    Aberrant salience processing has been proposed as a pathophysiological mechanism underlying psychiatric symptoms in patients with schizophrenia. The gaze trajectories of individuals with schizophrenia have been reported to be abnormal when viewing an image, suggesting anomalous visual salience as one possible pathophysiological mechanism associated with psychiatric diseases. This study was designed to determine whether visual salience is affected in individuals with schizophrenia, and whether this abnormality is unique to patients with schizophrenia. We examined the gaze behaviors of 1012 participants recruited from seven institutes (550 healthy individuals and 238, 41, 50 and 133 individuals with schizophrenia, bipolar disorder, major depressive disorder and autism spectrum disorder, respectively) when they looked at stationary images as they liked, i.e., free-viewing condition. We used an established computational model of salience maps derived from low-level visual features to measure the degree to which the gaze trajectories of individuals were guided by visual salience. The analysis revealed that the saliency at the gaze of individuals with schizophrenia were higher than healthy individuals, suggesting that patients’ gazes were guided more by low-level image salience. Among the low-level image features, orientation salience was most affected. Furthermore, a general linear model analysis of the data for the four psychiatric disorders revealed a significant effect of disease. This abnormal salience processing depended on the disease and was strongest in patients with schizophrenia, followed by patients with bipolar disorder, major depressive disorder, and autism spectrum disorder, suggesting a link between abnormalities in salience processing and strength/frequency for psychosis of these disorders.

    English, Scientific journal, Abnormalities in visual exploration affect the daily lives of patients with schizophrenia. For example, scanpath length during free-viewing is shorter in schizophrenia. However, its origin and its relevance to symptoms are unknown. Here we investigate the possibility that abnormalities in eye movements result from abnormalities in visual or visuo-cognitive processing. More specifically, we examined whether such abnormalities reflect visual salience in schizophrenia. Eye movements of 82 patients and 252 healthy individuals viewing natural and/or complex images were examined using saliency maps for static images to determine the contributions of low-level visual features to salience-guided eye movements. The results showed that the mean value for orientation salience at the gazes of the participants with schizophrenia were higher than that of the healthy control subjects. Further analyses revealed that orientation salience defined by the L + M channel of the DKL color space is specifically affected in schizophrenia, suggesting abnormalities in the magnocellular visual pathway. By looking into the computational stages of the visual salience, we found that the difference between schizophrenia and healthy control emerges at the earlier stage, suggesting functional decline in early visual processing.     English, Scientific journal, Various eye movement abnormalities and impairments in visual information processing have been reported in patients with schizophrenia. Therefore, dysfunction of saccadic eye movements is a potential biological marker for schizophrenia. In the present study, we used a pharmacological model of schizophrenia symptoms in marmosets and compared the eye movement characteristics of marmosets during free-viewing, using an image set identical to those used for human studies. It contains natural and complex images that were randomly presented for 8 s. As a pharmacological model of schizophrenia symptoms, a subanesthetic dose of ketamine was injected intramuscularly for transient and reversible manipulation. Eye movements were recorded and compared under a ketamine condition and a saline condition as a control. The results showed that ketamine affected eye movement characteristics during free-viewing. Saccades amplitude and scanpath length were significantly reduced in the ketamine condition. In addition, the duration of saccades was longer under the ketamine condition than under the saline condition. A similar tendency was observed for the duration of fixations. The number of saccades and fixations tended to decrease in the ketamine condition. The peak saccades velocity also decreased after ketamine injection whereas there was no difference in the main sequence relationship between saccades amplitude and peak velocity. These results suggest that ketamine affected visual exploration but did not affect the oculomotor aspect of saccades in marmosets, consistent with studies in patients with schizophrenia. Therefore, we conclude that the subanesthetic dose of ketamine is a promising pharmacological model of schizophrenia symptoms in common marmosets and can be used in combination with free-viewing paradigms to establish “translatable markers” for schizophrenia in primates., 14752825;31462937

