- Application of viability PCR to investigate the capsid integrity of human enteric viruses in drinking water sources and the persistence of those viruses in surface waters.

English, Scientific journal, Here, we investigated the occurrence of indigenous human enteric viruses with intact capsids (i.e., those that are potentially infectious) in drinking water sources by applying viability PCR using improved propidium monoazide with an enhancer (PMAxx-Enhancer-PCR). A comparison of the results obtained with PCR alone and PMAxx-Enhancer-PCR suggested that adenovirus, norovirus GI, and rotavirus present in drinking water sources tended to have damaged capsids and be inactivated by damage to the capsid. In contrast, astrovirus, norovirus GII, and enteroviruses tended to have intact capsids and be potentially infectious. To elucidate the main cause of damage to the viral capsid, we next investigated the effects of environmental stresses (water temperature, particulate matter, enzymatic activity, and sunlight) on viral persistence in surface waters used as drinking water sources through virus-spiking incubation experiments by using infectivity assays, PCR alone, and PMAxx-Enhancer-PCR. The pseudo-first-order decay rate constants of the viruses obtained with PMAxx-Enhancer-PCR were comparable to those obtained with PCR alone when only water temperature, particulate matter, and enzymatic activity were stresses. In contrast, when sunlight was present in addition to the other three stresses, PMAxx-Enhancer-PCR showed larger decay rate constants than PCR alone. Thus, sunlight appears to be an environmental stress that determines the capsid integrity of indigenous human enteric viruses in drinking water sources. Indigenous pepper mild mottle virus (PMMoV) was present at >100 times higher concentrations in drinking water sources than human enteric viruses in both cases of using PCR alone and PMAxx-Enhancer-PCR, supporting the suitability of PMMoV as a surrogate for human enteric viruses for evaluating virus removal in actual drinking water treatment plants. The incubation experiments suggested that PMMoV is more resistant to environmental stresses than human enteric viruses, partly due to its more robust capsid structure. This may contribute to the high concentration of PMMoV in drinking water sources. - Methomyl, a carbamate insecticide, forms oxygenated transformation products that inhibit acetylcholinesterase upon chlorination.

English, Scientific journal, The fates of organophosphorus insecticides during chlorination, a widely used disinfection step in drinking water treatment, have been extensively investigated. These compounds are known to partly transform into their corresponding "oxons", which retain the same mode of action-the inhibition of acetylcholinesterase (AChE)-as their parent compounds, but with markedly greater potency. In contrast, although carbamate insecticides share the same mode of action, their toxicological fates during chlorination remain entirely unexplored. Here, we investigated changes in anti-AChE activity during chlorination of a solution containing methomyl, a carbamate insecticide. The activity was assessed in the presence (indirect toxicity) and absence (direct toxicity) of metabolic activation. Methomyl reacted with free chlorine and almost completely disappeared at a chlorine-to-compound molar ratio of ≥4. Even so, the chlorinated samples retained both direct and indirect toxicities. Indirect toxicity was attributable solely to residual methomyl. Direct toxicity originated from residual methomyl and the TPs methomyl-sulfoxide and a dioxygenated derivative of methomyl-sulfoxide. No other TPs contributed to it. Methomyl-sulfoxide was identified via MS/MS spectral matching, but the dioxygenated TP could not be structurally confirmed. From a toxicological perspective, drinking water treatment plants using free chlorine for disinfection should monitor at least methomyl-sulfoxide in addition to methomyl in finished water. - Decomposition in vacuum ultraviolet-based advanced oxidation/reduction processes and its relationship to physicochemical properties of target contaminants.

English, Scientific journal, Vacuum ultraviolet (VUV)-based treatment for water supplies can potentially be applied to a wide range of compounds because they enable enhancement of either oxidative or reductive decomposition through the addition of ozone (O3) or the reduction of dissolved oxygen (DO), respectively. In the present study, we applied three VUV-based treatments (VUV alone, VUV/O3, and VUV/low-DO) to five ozone-resistant compounds (1,4-dioxane, epichlorohydrin, acephate, N-nitrosodimethylamine [NDMA], and dichloroacetonitrile [DCAN]), and evaluated the extent of oxidative and reductive enhancements for each contaminant. Simple correlation analysis revealed that oxidative and reductive enhancements were more effective for compounds with a high highest occupied molecular orbital (HOMO) energy level and low lowest unoccupied molecular orbital (LUMO) energy level, respectively. These findings suggested that the effective VUV-based treatment for a contaminant might be identified by simply calculating its HOMO and LUMO energy levels. Further optimizations of O3 and DO concentrations reduced the electric energy per order values for epichlorohydrin and DCAN to approximately 4/5 and 1/3, respectively, of their values for VUV alone. - Chlorine-substituted transformation products generated during chlorination of the organophosphorus insecticide disulfoton induce anti-acetylcholine esterase activity

Japanese, Scientific journal, A novel aluminum-based coagulant for effective virus removal was developed in the present study based on the investigation of roles of the basicity, sulfate ion and aluminum speciation in coagulation of virus. The coagulation process with the novel aluminum-based coagulant effectively removed viruses compared with other aluminum-based coagulants, and achieved apporimately 6-log removals not only in the weakly acidic and neutral pH conditions but also weakly alkaline pH conditions. In addition, Al13 and Al30 polymers were detecteded by electrospray ionization mass spectrometry and 27Al-NMR spectrometry in the novel aluminum-based coagulant. Accordingly, Al13 and Al30 species are probably dominant species to control the virus removal performance, and that lead effective removals of viruses in the coagulation process with novel aluminum-based coagulant even in the weakly alkaline pH condition. - Difference in behaviors of F-specific DNA and RNA bacteriophages during coagulation-rapid sand filtration and coagulation-microfiltration processes

水系感染症を引き起こす病原性ウイルスの中でも,感染事例が世界中で増加しているヒトノロウイルスは,未だ効率的な培養法が確立されていないことから,添加実験による浄水処理性の評価を行うために必要なウイルス量を確保することが極めて困難であり,培養可能な病原性ウイルスと比べて研究が格段に遅れている.そこで,本研究では,ヒトノロウイルスの物理的な浄水処理性を,遺伝子組換えバキュロウイルスとカイコを用いたタンパク質発現法により発現させたヒトノロウイルス外套タンパク粒子(VLPs)を用いて評価することを目的とした.
本年度は,凝集沈澱-MF膜ろ過処理におけるVLPsの処理性を調べた.凝集沈澱-MF膜ろ過処理においては,凝集剤としてPACIを用いた場合,4log以上のVLPsの除去率が得られた.従って,凝集沈澱-MF膜ろ過処理を用いることにより,米国環境保護局の要求する4logの腸管系ウイルスの除去率を達成し得ることが示唆された.一方,凝集剤としてalumを用いた場合,VLPsの除去率は約3logとなり,PACIを用いた場合の除去率よりも低くなった.従って,凝集沈澱-MF膜ろ過処理においては,PACIを用いることにより,alumを用いる場合に比べて効果的にVLPsを除去できることが分かった.加えて,VLPsと大腸菌ファージQβおよびMS2の処理性を比較したところ,いずれの凝集剤を用いた場合でも,QβおよびMS2の除去率は,VLPsの除去率よりも低くなった.従って,凝集沈澱-MF膜ろ過処理においては,Qβ,MS2共にヒトノロウイルスの安全側の指標と成り得る可能性が示された.
