes.

English, Scientific journal, Scanning ion conductance microscopy (SICM) is useful for imaging soft and fragile biological samples in liquids because it probes the samples' surface topography by detecting ion currents under non-contact and force-free conditions. SICM acquires the surface topographical height by detecting the ion current reduction that occurs when an electrolyte-filled glass nanopipette approaches the sample surface. However, most biological materials have electrically charged surfaces in liquid environments, which sometimes affect the behavior of the ion currents detected by SICM and, especially, make topography measurements difficult. For measuring such charged samples, we propose a novel imaging method that uses a double-barrel nanopipette as an SICM probe. The ion current between the two apertures of the nanopipette desensitizes the surface charge effect on imaging. In this study, metaphase chromosomes of Indian muntjac were imaged by this technique because, owing to their strongly negatively charged surfaces in phosphate-buffered saline, it is difficult to obtain the topography of the chromosomes by the conventional SICM with a single-aperture nanopipette. Using the proposed method with a double-barrel nanopipette, the surfaces of the chromosomes were successfully measured, without any surface charge confounder. Since the detailed imaging of sample topography can be performed in physiological liquid conditions regardless of the sample charge, it is expected to be used for analyzing the high-order structure of chromosomes in relation to their dynamic changes in the cell division. - Scanning ion conductance microscopy of isolated metaphase chromosomes in a liquid environment

Recent progress in atomic force microscopy (AFM) combined with cell microarray substrates revealed that the number distribution of the complex shear modulus of cells exhibited a log-normal and became narrower with frequency, according to the power-law rheology model. Japanese, The mechanical properties of live cells are associated with various cell functions, and thus it is crucial to determine the cell elastic modulus, which is one of the fundamental quantities of matter and changes with time and frequency. Atomic force microscopy (AFM) allows us to directly measure the relaxation modulus of single cells in stress and creep relaxation measurements as well as the cell complex modulus in the force modulation. Recent progress in AFM combined with microfabricated substrates revealed the cell-cell variability of cell mechanics, in which the cell modulus exhibits a log-normal distribution depending on the frequency, according to a power-law rheology model. The AFM with micropost substrates as a force sensor demonstrated the direct measurement of intracellular force propagation through cytoskeletal networks between focal adhesions. - Universal Rheological Properties of Cells in Intermediate States Observed in Stress Relaxation of Cell Sheets

English - 1031 Effect of Sympathetic Nervous Activity and Microtubule Polymerization on Contractility in Myocardial Cells

Japanese, Cytoskeleton plays important roles in the arrangement of intracellular organelles, the development of tissue remodeling, and the determination of mechanical properties of the cell. However, it is not understood how the cytoskeleton influence the contraction of myocardial cells. In the present study, the role of microtubule in the contraction of isolated myocardial cells was studied by hyperpolymerizing microtubule with treating paclitaxel and depoymerizing it with colchicine. Stimulation frequency, concentration of isoproterenol,, and concentration of extracellular calcium were altered to simulate the changes in sympathetic nervous activity. Then, the amount of cell shortening, velocity of shortening, and velocity of 50% relengthening were measured. As results, it was indicated that the microtubule acts as a dumper element (viscous component) for cell contraction. In addition to this, it was indicated that the microtubule acts as a resistant element (elastic component) when cell contraction was enhanced with increasing sympathetic nervous activity.