    English, Scientific journal, Visual selection in primates is intricately linked to eye movements, which are generated by a network of cortical and subcortical neural circuits. When visual selection is performed covertly, without foveating eye movements toward the selected targets, a class of fixational eye movements, called microsaccades, is still involved. Microsaccades are small saccades that occur when maintaining precise gaze fixation on a stationary point, and they exhibit robust modulations in peripheral cueing paradigms used to investigate covert visual selection mechanisms. These modulations consist of changes in both microsaccade directions and frequencies after cue onsets. Over the past two decades, the properties and functional implications of these modulations have been heavily studied, revealing a potentially important role for microsaccades in mediating covert visual selection effects. However, the neural mechanisms underlying cueing effects on microsaccades are only beginning to be investigated. Here we review the available causal manipulation evidence for these effects’ cortical and subcortical substrates. In the superior colliculus (SC), activity representing peripheral visual cues strongly influences microsaccade direction, but not frequency, modulations. In the cortical frontal eye fields (FEF), activity only compensates for early reflexive effects of cues on microsaccades. Using evidence from behavior, theoretical modeling, and preliminary lesion data from the primary visual cortex and microstimulation data from the lower brainstem, we argue that the early reflexive microsaccade effects arise subcortically, downstream of the SC. Overall, studying cueing effects on microsaccades in primates represents an important opportunity to link perception, cognition, and action through unaddressed cortical-subcortical neural interactions.   • The posterior parietal cortex contributes to visuomotor processing for saccades in blindsight macaques.

    English, Scientific journal, Patients with damage to the primary visual cortex (V1) lose visual awareness, yet retain the ability to perform visuomotor tasks, which is called "blindsight." To understand the neural mechanisms underlying this residual visuomotor function, we studied a non-human primate model of blindsight with a unilateral lesion of V1 using various oculomotor tasks. Functional brain imaging by positron emission tomography showed a significant change after V1 lesion in saccade-related visuomotor activity in the intraparietal sulcus area in the ipsi- and contralesional posterior parietal cortex. Single unit recordings in the lateral bank of the intraparietal sulcus (lbIPS) showed visual responses to targets in the contralateral visual field on both hemispheres. Injection of muscimol into the ipsi- or contralesional lbIPSs significantly impaired saccades to targets in the V1 lesion-affected visual field, differently from previous reports in intact animals. These results indicate that the bilateral lbIPSs contribute to visuomotor function in blindsight.

Eye movement abnormalities are commonly observed in patients with schizophrenia (SZs). Here we examined whether such abnormalities reflect an anomaly in inhibition of return (IOR), the mechanism that inhibits orienting to previously fixated or attended locations. We analyzed spatiotemporal patterns of eye movement during free-viewing of visual images including natural scenes, geometrical patterns, and pseudorandom noise in SZs and healthy control participants (HCs). SZs made saccades to previously fixated locations more frequently than HCs. The time lapse from the preceding saccade was longer for return saccades than for forward saccades in both SZs and HCs, but the difference was smaller in SZs. SZs explored a smaller area than HCs. Generalized linear mixed-effect model analysis indicated that the frequent return saccades served to confine SZs’ visual exploration to localized regions. The higher probability of return saccades in SZs was related to cognitive decline after disease onset but not to the dose of prescribed antipsychotics. We conclude that SZs exhibited attenuated IOR under free-viewing conditions, which led to restricted scene scanning. IOR attenuation will be a useful clue for detecting impairment in attention/orienting control and accompanying cognitive decline in schizophrenia., 14752825

We found that all species exhibit qualitatively similar saccadic kinematics and saliency-driven saccadic behavior albeit with different parameters. Our results suggest that the marmoset possesses similar neural mechanisms to macaque and human for saccadic control, and it is an appropriate model to study neural mechanisms for active vision and attention., 14752825

