- Electroporation-Mediated Gene Delivery in Pyropia yezoensis (Rhodophyta) Without Cell Wall Removal.

English, Scientific journal, This study aimed to assess the potential of a gene delivery technique in the red macroalga Pyropia yezoensis (Rhodophyta) using electroporation without removing the cell wall. An antibiotic resistance gene was introduced into P. yezoensis tissues containing cells with intact cell walls through electroporation, followed by selection with the corresponding antibiotic. No germlings survived in the non-electroporated control tissue fragments under antibiotic selection. In contrast, some germlings were observed to survive in the electroporated group. Furthermore, the presence of antibiotic resistance genes was confirmed in the genomic DNA of several antibiotic-resistant germlings. Although reporter genes such as β-glucuronidase (GUS) and green fluorescent protein (GFP) were also introduced as supplementary markers, their expression was not detectable under the tested conditions. These findings provide evidence supporting the successful introduction of antibiotic resistance genes into P. yezoensis cells via electroporation. This study offers a preliminary assessment of a gene delivery strategy in P. yezoensis that bypasses cell wall removal, presenting a straightforward method for introducing foreign genes into Pyropia. To the best of our knowledge, this is the first report to demonstrate successful gene transfer via electroporation in a macroalga without cell wall removal. These results provide valuable insights for the development of genetic transformation systems in red macroalgae. - Isolation and characterization of a spore development mutant in Pyropia yezoensis (Rhodophyta) induced via insertional mutagenesis.

English, Scientific journal, Mechanistic elucidation of spore development is essential for understanding reproductive strategies and improving seaweed cultivation. In this study, we isolated a mutant with spore development defects in Pyropia yezoensis (Rhodophyta) from among genetic transformants generated via insertional mutagenesis. We characterized the mutant phenotype and identified the genomic region containing the mutation. The mutant produced spores (archeospores, zygotospores, and conchospores) at every relevant stage of the life history. No evident differences were observed in the appearance of the released spores at any stage under light microscopy between the wild-type and mutant strains; however, the mutant exhibited a trend of reduced spore release compared with the wild-type. Additionally, the archeospores released by the mutant remained round after being released from the sporangium, failed to adhere to the substratum, did not germinate, and showed no cell wall development after 4 days of standard culture, in contrast to the wild-type. Similar results were observed for the zygotospores and conchospores. These findings indicate that the main characteristic of this mutant was poor spore release, and the released spores failed to germinate. Whole-genome sequencing revealed the introduction of exogenous DNA into the nuclear genome of the mutant and suggested the presence of duplications, deletion, and inversion in the mutant genome. Notably, a deletion of approximately 13 kb was detected in the mutant genome flanking the insertion, and this region contained a plant homeodomain, suggesting that homeodomain-containing proteins may regulate spore development in P. yezoensis. - Ishimozuku (Sphaerotrichia firma) lipids containing fucoxanthin suppress fatty liver and improve short chain fatty acid production in obese model mice

English, Scientific journal, Small heat shock proteins (sHSPs) are found in all three domains of life (Bacteria, Archaea, and Eukarya) and play a critical role in protecting organisms from a range of environmental stresses. However, little is known about their physiological functions in red algae. Therefore, we characterized the sHSPs (PysHSPs) in the red macroalga Pyropia yezoensis, which inhabits the upper intertidal zone where it experiences fluctuating stressful environmental conditions on a daily and seasonal basis, and examined their expression profiles at different developmental stages and under varying environmental conditions. We identified five PysHSPs (PysHSP18.8, 19.1, 19.2, 19.5, and 25.8). Real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR) analysis showed that expression of the genes PysHSP18.8, PysHSP19.5, and PysHSP25.8 was repressed at all the developmental stages under normal conditions, whereas PysHSP19.1 and PysHSP19.2 were overexpressed in mature gametophytes and sporophytes. Exposure of the gametophytes to high temperature, oxidative stress, or copper significantly increased the mRNA transcript levels of all the five genes, while exogenous application of the ethylene precursor 1-aminocylopropane-1-carboxylic acid (ACC) significantly increased the expression levels of PysHSP19.2, PysHSP19.5, and PysHSP25.8. These findings will help to further our understanding of the role of PysHSP genes and provide clues about how Pyropia species can adapt to the stressful conditions encountered in the upper intertidal zone during their life cycle. - カシスの魅力と健康機能性

