    The concept of the ecological niche, defined as the basic habitat requirements for a species, is central to understanding species geographic distributions and predicting their responses to environmental change. However, identifying the essential niche for large regional communities remains a challenge because niche axes can be “hidden” by the complexity of the underlying ecological processes. Here, applying advanced species distribution modelling to nationwide environmental DNA survey data, we identified hidden niche axes of the Japanese coastal fish community and investigated the response diversity to these axes. Our survey detected 1,220 coastal fish species. The hidden niche axes collectively explained most of the variation in fish biodiversity and revealed five biogeographic boundaries for the regional community. These niches of the Japanese fish community may primarily relate to several processes due to ocean currents, such as environmental filters, transport from source areas and dispersal barriers. We also found that the response diversity to niche axes was positively correlated with species richness, although local communities with particularly high response diversity were geographically biased. A better understanding of the niche axes of the regional ecological community should help to mitigate the loss of biodiversity and ecosystem services caused by ongoing environmental change.

  • Taxonomy of freshwater gobies of the genus Rhinogobius (Oxudercidae, Gobiiformes) from central Vietnam, with descriptions of two new species

    English, Scientific journal, Taxonomy of the genus Rhinogobius was investigated for the first time in Central Vietnam. Five species occurring in this region can be distinguished by shape of their head, number of pectoral-fin rays, vertebral counts, arrangements of scales and cutaneous sensory papillae, and coloration. Two species are described as new, Rhinogobius rong sp. nov. and R. nami sp. nov., and the remaining three were identified as R. taenigena, R. leavelli, and R. similis. Though the morphology of R. leavelli and R. similis differed slightly between localities, counts and measurements overlapped, and coloration was variable. This study tentatively identifies all of those populations as R. leavelli or R. similis, but further research using additional specimens from a wider geographic range is required to clear the identity of the Rhinogobius occurring in this region. Rhinogobius rong, R. similis, and R. leavelli are distributed in both Da Nang City and Thua Thien Hue Province, with the first two species occurring widely in coastal areas, and R. leavelli restricted to inland areas. On the other hand, Rhinogobius nami and R. taenigena occur only in inland areas of Da Nang City and Thua Thien Hue Province, respectively.
  • A key evolutionary step determining osmoregulatory ability for freshwater colonisation in early life stage of fish

    English, Scientific journal, Colonisation of freshwater habitats by marine animals is a remarkable evolutionary event that has enriched biodiversity in freshwater ecosystems. The acquisition of the tolerance to hypotonic stress during the early life stages is presumed to be essential for their successful freshwater colonisation, but very little empirical evidence has been obtained to support this idea. This study aimed to comprehend the evolutionary changes of osmoregulatory mechanisms that enhance the larval freshwater tolerance in amphidromous fishes, which typically spend their larval period in marine (ancestral) habitats and the rest of life history stages in freshwater (derived) habitats. We compared the life history patterns and changes in larval survivorship and gene expression depending on salinity among three congeneric marine-originated amphidromous goby species (Gymnogobius), which had been suggested to differ in their larval dependence on freshwater habitats. An otolith microchemical analysis and laboratory-rearing experiment confirmed the presence of freshwater residents only in G. urotaenia and higher larval survivorship of this species in the freshwater condition than the obligate amphidromous G. petschiliensis and G. opperiens. Larval whole-body transcriptome analysis revealed that G. urotaenia from both amphidromous and freshwater-resident populations exhibited the greatest differences in expression levels of several osmoregulatory genes, including aqp3, which is critical for water discharge from their body during early fish development. The present results consistently support the importance of enhanced freshwater tolerance and osmoregulatory plasticity in larval fish to establish freshwater forms, and further identified key candidate genes for larval freshwater adaptation and colonisation in the goby group.

    English, Scientific journal, AbstractThe silver pomfret (Pampus argenteus) is one of the most commercially important marine fish species in the Bay of Bengal, Indian Ocean. However, detailed information on the reproductive biology of silver pomfret is limited for the Bay of Bengal, Bangladesh. To understand their reproductive biology, especially the gonadal developmental cycle and spawning season, a total of 373 individuals were collected monthly from February 2016 to January 2018 and examined. The total length (TL) and weight (TW) of P. argenteus ranged from 18.82–35.73 cm and 89.26–617.60 g, respectively. The highest gonadosomatic index (GSI) values were observed from April–June with a second (smaller) peak in October. The highest oocyte diameter was observed in May (630.50 ± 96.70 μm), when the fish reached full maturity, and the lowest was observed in July (35.10 ± 7.12 μm), when new eggs began their development for the next spawning season. The maximum number of eggs in the ovary were found in the yolk granule stage in May with a number of post-ovulatory follicles, which indicated that the ovaries developed to a peak leading up to the spawning season. A number of spermatids were also seen in males in April–June along with females. From these detailed observations, it may be concluded that P. argenteus shows two spawning peaks, in May–June and October. This observation will be helpful for the artificial breeding, sustainable management and conservation of this species in the study area.
  • Life history and instream distribution of the endangered goby Acanthogobius insularis from Okinawa-jima Island, Japan