    Japanese, 【はじめに,目的】半側空間無視とは主に右大脳半球の損傷によって引き起こされ,損傷と反対側の空間の感覚刺激に対する反応が欠如・低下する現象の事を指す。半側空間無視は背側注意経路と腹側注意経路が関係しており(Corbetta,2002),近年の研究によると腹側注意経路の損傷により,半側空間無視が出現すると報告されている(Shulman GL 2011)。しかし,どのような神経機構によって無視が起こっているか,明確にわかっている事は少ない。また,半側空間無視の動物モデルも確立していない。しかし近年の解剖学的研究から,マカクザルにも注意の背側経路と腹側経路に相同な神経ネットワークがあることが示唆されている。マカクザルの半側空間無視モデルを作成する事により,神経メカニズムを明らかにし,さらに有効なリハビリテーションの手法の開発に繋げられると考える。そこで本研究では,マカクザル半側空間無視モデルを作成することを目的とする。【方法】本研究は2頭のマカクザルで脳損傷を作成した。2頭のサルは,側頭連合野と前頭連合野とを離断するために腹側注意経路の相同部位である右上側頭回を損傷した。サルの半側空間無視を評価する課題を確立するために,ヒト患者での評価法として用いられる線分抹消課題を参考にし,ケージ内およびモンキーチェア上で評価できる課題を作成した。ケージ内の評価として「食物選択課題」を2頭のサルに行わせた。この課題は左右に各3つの蓋つきの筒があり,左右の各1つに縞模様の筒がある。サルは縞模様の筒を選択すると報酬がもらえる。モンキーチェア上での評価として「刺激選択課題」を1頭のサルに行わせた。この課題はタッチパネルを使用し,10個の刺激に1つだけ色や形の異なるターゲットが存在する。サルは2秒以内にそのターゲットにタッチすると報酬がもらえる。また刺激のセットは4種類使用した。さらに2頭のサルで3Tの核磁気共鳴画像法装置を用いて安静時脳活動を記録した。【結果】ケージ内の評価では2頭とも術後1週間は損傷側の空間に比べ,非損傷側の空間を長時間無視する空間無視様の症状がみられた。さらに1頭では,術後1ヶ月後でも損傷側に比べ,非損傷側の空間に対する反応時間が長くなった。モンキーチェア上の評価では術後1週間は4種類すべてに損傷側に比べ,非損傷側で反応時間は長くなり,正答率も低下した。また2頭とも運動障害および一次的な知覚障害はみられなかった。安静時脳活動では上側頭回の損傷によって損傷後1週間の段階では頭頂連合野と前頭連合野の間の機能的結合が低下していた。【結論】本研究結果から,脳損傷による無視症状がマカクザルにおいても見られることが明らかになった。また安静時機能MRIによって無視症状に関連した脳活動を同定することができた。以上のことから,マカクザルが半側空間無視の動物モデル確立のために有望であると考える。

    我々の視知覚は眼球運動という行為によって構成される。この「アクティブビジョン」をシーン理解と眼球運動の制御の両面から明らかにしたい。自由エネルギー原理では、生体がシーン理解をするために情報獲得の期待値が高い部分に視線を向けるという考えから、従来の視覚サリエンスを拡張した「ベイジアン・サプライズ」という概念が提唱されている。そこで本研究は「ベイジアン・サプライズ」が脳内で処理されているか検証することを目的とした。
    そこで以下の2つの実験デザインを用いてマーモセットから脳活動を計測した。[視覚性ミスマッチ課題] マーモセットに視覚刺激を提示する。Oddball条件はstandard刺激とdeviant刺激から構成されている。Many standards条件は8種類の方位の刺激から構成されている。Oddball条件でのdeviant刺激とMany standards条件でのdeviant刺激の応答の差分から、予想外の刺激が出たことによる成分(サプライズ)を順応や方位選択性の成分から単離することが可能となる。また、視覚刺激にgrating刺激、random dot motion刺激、物体刺激を提示することで、系統的に視覚刺激への反応選択性を定量化する。[フリービューイング課題]ではマーモセットに自然画像を提示して視線を計測する。視覚刺激には公開されている自然画像データベースを用いることによって、新規な画像が提示されるようにする。
    [脳活動計測] これらの行動課題を用いて、マーモセット大脳表面をカバーする96ch ECoG電極を用いて局所電場電位を計測した。また別の個体では、内視鏡型Caイメージング装置を用いて、頭頂連合野から神経細胞活動を大量に同時記録を行った。

