While animal and human decision strategies are typically explained by model-free and model-based reinforcement learning (RL), their choice sequences often follow simple procedures based on working memory (WM) of past actions and rewards. Here, we address how working memory-based choice strategies, such as win-stay-lose-switch (WSLS), are represented in the prefrontal and motor cortico-basal ganglia loops by simultaneous recording of neuronal activities in the dorsomedial striatum (DMS), the dorsolateral striatum (DLS), the medial prefrontal cortex (mPFC), and the primary motor cortex (M1). In order to compare neuronal representations when rats employ working memory-based strategies, we developed a new task paradigm, a continuous/intermittent choice task, consisting of choice and no-choice trials. While the continuous condition (CC) consisted of only choice trials, in the intermittent condition (IC), a no-choice trial was inserted after each choice trial to disrupt working memory of the previous choice and reward. Behaviors in CC showed high proportions of win-stay and lose-switch choices, which could be regarded as “a noisy WSLS strategy.” Poisson regression of neural spikes revealed encoding specifically in CC of the previous action and reward before action choice and prospective coding of WSLS action during action execution. Animals optimize their actions by predicting and verifying their outcomes (e.g., rewards). Reward prediction often requires working memory (WM)-based information. To elucidate the neural basis of WM-based reward prediction, we compared the activity of dopamine (DA) neurons in the ventral tegmental area (VTA) in an alternate reward condition (WM-based) with that in a random (WM-free) reward condition in rats and mice. Positron emission tomography revealed greater VTA activation in the WM-based than the WM-free condition. Lateral and medial VTA neurons displayed differential electrophysiological spike activities reflecting WM-based and WM-free reward prediction error, respectively. Furthermore, phasic DA release in the dorsal and ventral striatum changed as WM-based and WM-free classical conditioning progressed. Consistent with our WM-based model, reward acquisition caused a DA dip only in the dorsal striatum. 英語, 研究論文(学術雑誌), We investigated the effect of area postrema lesions and selective vagotomy of afferent fibers on emetine-induced nausea in rats. We evaluated the acquisition of the conditioned taste avoidance (CTA) to 0.1% saccharin solution after conditioning with emetine dihydrochloride (5.54 mg/kg, i.p., 1% BW). The CTA was measured in three groups of rats: a bilateral subdiaphragmatic afferent vagotomy group, an area postrema lesion group, and a sham lesion group. The bilateral vagotomy and sham groups of rats showed acquisition of CTA within 2 days of the test date. Taste avoidance was never conditioned in the area postrema lesion group. These results indicate that the area postrema plays a crucial role in the induction of emetine-induced nausea. - An engineered channelrhodopsin optimized for axon terminal activation and circuit mapping.

英語, 研究論文(学術雑誌), Optogenetic tools such as channelrhodopsin-2 (ChR2) enable the manipulation and mapping of neural circuits. However, ChR2 variants selectively transported down a neuron's long-range axonal projections for precise presynaptic activation remain lacking. As a result, ChR2 activation is often contaminated by the spurious activation of en passant fibers that compromise the accurate interpretation of functional effects. Here, we explored the engineering of a ChR2 variant specifically localized to presynaptic axon terminals. The metabotropic glutamate receptor 2 (mGluR2) C-terminal domain fused with a proteolytic motif and axon-targeting signal (mGluR2-PA tag) localized ChR2-YFP at axon terminals without disturbing normal transmission. mGluR2-PA-tagged ChR2 evoked transmitter release in distal projection areas enabling lower levels of photostimulation. Circuit connectivity mapping in vivo with the Spike Collision Test revealed that mGluR2-PA-tagged ChR2 is useful for identifying axonal projection with significant reduction in the polysynaptic excess noise. These results suggest that the mGluR2-PA tag helps actuate trafficking to the axon terminal, thereby providing abundant possibilities for optogenetic experiments. - Effects of methyl methacrylate on the excitability of the area postrema neurons in rats.