【目的】腸肝組織においてコレステロールからの胆汁酸合成を制御するFXR(farnesoid X receptor)が雌雄生殖器にも存在しており,マウス精巣においてFXRがテストステロン合成に関与していることが報告されている。しかしながら,その詳細なメカニズムは不明な点が多く,生殖器におけるFXRの機能はほとんど解明されていない。そこで本研究は,これまでに本研究室で作出したFXRノックアウト(FXR-KO)雄マウスの生殖生理学的解析をおこない,精巣のFXRがテストステロン合成に寄与する機構を解明する。【方法】①FXR-KO雄マウスの妊孕能を調べるため,交配試験をおこなった。②野生型およびFXR-KOマウス精巣よりライディッヒ細胞を採取し,1%FBS,1%ITS添加DMEM培地で培養した。24時間培養後の培養上清中のテストステロン量,および③性ホルモン合成関連遺伝子の発現量を調べた。【結果】①交配試験の結果,平均産子数は野生型マウスで11.2匹,FXR-KOマウスで11.6匹と同程度で,FXR-KO雄マウスは正常な妊孕能を有していた。②ライディッヒ細胞培養上清中のテストステロン量を測定した結果,野生型マウスで15.9 pg/mL,FXR-KOマウスで46.6 pg/mLとFXR-KOマウスで有意に高かった。③ライディッヒ細胞の遺伝子発現解析の結果,FXR-KOマウスでテストステロン合成遺伝子であるStAR,CYP11a1,HSD3b,およびCYP17a1の発現が有意に上昇していた。以上の結果,精巣のFXRは性ホルモン合成関連遺伝子群の発現を抑制し,テストステロン合成を下方制御していることが示された。, The Society for Reproduction and Development, Japanese - Imaging with canceling of charging influence of sample surface using a scanning ion conductance microscopy with a theta nanopipette

A scanning ion conductance microscope (SICM) can image living samples in liquid by detecting ion current through a nanopipette. The nanopipette has an aperture of approximately 100 nm in diameter. In the SICM technique, it is difficult to obtain electrically charged samples because of the complicated ion current behaviors detected with the nanopipette in the vicinity of the surface. In this study, to observe charged samples, we used a theta type nanopipette as a probe of the SICM. Furthermore, we additionally developed a method for obtaining a charge map of the sample surface. Mechanical measurements of single cells are crucial for understanding various cell behaviors. The viscoelastic properties of a number of rat fibroblast cells in the process of transformations such as growth (Phase II), senescence (Phase III) and immortalization (IM) were investigated by atomic force microscopy (AFM) combining with a cell microarray technique. The complex shear modulus of cells as a function of frequency was fitted to a power-law model, consisting of a single power-law function in addition to the Newtonian viscosity. Both the mean value and standard deviation of the power-law exponent a strongly depended on the cell phases. As a result, the power-law behaviors of cells were changed in the process of transformation. The mechanical properties of live cells are associated with various cell functions, and thus it is crucial to determine the cell elastic modulus, which is one of the fundamental quantities of matter and changes with time and frequency. Atomic force microscopy (AFM) allows us to directly measure the relaxation modulus of single cells in stress and creep relaxation measurements as well as the cell complex modulus in the force modulation. Recent progress in AFM combined with microfabricated substrates revealed the cell-cell variability of cell mechanics, in which the cell modulus exhibits a log-normal distribution depending on the frequency, according to a power-law rheology model. Ventricular fibrillation (VF) and reentrant ventricular tachycardia (VT) are the major immediate causes of sudden cardiac death. Whereas VT is considered a rapid but well organized process, VF has been described as turbulent cardiac electrical activity, resulting from the random and aperiodic propagation of multiple independent wavelets throughout the cardiac muscle. Clinical studies have shown that VF is almost always preceded by VT of variable duration, from a few to many beats. However, the manner in which VT converts to VF, or vice versa is not yet fully understood despite more than a century of intensive study. We have previously demonstrated that the perfusion of isolated hearts with ruthenium red (RR), a blocker of the mitochondrial Ca^<2+> uniporter, converts pacing-induced sustained VF to VT. In addition, we have also demonstrated that the perfusion of isolated rat hearts with RR during sustained VF results in the close balance between myocardial energy supply and demand, suggesting that the balance was lost during VF, leading to the significant decrease of cellular ATP in the myocardium during VF. In this study, we investigated whether high-frequency electrical stimulation affected the intracellular concentration of Ca^<2+> and Mg^<2+> dynamics in isolated adult rat cardiomyocytes. The present study revealed that the persistent high-frequency stimulation resulted in the collapse in an ionic homeostasis in cardiomyocytes. In our group, the tiny robot with the pico-litter dispensing resolution which can be applied in the field of μTAS is developed. This dispensing mechanism is composed with a thin glass capillary and the single thin needle which driven through it. Then the liquid in the tube can be pushed out and the small amount of the liquid can be squeezed on the surface. This simple manner can allow dispensing with the resolution of a pico litter oder possible. This report describes the micro emulsion generation mechanism made by applying pico-litter dispenser. And the control of the diameter of the emulsion was experimentally examined by using this mechanism. 本研究ではμTAS分野において活用可能な微少量の液体をハンドリングするといった分注機構を搭載させた超小型ロボットの開発を目的としている。
そこで今回は、超小型ロボットに搭載可能な小型単純構造であり、様々な粘度の試料液体を吐出することが可能、吐出量を調整することが可能、さらに[pl]オーダーの吐出量を目標とした極微量液体吐出機構の試作・開発を行なった。