    A.時間論グループ(田口、西郷、谷、田中、富山、栁川、武内)は、前年度の研究を受け、現象学的な「時間」の捉え方について、さらに踏み込んだ考察を行った。現象学的時間論を専門とする栁川耕平氏を博士研究員として招聘し、月に1回ほどのペースで研究会を行った。現象学、圏論(数学)、ロボティクス、精神病理学等の知見を持ち寄り、離人症や統合失調症、自閉症などにおける時間意識の変容をできるだけ構造化して理解し、圏論を用いて定式化する方法を議論した。とりわけ現象学で言う把持(retention)と予持(protention)の相互媒介構造を圏論によって形式的に表現する可能性が開かれてきた。また、時間と身体性のつながりについても議論し、原初的な「感じ」の無数のうごめきから、「矢印」的な動きの形が生成してくるプロセスとして、時間の原初的な生成を描くという方向性での議論が進展した。
    B.計算論的意識論グループ(吉田、山下、西尾、宮園、協力者・鈴木)は、前年度の議論から浮上してきた新しいアイデア、すなわち「サリエンス」と「アフォーダンス」を結びつけて理解するというアイデアを追究し、同じく月に1回ほどのペースで研究会を行った。とりわけ統合失調症に関する「異常サリエンス仮説」と、精神疾患へのアフォーダンス概念の適用について詳しく検討し、両者を「行為」を媒介として結びつける可能性が示された。予測符号化理論および自由エネルギー原理との関連も多角的に議論し、それにもとづいて「知覚サリエンス」と「動機サリエンス」の本質的な近縁性を浮き彫りにすることができた。
    これらの研究の成果は、論文と学会発表により部分的に公表した。また、2021年12月に北海道大学人間知・脳・AI研究教育センター主催で行われた国際会議The free energy principle of the brainなどにおいてもその成果の一端を公表した。

    Habit forms the fundamental basis of cognition and action. However, current philosophy and sciences of the mind lack an appropriate theoretical framework to study its significance for human experience. This project explored the unique role of habits in various dimensions of experience based on the enactive approach, an approach to mind and cognition developing in the field of cognitive sciences, while incorporating insights from phenomenological, pragmatist, and Japanese traditions of philosophy. It was divided into three sub-topics: (1) Developing a novel conception of habits; (2) Developing a habit-centred model of the mind; (3) Exploring its implications for the sciences of the mind.

  • Marmoset model of unilateral spatial neglect

    本研究では、マーモセットを用いた半側空間無視モデルの確立と、大脳注意経路のネットワーク活動の計測、モデル化、介入を目指した研究を実施した。
    損傷標的部位および損傷作成法の確定: これまでの研究代表者によるマカクザルを対象とした実験では、ヒト腹側注意経路の相同部位の損傷を行い、半側空間無視と類似した症状を再現している。マーモセットにおける皮質注意経路との相同部位を見つけるため、96chECoG電極をマーモセットに埋設して、視覚性ミスマッチ課題および聴覚性ミスマッチ課題を用いて脳活動を計測した。その結果、側頭連合野のTPO野が視覚性および聴覚性のサプライズの両方に関連することを見出した。以上の結果から、本研究の損傷標的部位をrTPO野に決定した。
    行動の評価法の確立: これまでの研究代表者によるマカクザルを対象とした実験では、フリービューイング中の視線の計測を行い、損傷作成後に視線が損傷と同側(右側)に偏ることを明らかにしている。そこで研究代表者はフリービューイング中のマーモセットの視線計測を行った。視線の位置の視覚サリエンスを定量化することで、マーモセットの視線が視覚サリエンスの高いところに誘引されることを明らかにした。以上によって脳損傷作成後の行動評価法を確立した。
    脳活動計測法の確立: これまでの研究代表者によるマカクザルを対象とした実験では、安静時fMRIを用いて、背側注意経路DANの半球間での活動相関が損傷後に変動することを明らかにしている。マーモセットにおいては、両半球の頭頂連合野PPCにウイルスベクターを注入することで、両半球のPPCからのCaシグナルを同時計測した。半球内の活動で弱い正の相関が見られた。以上によって、損傷前後でのDANのネットワークの変動を計測する方法を確立した。
    なお、予算ではその他として、動物実験施設使用料7ヶ月分2,142,860円の支出を行った。