    English, Scientific journal, The genus Acanthogobius of gobiioid fish has been reported for six species from East Asia, and inhabits estuarine and coastal waters. Within this genus, Acanthogobius insularis is an endangered and endemic goby in the Amami-oshima and Okinawa-jima Islands, southern Japan, and its range is restricted to the lowermost course of a few river basins. Basic knowledge on this species is scarce in spite of its vulnerable conservation status. The purpose of this study was to elucidate the life history of A. insularis. Monthly sampling was conducted at five stations in the Taiho River, Okinawa-jima Island, from November 2014 to November 2015. Monthly standard length (SL) distributions were unimodal except in April during the recruiting period, suggesting that A. insularis is an annual species. Analysis of the gonadosomatic index and histological observations of the ovaries revealed that this species spawns from January to May. The beginning of the spawning season seems to be related to a decline in water temperature in December. Growth rates appeared to be lower from April to December and higher in winter months. From monthly collections, A. insularis was found to move upstream with growth, and gather at spawning grounds during the reproductive season. Acanthogobius insularis might be threatened by increasing water temperature due to climate change, since low water temperatures appear to be important for their reproduction and growth. Moreover, habitat diversity, from tidal flats for recruiting grounds to upstream sites with cobbles for spawning, is needed to complete their life cycle, and should be conserved.

    English, Scientific journal, To determine the periodicity of growth increment deposition in the otoliths of newly recruited larvae of the amphidromous goby Sicyopterus japonicus, an otolith validation experiment was performed. Larval gobies collected at the river mouth of the Ota River, Wakayama, Japan were immersed in an alizarin complexone (ALC) solution to mark the timing in their otoliths. After being held under natural light conditions for 15 days, they were treated with ALC again to make the second time mark. The number of growth increments between two ALC marks in the otoliths was 14.1 on average, which roughly coincided with the number of days between two ALC treatments. This experiment validated the daily periodicity of otolith growth increment deposition in S. japonicus showing that the otolith increments of the species can be used for age determination with daily precision.
  • Larval transport of the amphidromous goby Sicyopterus japonicus by the Kuroshio Current

    English, Scientific journal, Oceanic larval transport of an amphidromous goby, Sicyopterus japonicus, was simulated using a Lagrangian modeling approach to validate the hypothesis that the Kuroshio Current could transport their larvae from Taiwan to northern Japan, which is the geographic range of this species. Simulated particles released at a 50 m depth from the off southernmost part of Taiwan reached Japan from southern Kyushu to northern Japan after 180 days. In contrast, many particles released at 120 m were trapped in eddies in the northwestern part of the subtropical gyre for 90 to 120 days and never reached the southern Japanese coast. These simulations showed that transport at a 50 m depth would enable many larvae to distribute over their entire species range after being transported away from Taiwan. The otolith growth increments of 30 larvae from Wakayama in April to August 2007 showed their oceanic larval duration ranged from 185 to 270 days, suggesting they have sufficient time as larvae for their transport from Taiwan to northern Japan. These results supported the hypothesis of larval transport described above and suggested a possibility that the Kuroshio would play an important role to maintain the apparent lack of population structure of S. japonicus from Taiwan to northern Japan.

    English, Scientific journal, To examine the genetic diversity of Sicyopterus japonicus, 448 sites of the control region of the mitochondrial DNA were analyzed in 77 specimens from the four localities of Okinawa, Kochi, Wakayama, and Shizuoka in Japan. A total of 74 haplotypes were found in the individuals examined. The same haplotypes occurred in Okinawa and Kochi, Kochi and Wakayama, and two specimens in Wakayama. The average sequence in genetic characteristics within localities varied from 1.7% in Kochi, 1.6% in Wakayama to 1.4% in Okinawa. There was no significant difference in genetic characteristics among the three locations of Wakayama, Kochi, and Okinawa (10,000 times permutation test, P>0.05). The number of specimens from Shizuoka was too small to compare to the other three locations. The neighbor-joining tree of the mitochondrial DNA haplotypes for all specimens constructed from the Kimura's two-parameter distances suggested no evidence of genetic subdivision of S. japonicus. These results suggested that this species has a single panmictic population and their larvae probably have a high dispersal ability during their oceanic stage.