Ventricular fibrillation (VF) and reentrant ventricular tachycardia (VT) are the major immediate causes of sudden cardiac death. Whereas VT is considered a rapid but well organized process, VF has been described as turbulent cardiac electrical activity, resulting from the random and aperiodic propagation of multiple independent wavelets throughout the cardiac muscle. Clinical studies have shown that VF is almost always preceded by VT of variable duration, from a few to many beats. However, the manner in which VT converts to VF, or vice versa is not yet fully understood despite more than a century of intensive study. We have previously demonstrated that the perfusion of isolated hearts with ruthenium red (RR), a blocker of the mitochondrial Ca^<2+> uniporter, converts pacing-induced sustained VF to VT. In addition, we have also demonstrated that the perfusion of isolated rat hearts with RR during sustained VF results in the close balance between myocardial energy supply and demand, suggesting that the balance was lost during VF, leading to the significant decrease of cellular ATP in the myocardium during VF. In this study, we investigated whether high-frequency electrical stimulation affected the intracellular concentration of Ca^<2+> and Mg^<2+> dynamics in isolated adult rat cardiomyocytes. The present study revealed that the persistent high-frequency stimulation resulted in the collapse in an ionic homeostasis in cardiomyocytes. In addition, the intracellular concentration of Mg^<2+> increased significantly in association with the high-frequency stimulation-induced hypercontraction, suggesting that the myocardial ATP content was markedly reduced during the stimulation. - Changes in the concentrations of Ca2+ and Mg2+ induced by high-frequency electrical stimulation in adult rat cardiomyocytes.

In our group, the tiny robot with the pico-litter dispensing resolution which can be applied in the field of μTAS is developed. This dispensing mechanism is composed with a thin glass capillary and the single thin needle which driven through it. Then the liquid in the tube can be pushed out and the small amount of the liquid can be squeezed on the surface. This simple manner can allow dispensing with the resolution of a pico litter oder possible. This report describes the micro emulsion generation mechanism made by applying pico-litter dispenser. And the control of the diameter of the emulsion was experimentally examined by using this mechanism. 本研究は,走査型イオンコンダクタンス顕微鏡(SICM)を用いて取得できる生きた細胞の頂上膜面の細胞質突起の画像からダイナミクスを解析し,そのダイナミクスと細胞機能の関係性を解明することを目的としている。そのダイナミクスの解析には,本研究を通して構築する機械学習を用いたコンピュータ支援構造判別評価システムを使用し,困難さを多く含む画像判別を自動化することでより効率的におこなう事を並行して実施するものである。
取り組んでいる課題を以下に示した。
1. 画像データセットの収集と機械学習用データを用いたシステムの構築: 申請者は現在までに各種実験を通して撮像したSICM像を所有し,継続的に画像データセットの収集および本研究に学習データとして用いるデータセットを選別しタグ付け作業をおこなった。また,システムの構築は,フリーウエアで構築を進め,テスト画像データ等を処理させるなど適したものの選定をおこない,機械学習の作業環境整備を進め,識別アルゴリズムの作成に着手した。
2. 各細胞の細胞質突起の構造観察: SICMを用いた画像取得は,探針であるガラスピペットの形状や細胞の状態に依って適した画像取得に関するパラメータは異なる。本研究では,細胞質突起の観察における各種条件を精査することで,現状において適した画像取得パラメータを見出した。
3. Tet受容体を恒常的に発現する細胞の樹立: 遺伝子導入により細胞質突起構造の遷移状態をコントロール可能なCa9-22細胞の作製を試みた。当初はTetの発現を確認したが,発現していても数日で発現が消失することが明らかとなった。そのため,非メチル化プロモーターに組み換えた発現ベクターを作製し,細胞にトランスフェクションおよびセレクションを行うことでTet受容体恒常発現細胞(TetR-Ca9-22 cells) の樹立に成功した。