  • Visual attention and visual awareness in a monkey model of unilateral spatial neglect

    The aim of this study is to understand the mechanisms of the symptoms and recovery from unilateral spatial neglect. For this purpose, we examined whether the damage in the brain replicate the symptoms of unilateral spatial neglect in macaque and marmoset monkeys and investigated the mechanism of the symptoms. We developed behavioral methods for testing visual attention and visual awareness. We also identified attention networks using resting-state functional magnetic resonance imaging in macaque monkeys. We also developed behavioral methods for testing visual attention with free-viewing and identified attention networks using electrocorticograms in marmoset monkeys.

  • Dynamic circuit shift during functional recovery from brain and/or spinal cord injury

    To clarify the circuit shift mechanism during the recovery from brain and spinal cord injury, we investigated the spinal cord injury and primary visual cortex injury models in macaque monkeys. In the former case, dexterous hand movements recover after the corticospinal tract lesion at C4/C5 segment, and we have demonstrated by using pathway-selective blocking with double viral vector infection technique that the propriospinal neurons in the mid-cervical segments are critically involved in the recovery during the early recovery phase, however during the late recovery phase, other descending pathways also participate in the recovery process and the contribution of propriospinal neurons becomes partial. In the latter case, we have demonstrated by using the same double viral vector infection technique that after the primary visual cortex lesion, the superior colliculus to pulvinar pathway is involved in the recovery of visually guided saccadic eye movements.

    When humans normally move their bodies, they get sense of agency (the subjective awareness that one is excusing and controlling one’s own actions) and sense of body ownership (the awareness that one is the owner of an action or movement). Senses of agency and body ownership constitute bodily self-awareness. Our research project found that neural substrates for bodily self-awareness mainly exist in neural networks connecting the inferior parietal lobe and the inferior frontal gyrus in the right cerebral hemisphere. This finding was obtained by our behavioral experiments, measurements of brain activity, and non-invasive brain stimulation in normal human subjects as well as our analysis of neural networks in schizophrenic patients with altered bodily self-awareness. We also revealed neural mechanisms of sensory suppression, which is the basis of bodily self-awareness, by recoding activity of individual neurons in monkeys.

    In this study, we used the macaque monkeys with unilateral V1 lesion as an animal model of blindsight, and clarified (1) that visual awareness was actually impaired by showing the reduction in sensitivity of object detection, (2) that the “retina - superior colliculus - pulvinar and lateral geniculate nucleus - extrastriate visual cortices - parietal cortex” pathway underlies blindsight, and (3) that complex cognitive functions are maintained, and (4) neural correlates of the “partial awareness” could be detected in the frontal association cortex.