英語, 研究論文(学術雑誌), OBJECTIVES: The aim of the present study is to demonstrate the effects of inhaled methyl methacrylate (MMA) on the excitability of neurons in the area postrema (AP). We also investigated the relation between vagal afferent inputs and responding cells in the AP. METHODS: We set up two groups of experimental animals, such as rats inhaling MMA and rats inhaling room air. To visualize the changes of AP neuron excitability after inhalation of MMA for 90 min, c-Fos protein expression was identified and quantified by immunohistochemical analysis. Some rats receiving ventral gastric branch vagotomy were also subjected to the abovementioned experiment. RESULTS: The number of c-Fos-immunoreactive (Fos-ir) cells in the MMA group was more than six times greater than that of the control group (statistically significant, p < 0.01). In vagotomized rats inhaling MMA, markedly smaller number of Fos-ir cells was identified in the AP compared to that of rats inhaling MMA without vagotomy. Abstract
The striatum is a major cortical input site of the basal ganglia and plays a critical role in the control of orofacial movements such as licking. However, how striatal activity relates to the spatial features of licking behavior remains unclear. In this study, we examined whether neural activity in the striatal matrix and striosomal compartments is associated with the spatial position of a licking target during an operant task. Head-fixed mice performed a licking task in which the target positions were varied across three spatial dimensions. Using fiber photometry in Calb1-IRES-Cre and Pdyn-IRES-Cre mice, we recorded calcium signals from matrix and striosomal neurons. Associations between neural activity, target position, and behavioral variables were quantified using linear mixed-effects modeling with cross-validation. Matrix activity prior to licking onset was primarily associated with the dorsal–ventral target position and reaction time. During licking, matrix activity was modulated by anterior–posterior and medial–lateral positions, independent of reaction time and lick count. In contrast, striosomal activity during licking was predominantly associated with the dorsal–ventral position. These findings demonstrate that neural matrix activity is systematically associated with spatial features of licking behavior, with distinct contributions before and during movement. Our results suggest that striatal matrix circuits provide task-relevant spatial signals for the control of orofacial actions.
Significant Statement
We show that neural activity in the striatal matrix is associated with the three-dimensional position of a licking target during an operant task. Activity prior to licking onset reflects dorsal–ventral position, whereas activity during licking is modulated by the anterior–posterior and medial–lateral positions. Abstract
Licking is a continuous tongue thrust observed during drinking in rodents and humans and is often studied as an essential tongue movement for feeding and swallowing. The striatum, a component of the basal ganglia, plays a critical role in licking onset; however, it is unclear how the two compartments of the striatum—the matrix and striosomes—contribute to the control of licking onset. In this study, we used male and female transgenic mice that selectively expressed Cre recombinase in matrix or striosome neurons and subjected them to operant conditioning based on licking of a spout, during which neuronal activity in both compartments was measured using fiber photometry. Only matrix neurons showed responses prior to licking onset. In addition, the matrix neural response before licking onset was larger when mice licked a spout ipsilateral to the recording hemisphere of the brain than that observed when licking the contralateral spout. This response was observed similarly in mice conditioned to receive a reward regularly and those conditioned to receive a reward randomly, suggesting that the response was unrelated to whether the reward was predictable or unpredictable. Matrix neural activity was negatively correlated with the number of licks during the water intake behavior following the first lick. These findings suggest that matrix neurons are involved in the preparatory process for licking onset as well as in the regulation of licking frequency during water intake.
Significant Statement
This study demonstrated that during the expression of operant conditioning behaviors based on licking, striatal matrix neurons showed responses prior to licking onset. Additionally, these responses were larger when the mouse licked the spout ipsilateral to the brain hemisphere undergoing recording than when a spout in the contralateral direction was licked. This result was also true for mice conditioned using either regular or random reward conditions. Additionally, the number of licks during water ingestion behavior following the initial lick was negatively correlated with matrix neuron activity. Abstract
The striatum consists of two anatomically and neurochemically distinct compartments, striosomes and the matrix, which receive dopaminergic inputs from the midbrain and exhibit distinct dopamine release dynamics in acute brain slices. Striosomes comprise approximately 15% of the striatum by volume and are distributed mosaically. Therefore, it is difficult to selectively record dopamine dynamics in striosomes using traditional neurochemical measurements in behaving animals, and it is unclear whether distinct dynamics play a role in associative learning. In this study, we used transgenic mice selectively expressing Cre in striosomal neurons, combined with a fiber photometry technique, to selectively record dopamine release in striosomes during classical conditioning. Water-restricted mice could distinguish the conditioned stimulus (CS) associated with saccharin water from the air-puff-associated CS. The air-puff-associated CS evoked phasic dopamine release only in striosomes. Furthermore, air puff presentation induced dopamine release to striosomal neurons but suppressed release to putative matrix neurons. These findings suggest that dopamine is released in a differential manner in striosomes and the matrix in behaving animals and that dopamine release in striosomes is preferentially induced by the air-puff-associated CS and air puff presentation. These findings support the hypothesis that striosomal neurons play a dominant role in aversive stimuli prediction.

Significance Statement
Working memory-based decision strategies, such as win-stay-lose-switch (WSLS), are widely observed in humans and animals. To address neuronal bases of these strategies, we recorded neuronal activities of rat prefrontal and motor cortico-basal ganglia loops during continuous/intermittent choice tasks. The rat choice strategy was a noisy WSLS in the continuous choice condition, whereas non-WSLS was selected in the intermittent choice condition. In the continuous choice condition, the primary motor cortex and the dorsolateral striatum in the motor loop more strongly conveyed information about previous choices, rewards, and their interactions than the medial prefrontal cortex and the dorsomedial striatum in the prefrontal loop.