English, Scientific journal, Fucoxanthin is a specific carotenoid in brown seaweeds with remarkable biological properties. Ishimozuku (Sphaerotrichia divaricata), an edible brown alga from northern Japan, has morphology that is almost identical to that of Okinawa-mozuku (Cladosiphon okamuranus) harvested off Okinawa, Japan. However, because of Ishimozuku's lower availability compared to Okinawa-mozuku, the contents of its nutrient compounds remain unclear. The present study analyzed fucoxanthin and anti-oxidant compound contents of Ishimozuku harvested off the northern coast of Japan from 2014 to 2016. First, 80% ethanol extract solutions were prepared from Ishimozuku harvested from several west coast areas of Aomori, Japan. Then, polyphenol content was analyzed using the Folin⁻Ciocalteu method. Then anti-oxidative effects were analyzed by their 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and hydrogen peroxide scavenging activity. Furthermore, fucoxanthin contents were measured using high performance liquid chromatography (HPLC) analysis. Fucoxanthin contents of Ishimozuku were 105.6⁻1148.5 μg/g dry weight. Total polyphenol contents of Ishimozuku were of 0.296⁻0.958 mg/g dry weight: higher than Okinawa-mozuku (0.082 ± 0.011 mg/g dry weight). The anti-oxidation effects of Ishimozuku accompanied the polyphenol content. These results suggest that Ishimozuku contains various anti-oxidant components and has high potential to provide the promotion of human health. - Development of a nuclear transformation system with a codon-optimized selection marker and reporter genes in Pyropia yezoensis (Rhodophyta)

English, Scientific journal, Marine macroalgae play an important role in marine coastal ecosystems and are widely used as sea vegetation foodstuffs and for industrial purposes. Therefore, there have been increased demands for useful species and varieties of these macroalgae. However, genetic transformation in macroalgae has not yet been established. We have developed a dominant selection marker for stable nuclear transformation in the red macroalga Pyropia yezoensis. We engineered the coding region of the aminoglycoside phosphotransferase gene aph7″ from Streptomyces hygroscopicus to adapt codon usage of the nuclear genes of P. yezoensis. We designated this codon-optimized aph7″ gene as PyAph7. After bombarding P. yezoensis cells with plasmids containing PyAph7 under the control of their endogenous promoter, 1.9 thalli (or individuals) of hygromycin-resistant strains were isolated from a 10-mm square piece of the bombarded thallus. These transformants were stably maintained throughout the asexual life cycle. Stable expression of PyAph7was verified using Southern blot analysis and genomic PCR and RT-PCR analyses. PyAph7 proved to be a new versatile tool for stable nuclear transformation in P. yezoensis. - Structural features and gene-expression profiles of actin homologs in Porphyra yezoensis (Rhodophyta).

English, Scientific journal, The marine red alga Porphyra yezoensis contains an actin gene family consisting of at least four isoforms (PyACT1, 2, 3 and 4). The amino acid identity between isoforms exceeds 83%, and each contains a putative nuclear export signal (NES). We scanned the sequences for amino acids in regions homologous to the intermonomeric interface of actin filaments. Few residues expected to engage in cross-linking were conserved between the four isoforms. The results of the sequence analyses suggest that PyACT2 probably functions in the nucleus as a monomer (G-actin) or in other unconventional forms. In addition, the distribution and position of the introns were different from those in florideophycean actin genes. The expression level of PyACT3 in matured gametophytes was significantly higher than in those in a vegetative state, although the mRNA was detected at similar levels in both apical and basal parts of thalli. The expression levels of PyACT2 and 4, on the other hand, did not change significantly between the matured and vegetative gametophytes. English, Scientific journal, As a part of the construction of a Porphyra yezoensis Ueda genetic linkage map, we conducted intraspecific cross-experiments and subsequent screening of cross-fertilized conchocelis by cleaved amplified polymorphic sequence (CAPS) analysis. The cross-experiments were carried out between males of the wildtype (KGJ) and females of the recessive green mutant (TU-2) using two methods, controlled and random crosses. A total of 42 and 186 wildtype-colored conchocelis colonies were obtained from the former and latter experiments, respectively. Among those, 49 DNA samples (14% and 23% obtained from the former and latter crosses, respectively) showed biparental CAPS patterns in the two gene regions (EF-1α open reading frame [ORF] region and V-ATPase). This study represents the first report in which the cross-fertilized conchocelis of P. yezoensis has been directly confirmed by molecular marker. The combination of the simple DNA extraction and CAPS analysis may be applicable in genetic studies of other macroalgae that are monoecious and/or grow slowly in laboratory culture. - Factors influencing efficiency of transient gene expression in the red macrophyte Porphyra yezoensis