    半側空間無視とは、主に右大脳半球の損傷によって引き起こされる、損傷対側の空間の感覚刺激に対する反応が低下する現象のことを指す。半側空間無視の動物モデルはまだ確立していない。近年の解剖学や機能イメージングの研究からは、マカクザルにも注意の背側経路と腹側経路に相同な神経経路があることが示唆されている。そこで本研究は以下を目的とした:A) マカクザルでの腹側注意経路の相同部位を損傷させることによって半側空間無視の動物モデルを確立する。B) 視線と頭部のトラッキングを用いて、動物モデルで網膜中心座標、頭部中心座標での処理がどのように影響を受けているかを解明する。C) そして機能的MRイメージング法を用いて、無視症状の発現と回復過程が注意経路の機能障害と関連しているか検証する。(A) 注意の腹側注意経路の一部である右上側頭回(STG)へ損傷を加えて、注意機能への影響を定量化した。昨年の2頭のニホンザルに加えて本年度はさらに2頭のサルで損傷実験を行った。Target-choice taskにおいて損傷対側の標的刺激に対する応答潜時は右STG損傷後3ヶ月にわたって増加していることが明らかになった。(B) 同じ動物において視線と頭部のトラッキングを行った。3頭のサルで損傷前後に静止画像を見ている間の計測を行ったところ、損傷後3ヶ月にわたって視線位置が損傷側と同側に偏位することを見出した。(A)および(B)の結果から、本研究は右STG損傷によってサル半側空間無視モデルの確立に成功したと結論づける。(C) 機能的MRイメージング法を用いて半側空間無視モデル動物での安静時脳活動を計測した。その結果、損傷後には損傷同側内の背側注意経路の機能的結合の低下と損傷対側内の背側注意経路の機能的結合の上昇という不均衡が起きたことを示唆するデータを得た。

  • Awareness and attention in animal models of blindsight and spatial neglect

    The aim of this research is to establish an animal model of spatial neglect. For this purpose, we made a lesion in the right superior temporal gyrus and evaluated their behavior with a target-selection task and eye-tracking. We found that neglect-like behavior was sustained for more than 1 month after the lesion. Thus we conclude that we established an animal model of spatial neglect. We also measured BOLD activity of the same monkeys before and after the lesion and found that the functional connectivity within the attention network is reduced and recovered, which was correlated with behavioral markers of spatial neglect after the lesion.

    It is known that damage to the primary visual cortex (V1) impairs visual awareness in the contralesional visual field, but some patients show residual vision which guides eye or hand movements to the objects presented in the impaired visual field. This ability is called “blindsight”. In this study, we combined studies on monkeys with unilateral V1 lesion, a human blindsight subject, in-vitro or in-vivo recording in the mouse superior colliculus and large-scaled network simulations, we investigated the neuronal mechanism of saliency detection by the blindsight. The results showed that the neural pathways from the retina to the superior colliculus, pulvinar, to the fronto-parietal cortical network are involved in the blindsight, and among them, the typical Mexican-hat like center excitation-surround inhibition local circuit structure extending in the spatial map of the midbrain superior colliculus, the input structure of the network, plays a key role in the saliency detection.

  • マカクザル半側空間無視モデルを用いた空間および身体の認知機構の解明

    半側空間無視とは脳損傷と反対側の空間の感覚刺激(視覚、聴覚、体性感覚)に対する反応が欠如・低下する現象のことを指す。これは感覚障害(同名半盲など)や運動障害(片麻痺など)によっては説明できない認知的障害である。本研究は半側空間無視の動物モデルを確立することを最大の目標とする。マカクザルの頭頂連合野と前頭連合野とを繋ぐ神経線維である上縦束(SLF)を切断することで無視症状を引き起こすことを目指した。この術式の確立のために上縦束を切断の手術を行った。手術終了後に行動観察を行った。動物モデルは術後1日後には回復した。顕著な運動麻痺などは起こっていないことから、手術は運動機能そのものへの直接的な影響はないことが推測される。また、刺激が一つである場合にはそれを目で追い、顔を向けることができることを確認した。このことは視野欠損及び眼球運動系の障害がないことを示している。ところが刺激を左右同時に提示したときには損傷と同側の刺激を常に選ぶ。つまり、無視症状に伴う「消去」の現象が見られることを確認した。また、目立たないように損傷側に刺激を提示した場合にはそれを無視した。つまり、空間無視の症状が見られた。また、損傷対側の身体に触れられても反応しない、といった身体無視の症状も見られた。以上のことから、半側空間無視の動物モデルを確立することに成功したと考えられる。また、行動評価法のひとつとして、ビデオクリップを観察中の実験動物の眼球運動を計測して、視覚的顕著性(saliency)の計算論的モデルを応用することによって、無視症状の行動評価を行うための実験系を確立した。これまでに第一次視覚野を損傷させたマカクザル盲視モデルでこの方法を応用して有用性を確認した。