    2021年度は、沖縄島の河川および河口周辺の海域において、河川性(一生を川で過ごす)のアオバラヨシノボリとキバラヨシノボリおよび両側回遊性(仔魚期を海で過ごす)のヨシノボリ属複数種の仔稚魚を採集した。採集された仔稚魚サンプルの形態観察を行い、孵化直後から着底後までさまざまな発育段階の仔稚魚が含まれることを確認した。両側回遊性の種の多くは形態による種同定が困難であったため、RNAを抽出しRAG2遺伝子の配列により種同定を行い、当初目的のアヤヨシノボリとクロヨシノボリに加え、シマヨシノボリも多く含まれていることを確認した。これら5種の仔稚魚サンプルをできる限り多くの発育過程が含まれるように選定し、RNAシーケンシングを行った。
    フィリピンのパラワン島で採集されたヨシノボリ属2種の分類と系統の研究を進め、11月に新種Rhinogobius estrellaeとRhinogobius tandikanとして記載した論文を公表した。また同じ論文の中で、ミトコンドリアゲノムの解析により、これら2種がゴクラクハゼとともに本属中最も古い系統に含まれることを示した。ヨシノボリ属は温帯と亜熱帯の大陸や島嶼を中心に分布し、パラワン島はこれまで知られていたヨシノボリ属の分布域のはるか南に位置する。新種の発見により、ヨシノボリ属の分布の南限が更新された。異なる環境間で回遊パタンや卵サイズを比較し、回遊パタン進化の適応的意義を解明することが本研究の目的の1つであり、本研究において両種は、熱帯島嶼に住むヨシノボリ属として非常に興味深い対象となる。新種記載され系統関係が示されたことにより、これらの種の回遊パタンや生活史の研究を進め、公表する道筋をつけることができた。
    また、2021年度は、回遊パタンを調べるための耳石サンプルの選定、準備を進め、2月からは、卵サイズの研究のための成熟雌の採集を進めている。

    琉球列島に生息する両側回遊魚の大規模回遊種と局地的回遊種のそれぞれ複数種をモデルとして, 仔魚の行動特性, 流下と加入の生態特性, 海洋における分散過程について比較研究を行い, ハゼ科における両側回遊生態を明らかにし, その進化の過程を考察することを目的とした。
    2011年からの継続として, 両側回遊魚のボウズハゼとヨシノボリ属仔魚の海から河川への加入生態を明らかにするため, 毎月1,2回, 沖縄島の有津川で潜水調査を行った。仔魚の加入は1月から観察され, 4月から6月がピークであった。ボウズハゼの加人期間は本州よりも長く, 本種の回遊生態が緯度により変動することを明らかにした。ボウズハゼ仔魚について耳石による日齢査定を行ったところ, 仔魚は聯化直後から約200日間を海で過ごすことが分かった。
    大規模回遊種ルリボウズハゼとナンヨウボウズハゼの産着卵から艀化仔魚を得て, 超小型ワムシ(プロアレス), SS型ワムシを給餌し, 飼育を行った。艀化後0,1,2,3,5,7日後の昼と夜の仔魚の比重を測定した結果, 比重は, 艀化直後には大きな値を示すものの, 発育とともに急減し, さらに昼夜で変化することがわかった。
    沖縄島において, 通し回遊性ハゼ亜目魚類のうち大規模回遊種ナンヨウボウズハゼと局所的回遊種ナガノゴリ, ゴクラクハゼを採集した。フランス国立自然史博物館において, レーザーアブレーションICP-MS(誘導結合プラズマ質量分析装置)を用いて, これらの耳石に含まれるSr, Ca, Mg, Baの各元素量を測定し, 回遊履歴を分析した結果, 3種はいずれも両側回遊型の回遊パタンを示すものの, 河川への加入生態を反映して, 異なる移動形態を持つことが明らかとなった。
    以上の結果より, ハゼ亜目魚類の通し回遊は, 回遊規模に加えて, 海洋での生態, 河川と海との回遊パタンなど様々な要因により形成, 維持されてきたと推察された。