English, Scientific journal, Asexual reproduction via archeospores in Porphyra yezoensis Ueda gametophytes is a very valuable character to nori farming; however, there is little information available on the molecular basis of the developmental process. To identify genes involved in the Porphyra asexual sporulation, we compared the gene expression profiles derived from four developmental stages of the life cycle (three from gametophytes; one from sporophytes) using cDNA macroarray, which includes 4,896 nonredundant expressed sequence tag (EST) groups. Candidate genes were screened by two different macroarray data analyses combined with reverse transcription-PCR (RT-PCR) analysis or Northern analysis. RT-PCR analysis revealed that nine genes (one: similarity to 5'-3' exoribonuclease; the other eight: no sequence similarity to known proteins) were expressed with a gametophyte (G)-specific manner, and two genes (named ASPO2608, ASPO1527) were expressed only in gametophytes that formed archeospores. The deduced amino acid sequences for the latter two genes are predicted to contain signal peptides for secretion at their N-termini. Northern analysis revealed that expression levels of Calvin cycle genes in the gametophytic stage that formed archeospores (G-A stage) were higher than those of the gametophyte blade with no archeospores (G-NA stage). In the macroarray analysis based on the rank data of G-preferentially expressed genes, which were detected in the previous P. yezoensis EST analysis, one gene encoding the cyclase associated protein (CAP) exhibited a change upwardly in the G-A stage >1,000 ranks to the G-NA stage. Japanese, We extracted a xylanase and a mannanase from Ezo abalone Haliotis discus hannai, and examined the enzyme activity and the ability for cell wall degradation of Porphyra yezoensis. Abalone xylanase was partially purified from the crude enzyme prepared from the digestive fluid by column chromatography on a TOYOPEARL CM-650M. Abalone mannanase was successfully purified from the crude enzyme by column chromatographies on TOYOPEARL CM-650M, hydroxyapatite, and TOYOPEARL HW-50E The gametophytic thalli of P.yezoensis could not be degraded by the xylanase alone. The purified mannanase could degrade the thalli into cell masses consisting of 10-20 cells; however, it produced practically no protoplasts from the thalli. When the thalli were incubated both with the xylanase and the purified mannanase, some protoplasts could be produced from the thalli. The two enzymes may be useful for the studies on cell engineering and asexual seedling production of this alga. - A Preliminary Genetic Analysis of Several Industrially Important Traits in Porphyra yezoensis (Bangiales, Rhodophyta)

Japanese, Inheritance of several phenotypes of Porphyra yezoensis was investigated by a cross experiment. The following phenotypes, regarded as important characters in layer cultivation, were subjected in the present study: color, twist, corrugation, hardness, amount of monospores, maturing pattern, maturing rate, thallus collapse and thallus form. Among those, color, twist, hardness and maturing pattern were appeared to be stable and reliable genetic traits. Furthermore, linkage of two phenotypes, twist and maturing pattern, was suggested. This is the first report of a genetic analysis focusing on the important characters for industrial quality, except for color inheritance, and it may be helpful for further investigation of improvement of breeds in Porphyra. - 海産紅藻スサビノリの配偶体における薬剤感受性に関する研究

Japanese, Symbiotic bacteria BPY-W6 and BPY-W8 were added to an axenic culture strain (TU-1) of Porphyra yezoensis, and the length and width of gametophyte of the organism were measured. Axenic cultures of the gametophyte were used as a control. Area and morphological index value (MIV) were calculated on the basis of the length and width measured. After culture for 42 days, the form of the control strain became the callus-like masses, and the strains which added bacteria became monostromatic gametophytes. The strain in which the average length was most lengthened was one added BPY-W8. The strain in which the average width was most lengthened was one added BPY-W6. The strain in which the average area increased most was one added BPY-W6. Throughout the measurement period, MIV for the strain added BPY-W8 was bigger than that of the strain added BP-YW6. It was suggested that BPY-W6 was the bacteriostrain of which the effect for the elongation of the width is big and tended to form the ova-like gametophyte, and BPY-W8 was the bacteriostrain of which the effect for the elongation of the length is big and tended to form the elliptical-like gametophyte from these results. - Isolation and characterization of an elongation factor-1α gene in Porphyra yezoensis (Rhodophyta)

The purpose of this study is to establish the genome editing technology of Neopyropia yezoensis by applying our gene transfer technology. Although this study could not demonstrate genome editing by the microinjection method, it was confirmed that the reporter gene introduced by the method was transiently expressed in Neopyropia yezoensis cells. It was suggested that the microinjection method contributes to gene transfer technology including genome editing of Neopyropia yezoensis.

The purpose of this study, using the transformation techniques of Pyropia yezoensis, was to research and develop new breeding base technology to contribute to the nori breeding. In this study, we tried to produce transformants with a reporter gene. However, transformant candidates to clearly express the reporter gene was not confirmed. As a result, transformant candidates available for demonstration of meiosis P. yezoensis was not obtained. In conclusion, to increase the transformation efficiency, it is suggested that the gene transfer technology of P. yezoensis be still improved.

目標達成するため、初めにホタテウロが可溶化(スラリー状)となる適切な条件を探索し、スラリー状にする条件を確立することに成功した。次に競争吸着法により、脱カドミウムしたホタテウロはカドミウム濃度が1.7mg/kgを示し、飼料安全法の3.0mg/kgを下回った。したがって、目標である競争吸着法によるホタテウロのカドミウム分離条件の確立を達成した。脱カドミウムしたホタテウロは液状のため乾燥する必要があり、脱脂米糠に吸着させ乾燥後、大豆粕の代替として利用できるかどうかの検討を行った結果、代替できることが分かった。今後は実際に畜産用飼料として利活用可能か実証への展開が考えられる。