    The aim of this study is to elucidate the neural mechanism of visual awareness using animal model of blindsight. We recorded the neural activities of the superior colliculus of the monkeys and found that their visual response is stronger when the monkeys were able to detect visual stimuli that when they failed to it. We found that such activity was developed during the transmission of visual information through the superficial layer of the superior colliculus to the intermediate later of the superior colliculus. These findings suggest that neural correlate of visual awareness develops within the local circuit of the superior colliculus.

    It has been reported that macaque monkeys with unilateral lesions in V1 exhibited analogous behavior to the human blindsight patients (Cowey and Stoerig, 1995). Here we examined behavioral report of visual awareness in monkeys with unilateral V1 lesions using saccade tasks and sought neural activity specific to blindsight. First, we examined whether the monkeys are able to maintain short-term memory of the stimuli presented in their contralateral (affected) hemifield. The monkeys were tested with a memory-guided saccade task with a 2 sec-delay. The success ratio was over 80%, significantly higher than chance. Then we recorded the neuronal activity from the superior colliculus (SC) during the memory-guided saccade task We found that a majority of the neurons recorded from the ipsilesional SC showed a delay activity selective for spatial locations of targets. On the other hand, the ratio of neurons with a delay activity was lower in the contralesional SC. These results suggest that the monkeys with V1 lesion retain a certain level of visual awareness and that it was represented in the SC. Then we examined their visual awareness more directly using comparison between the performance of a forced-choke (FC) task., in which localization of target positions was required, and that of a yes-no (YN) task, in which detection of the targets was required. The performance of the FC task was better than that of the YN task when stimuli were presented in the affected hemifield. Such a dissociation &performance was not observed when stimuli with low luminance contrasts were presented in the ipsilateral ('normal') hemifield, indicating that the dissociation is specific to the V1 lesion, not a general phenomena occurring in near threshold vision. Then we examined whether the neural activity of SC during the YN task is dependent on the monkeys' report on visual awareness. We found that, in the ipsilesional SC, the neural response to the visual stimuli in the affected hemifield was larger when the monkeys successfully detect the targets than when the monkeys missed them. Such modulation was not found in the neural response to the near threshold stimuli in the normal hemifield when we recorded from the contralesional SC. These results suggest that the modulation of neural activity found in the ipsilesional SC is a neural correlate of reduced visual awareness that specifically induced by V1 lesion.

  • ニホンザル盲視モデルにおける視覚的意識に関する認知神経科学的研究

    マカクザルの第一次視覚野を除去して作成した盲視動物モデルを用いて視覚的気づきの神経機構を明らかにすることを目的とした。視覚的気づきはサッケードを用いた検出報告課題を用いて評価した。この課題では損傷視野に標的刺激が現れる条件では標的にサッケードすれば報酬が与えられ、標的刺激が現れない条件(ST-)では注視点が消えてからも注視を維持することによって報酬が与えられる。損傷側の半球の上丘中間層より課題遂行中のニューロン活動を記録した。
    上丘ニューロン活動は、標的刺激が提示される条件(ST+)のうち、正しく標的の提示を報告できた試行(Hit)と報告できなかった試行(Miss)とでは、標的提示時の視覚応答がHit試行でより大きくなっていた。この活動はサッケード準備活動などでは説明できない。また、このような活動は健常側の上丘では見られなかった。よって、このような神経活動の修飾はV1切除によって特異的に引き起にされた視覚的気づきに対応した神経活動であると結論づけることができる。
    サッケードの応答潜時の分布をdiffusion modelでモデル化することによって意思決定の過程を推測することが可能である。盲視モデルサルの強制選択課題(視覚標的は損傷視野のどこかに必ず現れる)での応答潜時の分布をモデル化し、意思決定における閾値(Decision threshold)が損傷測の視野で低下していることを見出した。ひきつづき、同様なモデル化を検出報告課題でも行うことによって盲視モデルサルにおける視覚的気づきと意思決定との関係を明らかにする予定である。