To elucidate the underlying mechanism of secretory leukocyte protease inhibitor (SLPI)-induced cell migration, we compared SLPI-deleted human gingival carcinoma Ca9-22 (ΔSLPI) cells and original (wild-type: wt) Ca9-22 cells using several microscopic imaging methods and gene expression analysis. Our results indicated reduced migration of ΔSLPI cells compared to wtCa9-22 cells. The lamellipodia/dorsal ruffles were smaller and moved slower in ΔSLPI cells compared to wtCa9-22 cells. Furthermore, well-developed intermediate filament bundles were observed at the desmosome junction of ΔSLPI cells. In addition, Galectin4 was strongly expressed in ΔSLPI cells, and its forced expression suppressed migration of wtCa9-22 cells. Taken together, SLPI facilitates cell migration by regulating lamellipodia/ruffles and desmosomes, in which Galectin4 plays an important role.

Scanning Ion Conductance Microscopy (SICM) is the family of scanning probe microscopy. This technique uses a glass nanopipette filled with an electrolyte as a probing tip and regulates the tip-surface distance by detecting the ion current passing through a small hole of the pipette. Thus, SICM can obtain images of three-dimensional structures in liquid conditions without mechanical contact. In this study, we succeeded in establishing a method for observing the three-dimensional dynamics of microvillous structures including dorsal ruffles on the cell membrane surface.
By using SICM and fluorescence microscopy, we observed the dynamics of the dorsal surface ruffles and the intracellular cytoskeleton related to cell motility. Furthermore, we observed the dorsal surface ruffles of cancer cells, and showed that the structure and dynamics of dorsal surface ruffles differ greatly depending on the grade of cancer cells.

Scanning ion conductance microscopy (SICM) uses a glass micropipette as a sensitive probe, which contains electrode in an electrolyte. This technique allows force-free topographic imaging of soft samples under liquid conditions. Thus, we have been interested in the applicability of SICM to imaging of different types of biological samples, such as collagen fibrils, chromosomes and cultivated cells. In this study, we succeeded in obtaining images of tissue blocks such as trachea, kidney and lymph node. The comparison of the SICM and scanning electron microscopy (SEM) images showed that SICM is useful for observing the three-dimensional structure of cells and tissues in liquid conditions with the quality comparable to the SEM image. We also succeeded in obtaining images of tissue sections for studying such cell organelles as mitochondria. Finally, we showed the possibility of measurement of charge distribution of sliced tissues by SICM

We investigated the possibility of observing live cells and tissues by atmospheric scanning electron microscopy (atmospheric SEM). Our study revealed that this technique is powerful for observing plant tissues including leaf surfaces, but has some difficulty for visualizing the surface of animal cells and tissues due to several reasons such as surface unevenness, low signal intensity and the presence of surface water . To improve the quality of cell observation by atmospheric SEM, preparation techniques including the ion liquid treatment were tested in this study.