  • ニホンザル盲視モデルによる視覚的意識と注意との関係に関する認知神経科学的研究

    二匹のニホンザルにおいて各種の眼球運動課題の習得を済ませた後、片側の第一次視覚野の除去手術を行った。手術後の再トレーニングによって以下の知見を得た。
    (1)術後の欠損半視野において、強制選択型の視覚誘導性眼球運動課題を遂行できることを確認した。欠損半視野で弁別できる標的刺激の閾値は手術前と比べて上昇していた。このことは、第一次視覚野を介さない経路によって残存視力による視覚運動変換が行われていることを示している。
    (2)強制選択型の視覚誘導性眼球運動課題において、眼球運動の最高速度が欠損半視野へ急速眼球運動するときには上昇していることを見出した。また、眼球運動の軌跡は正常半視野においては標的へと正確に到達するように屈曲していたのに対して、欠損半視野ではこのような屈曲の程度は少なかった。これらのことは、第一次視覚野を介さない経路による視覚運動変換での眼球運動の制御が不充分になっていることを示している。
    (3)強制選択型の視覚誘導性眼球運動課題において、注視点消去から視覚標的提示までに100msのgapを導入することによって、急速眼球運動の応答潜時が低下することを見出した(gap効果)。この応答潜時(120ms程度)は"express saccade"として知られるものに近い。このことは、第一次視覚野を介さない経路による視覚運動変換がgap効果によって修飾されることを示している。
    (4)欠損半視野に提示したprecue刺激が標的刺激を予告するとき、視覚誘導性眼球運動課題の弁別閾値および反応潜時の低下が起こることを見いだした。このことは、第一次視覚野を介さない経路が空間的注意を向けるメカニズムにも寄与していることを示している。
    (5)欠損半視野に提示した手がかり刺激を用いて、記憶誘導性眼球運動課題を遂行できることを見出した。このことは、第一次視覚野を介さない経路が手掛かり刺激の位置情報を遅延時間のあいだワーキングメモリーとして保持することおよび急速眼球運動の標的として合目的な行動に利用することに寄与していることを示している。

  • Probing the cognitive brain functions with the oculomotor system

    In this study, we studied the fundamental structure of the neural circuits controlling saccades and how the activity of the circuits can be modulated in a context dependent manner.
    (1)At the local circuits level, we clarified the neural mechanism of burst generation of the neurons in the deeper layer of the superior colliculus (SC). We found that the burst is generated via NMDA receptor-mediated transmission and local recurrent network. Release from GABAergic inhibition is essential for the circuit to exhibit burst.
    (2)We recorded the activities of neurons in the pedunculopontine tegmental nucleus (PPTN), which is the origin of cholinergic input to the SC, during visually guided saccade task. We have found that a group of PPTN neurons showed tonic discharge while monkeys are fixating the fixation point and make saccades. When a large reward volume was expected, the neuron showed higher activity, while when small reward was predicted, the activity was low. These results suggest that these PPTN neurons encode predicted reward. Considering that the PPTN neurons project to the dopamine neurons in the substantia nigra, which have been shown to encode reward prediction error, activity of the PPTN neurons were supposed to be used for calculation of reward prediction error at the dopamine neurons.
    (3)We investigated the saccades of monkeys with unilateral lesion of the primary visual cortex (V1) in monkeys. These monkeys with V1 lesion could perform visually guided saccades to the target presented in the blind field. These monkeys could perform memory guided saccades to the cue in the blind field and could exert top down attention to the cue in the blind field. These results suggested that the visual information through the pathway bypassing the V1 can trigger working memory and the visual processing in the pathway is under the influence of top down attention.
    (4)We have established the high speed video oculography to detect saccades in mice. Saccade-like rapid eye movements could be induced by SC stimulation in mice. Thus, we have established the experimental system to study the molecular basis of cognitive functions using mice model.
